    Transfer of Flexibacter sancti, Flexibacter filiformis, Flexibacter japonensis and Cytophaga arvensicola to the genus Chitinophaga and description of Chitinophaga skermanii sp nov

    Analysis of the 16S rRNA gene sequences of species currently assigned to the genus Flexibacter has shown extensive intrageneric phylogenetic heterogeneity. It has been shown in previous studies that the species [Flexibacter] sancti, [Flexibacter] filiformis and [Flexibacter] japonensis were most closely related to Chitinophaga pinensis. In addition, [Cytophaga] arvensicola and species of the genus Terrimonas also clustered into this phylogenetic group. Although the similarities of 16S rRNA gene sequences were low (88.5-96.4%), there is no evidence for clear phenotypic differences between these organisms that justify assignment to different genera. A proposal is made to transfer these species to the genus Chitinophaga as Chitinophaga sancti comb. nov., Chitinophaga filiformis comb. nov., Chitinophaga japonenis comb. nov. and Chitinophaga arvensicola comb. nov. on the basis of phylogenetic and phenotypic data. Furthermore, a novel species is described within this genus, Chitinophaga skermanii sp. nov., with strain CC-SG1B(T) (=CCUG 52510(T)=CIP 109140(T)) as the type strain

    Impact of Biological Sex on Immune Activation and Frequency of the Latent HIV Reservoir during Suppressive Antiretroviral Therapy

    Background: Persistent HIV infection of long-lived resting CD4 T cells, despite antiretroviral therapy (ART), remains a barrier to HIV cure. Women have a more robust type 1 interferon response during HIV infection relative to men, contributing to lower initial plasma viremia. As lower viremia during acute infection is associated with reduced frequency of latent HIV infection, we hypothesized that women on ART would have a lower frequency of latent HIV compared to men. Methods: ART-suppressed, HIV seropositive women (n = 22) were matched 1:1 to 22 of 39 ART-suppressed men. We also compared the 22 women to all 39 men, adjusting for age and race as covariates. We measured the frequency of latent HIV using the quantitative viral outgrowth assay, the intact proviral DNA assay, and total HIV gag DNA. We also performed activation/exhaustion immunophenotyping on peripheral blood mononuclear cells and quantified interferon-stimulated gene (ISG) expression in CD4 T cells. Results: We did not observe evident sex differences in the frequency of persistent HIV in resting CD4 T cells. Immunophenotyping and CD4 T-cell ISG expression analysis revealed marginal differences across the sexes. Conclusions: Differences in HIV reservoir frequency and immune activation appear to be small across sexes during long-term suppressive therapy

    Human infection challenge in the pandemic era and beyond, HIC-Vac annual meeting report, 2022

    HIC-Vac is an international network of researchers dedicated to developing human infection challenge studies to accelerate vaccine development against pathogens of high global impact. The HIC-Vac Annual Meeting (3rd and 4th November 2022) brought together stakeholders including researchers, ethicists, volunteers, policymakers, industry partners, and funders with a strong representation from low- and middle-income countries. The network enables sharing of research findings, especially in endemic regions. Discussions included pandemic preparedness and the role of human challenge to accelerate vaccine development during outbreak, with industry speakers emphasising the great utility of human challenge in vaccine development. Public consent, engagement, and participation in human challenge studies were addressed, along with the role of embedded social science and empirical studies to uncover social, ethical, and regulatory issues around human infection challenge studies. Study volunteers shared their experiences and motivations for participating in studies. This report summarises completed and ongoing human challenge studies across a variety of pathogens and demographics, and addresses other key issues discussed at the meeting

    Reliable prediction methods for the frictional pressure drop in liquid-vapor flows at zero gravity are essential to the design of two-phase thermal control and fluid transfer systems aboard spacecrafts. The present investigation is concerned with the prediction of the frictional pressure drop in steady, fully developed, turbulent liquid-vapor duct flows in both dispersed and annular flow regimes. The dynamics of such flows are simulated by using two immiscible, neutrally buoyant liquids, water and n-butyl benzoate, in an earth based flow facility.In the case of dispersed flow, simulation experiments were conducted with n-butyl benzoate droplets in flowing water. The theoretical basis of the simulation is given. Experiments showed that, for a fixed combined flow rate of water and benzoate, the frictional pressure drop is unaffected by large changes in the volume fraction of benzoate drops and their size distribution. Measured power spectra of the static wall pressure fluctuations induced by the turbulent water-benzoate flow also revealed that their dynamics is essentially unaltered by the presence of the droplets. These experimental findings, together with the theoretical analysis, led to the conclusion that the pressure drop in fully developed, dispersed liquid-vapor flow in straight ducts of constant cross section at 0-g is identical to that due to liquid flowing alone at the same total volumetric flow rate and, therefore, can be readily determined.A method for predicting the frictional pressure drop in liquid-vapor annular flows in 0-g has also been developed. Based on an analysis and published correlations, a means of relating the measured 1-g frictional pressure drop in water-benzoate flow to the liquid-vapor pressure drop in 0-g was developed. The use of empirical correlations was necessary because of the lack of rigorous theoretical analysis for the dynamics of annular flows. Two different correlations were used, and it was found that the results were in good agreement.Based on the results of this investigation, the frictional pressure drop in steady, fully developed, turbulent, liquid-vapor duct flow in 0-g is found to be consistently less than that in 1-g under otherwise identical conditions.
