    ANTIDIABETIC AND ANTIDYSLIPIDEMIC ACTIVITY OF ETHYL ACETATE FRACTIONS OF XYLOCARPUS GRANATUM AND XYLOCARPUS MOLLUCCENSIS ON HIGH FRUCTOSE HIGH FAT AND HIGH SUCROSE HIGH FAT FED-LOW DOSED STREPTOZOTOCIN TREATED DIABETIC RATS

    Objectives: The present study was carried out to investigate the antidiabetic and antidyslipidemic effect of standardized fractions of X. granatum (CDR-134 F194) and X. molluccensis (CDR-267 F018) by measuring the status of blood glucose, serum insulin, lipid levels, hepatic and renal function markers of high fructose high fed streptozotocin treated rats and high sucrose high fat diet fed-low dosed Streptozotocin treated diabetic rats.Methods: Male rats of Sprague Dawley strain of body weight around 150 g when kept on high fructose high fat diet and high sucrose high fat diet for two weeks, respectively, showed abnormal glucose tolerance, dyslipidemia and obesity and at this stage when streptozotocin was given intraperitoneally at 45.0 mg/kg body weight caused persistent hyperglycemia in them addition to dyslipidemia along with impairment in their hepatic and renal functions.Results: The standardized fractions of X. granatum (CDR-134 F194) and X. molluccensis (CDR-267F018) when given to these high fructose high fat fed low dosed streptozotocin treated diabetic rats or high sucrose high fat diet fed-low dosed streptozotocin treated diabetic rats for 10 consecutive days showed significant improvement in their glucose intolerance, decline in their serum triglycerides and LDL-cholesterol levels. These CDR-134 F194 and CDR-267 F018 treated rats also showed elevation in their HDL-cholesterol levels and improvement in their hepatic and renal functions as evidenced by decline in SGOT, SGPT, urea, uric acid and creatinine levels.Conclusion: The present study thus concludes that the antidiabetic efficacy of standardized fractions of X. granatum (CDR-134 F194) and X. molluccensis (CDR-267F018) have favorable effect in bringing down the severity of hyperglycemia, hyperlipidemia, decline the increased level of renal and hepatic function markers and also improving glucose tolerance activity

    RAPD-based genetic relationships in different Bougainvillea cultivars

    The present study deals with authenticating existing knowledge about 21 Bougainvillea cultivars comprisingof 9 hybrids and their parents through RAPD analysis. The 19 degenerate primer sets generated 234 bands from which 158(67.5%) were polymorphic. The UPGMA based dendrogram divided 21 cultivars into two major groups with Jaccard’ssimilarity coefficient ranging from 0.51 to 0.942. Group A had three cultivars namely Trinidad, Formosa and Dr. H. B. Singhin which Dr. H.B. Singh was confirmed as a hybrid of other two cultivars. Group B was sub divided into 8 clusters. Theparentages of 7 out of 8 hybirds have been confirmed based on clusters.     Response of potato clones to planting dates in Pokhara, Kaski, Nepal

    A field trial was conducted to optimize the planting date and appropriate clone for ware potato production at Horticulture Research Station, Malepatan, Pokhara from October 2015 to March 2016. The trial was laid out in two factors Factorial Randomized Complete Block Design. There were nine treatments with three replications. The treatments comprised of three dates of planting (30th October, 14th and 29th November) and three potato clones (Janakdev, Lbr 40 and PRP 266264.1). The results showed that the main effects of planting dates and potato clones significantly (P<0.05) affected most growth, yield and yield attributing parameters of the crop. Interaction effect of planting dates and potato clones also significantly influenced the number of above-ground stems per plant, ground cover percentage, number of tubers per plant, number and weight of small size tuber (<25 g), weight of medium or seed size (25-50 g) tuber and large size (>50 g) tuber, number and yield of tubers per plot, and yield t/ha. The highest yield (37.05 t/ha) was obtained from 14th November planting. The clone Lbr 40 produced a higher yield (36.05t/ha). The clone PRP 266264.1 planting on 30th October planting produced significantly a higher yield (41.34t/ha). The clone Lbr 40 produced significantly highest number and weight of large size (>50 g) tuber. Number of small size (<25 g) tuber per plot or unmarketable tuber significantly increased on delayed planting

    ANTIHYPERGLYCEMIC AND ANTIDYSLIPIDEMIC ACTIVITY IN ETHYL ACETATE FRACTION OF THE FRUITS OF XYLOCARPUS GRANATUM AND XYLOCARPUS MOLUCCENSIS

    Objectives: Although various species of Xylocarpus i. e. Granatum, moluccensis are known for their medicinal properties. Yet, its anti-diabetic activity remains to be defined. So the aim of this study was to evaluate the antihyperglycemic and antidyslipidemic activity in ethyl acetate fraction of the fruits of X. granatum and X. moluccensis on validated animal models as well as in-vitro glucose uptake stimulatory effect and their cytotoxicity effect in L6 skeletal muscle cells.Methods: The ethyl acetate fraction of the fruits of X. granatum and X. moluccensis were administered to diabetic groups daily up to 10 days for prolonged study. Biochemical parameters notably glucose tolerance, insulin level, lipid profile were assessed. The ethyl acetate fraction of the fruits of X. granatum and X. moluccensis were also tested for glucose uptake effect by skeletal muscle cells in the concentration dependent manner.Results: The present study show that the ethyl acetate fraction of the fruits of X. granatum as well as X. moluccensis are effective in improving glucose tolerance, declining blood glucose as well as serum cholesterol and triglycerides levels in low dosed streptozotocin-induced diabetic rats and dyslipidemic hamsters, respectively. These fractions were also found efficient in increasing glucose uptake by L6 skeletal muscle cells but did not show any effect on cell viability of L6 skeletal muscle cells.Conclusion: Based on the results, the present study revealed that ethyl acetate fraction of the fruits of X. granatum and X. moluccensis lowered blood glucose profile by increasing the glucose uptake by L-6 and this may be the possible mechanisms for the antidiabetic and antidyslipidemic action

    Revealing the therapeutic potential of teriparatide: a review

    Teriparatide is an FDA-approved medication for osteoporosis that presents promising results in treating various musculoskeletal conditions. It helps in improving the bone mineral density and preventing fractures in individuals with osteoporosis. Its effectiveness in treating non-union and delayed union fractures, atypical femoral fractures, and spinal fusion procedures makes it valuable in improving bone healing and reducing complications. Teriparatide also improves bone density and strength in individuals with osteogenesis imperfecta, helps prevent and treat hypocalcaemia post-thyroidectomy, and helps in the management of hypoparathyroidism. In MRONJ, teriparatide improves lesion resolution and reduces bony defects. Furthermore, it potentially prevents bone metastasis in cancer patients without stimulating tumour growth. Nevertheless, teriparatide may cause short-term side effects like nausea and long-term concerns pertaining to the risk of osteosarcoma. Recent European alliance of associations for rheumatology guidelines have highlighted teriparatide's superior effectiveness in achieving bone mineral density thresholds and reducing fracture risks. Further clinical trials are necessary to determine optimal dosages and treatment durations of teriparatide. The off-label use of teriparatide should be considered only under the guidance of a healthcare professional when standard options are unavailable or inadequate

    Acute leukemia after cytotoxic treatment in a child with nephrotic syndrome

    Renal involvement in acute lymphoblastic leukemia (ALL) occurs due to several factors including leukemic infiltration of the kidneys, therapy-related side effects such as tumor lysis syndrome, nephrotoxic drugs, and septicemias. A 3-year-old boy with nephrotic syndrome (NS) who was previously treated with prednisolone and cyclosporine A for 14 months after the initial diagnosis of NS, presented to the emergency department with fever, breathing difficulty, generalized edema, and body pain with pallor, without evidence of lymphadenopathy, hepatosplenomegaly, petechiae, or purpura. On investigation, peripheral blood smear showed blast cells >80% and bone marrow aspiration showed complete replacement of the marrow with L1 lymphoblasts, consistent with a diagnosis of ALL. The exact mechanism of developing acute leukemia after cytotoxic treatment has not been established; the possibility must be considered that the incidence of this malignant disease is increased after cytotoxic treatment for nonmalignant diseases

    Agarose processing in protic and mixed protic–aprotic ionic liquids : dissolution, regeneration and high conductivity, high strength ionogels

    We have shown that low viscosity alkyl or hydroxyalkyl ammonium formate (ILs) can dissolve agarose, and higher dissolution can be achieved in the mixed, alkyl or hydroxyalkyl ammonium + imidazolium or pyridinium ILs. The polarity parameters α, β, π*, ET(30) and ETN of these IL systems were measured to explain their dissolution ability for agarose. Dissolved agarose was either regenerated using methanol as a precipitating solvent or ionogels were formed by cooling the agarose–IL solutions to ambient temperature. Exceptionally high strength ionogels were obtained from the agarose solutions in N-(2-hydroxyethyl)-ammonium formate or its mixture with 1-butyl-3-methylimidazolium chloride. Regenerated material and ionogels are characterized for their possible degradation/conformational changes and gel properties (thermal hysteresis, strength, viscoelasticity and conductivity) respectively. A high strength, high conducting ionogel was demonstrated to be able to build an electrochromic window. Such ionogels can also be utilized for other soft matter electronic devices and biomedical applications

    Effect of application of some organic amendments on soil properties, extractable micronutrients and their chemical fractions in a sandy loam soil

    In a 120-day incubation study, farmyard manure (FYM), mushroom compost (MC), poultry manure (PM), vermi-compost (VC), biogas slurry (BS) and biochar from Lantana sp.; a common weed (BC) each added at 15 t ha-1 significantly increased soil electrical conductivity and organic C content, however, only VC failed to increase soil organic C content significantly but increased soil pH when compared with the control sample. Among organic amendments, FYM, MC, VC and BS increased DTPA-extractable Zn and Mn in soil significantly when compared with the control sample, while BC was significantly effective in regard to increasing both DTPA-extractable Mn and hot water soluble B when compared with the control sample. Application of FYM and MC significantly increased DTPA-extractable Cu and Fe, respectively when compared with the control sample. Simple correlation and path coefficient analysis were performed to identify key chemical fractions of micronutrients in soil having the direct and indirect effects on availability of micronutrients in organically amended soils. Based on soil extractable concentrations of micronutrients in organically amended soils, FYM appeared to be the best source of Zn, Cu and Mn, while MC could serve as the best source of Fe.
