The annual ammonia budget of fertilised cut grassland – Part 1: Micrometeorological flux measurements and emissions after slurry application
Two commercial ammonia (NH<sub>3</sub>) analysers were customised to allow continuous measurements of vertical concentration gradients. The gradients were used to derive ammonia exchange fluxes above a managed grassland site at Oensingen (Switzerland) by application of the aerodynamic gradient method. The measurements from July 2006 to October 2007 covered five complete growth-cut cycles and included six applications of liquid cattle slurry. The average accuracy of the flux measurements during unstable and near-neutral conditions was 20% and the detection limit was 10 ng NH<sub>3</sub> m<sup>&minus;2</sup> s<sup>&minus;1</sup>. Hence the flux measurements are considered sufficiently accurate for studying typical NH<sub>3</sub> deposition rates over growing vegetation. Quantifying the overall emissions after slurry applications required the application of elaborate interpolations because of difficulties capturing the initial emissions during broadspreading of liquid manure. The emissions were also calculated with a mass balance method yielding similar fluxes. NH<sub>3</sub> losses after slurry application expressed as percentage of emitted nitrogen versus applied total ammoniacal nitrogen (TAN) varied between 4 and 19%, which is roughly a factor of three lower than the values for broadspreading of liquid manure in emission inventories. The comparatively low emission factors appear to be a consequence of the low dry matter content of the applied slurry and soil properties favouring ammonium adsorption
The annual ammonia budget of fertilised cut grassland – Part 2: Seasonal variations and compensation point modeling
The net annual NH<sub>3</sub> exchange budget of a fertilised, cut grassland in Central Switzerland is presented. The observation-based budget was computed from semi-continuous micrometeorological fluxes over a time period of 16 months and using a process-based gap-filling procedure. The data for emission peak events following the application of cattle slurry and for background exchange were analysed separately to distinguish short-term perturbations from longer-term ecosystem functioning. A canopy compensation point model of background exchange is parameterised on the basis of measured data and applied for the purposes of gap-filling. The data show that, outside fertilisation events, grassland behaves as a net sink for atmospheric NH<sub>3</sub> with an annual dry deposition flux of &minus;3.0 kg N ha<sup>&minus;1</sup> yr<sup>&minus;1</sup>, although small NH<sub>3</sub> emissions by the canopy were measured in dry daytime conditions. The median &Gamma;<sub><i>s</i></sub> ratio in the apoplast (=[NH<sub>4</sub><sup>+</sup>]/[H<sup>+</sup>]) estimated from micrometeorological measurements was 620, equivalent to a stomatal compensation point of 1.3 &mu;g NH<sub>3</sub> m<sup>&minus;3</sup> at 15 &deg;C. Non-stomatal resistance to deposition <i>R<sub>w</sub></i> was shown to increase with temperature and decrease with surface relative humidity, and <i>R<sub>w</sub></i> values were among the highest published for European grasslands, consistent with a relatively high ratio of NH<sub>3</sub> to acid gases in the boundary layer at this site. Since the gross annual NH<sub>3</sub> emission by slurry spreading was of the order of +20 kg N ha<sup>&minus;1</sup> yr<sup>&minus;1</sup>, the fertilised grassland was a net NH<sub>3</sub> source of +17 kg N ha<sup>&minus;1</sup> yr<sup>&minus;1</sup>. A comparison with the few other measurement-based budget values from the literature reveals considerable variability, demonstrating both the influence of soil, climate, management and grassland type on the NH<sub>3</sub> budget and the difficulty of scaling up to the national level
Virtual disjunct eddy covariance measurements of organic compound fluxes from a subalpine forest using proton transfer reaction mass spectrometry
International audienceA `virtual' disjunct eddy covariance (vDEC) device was tested with field measurements of biogenic VOC fluxes at a subalpine forest site in the Rocky Mountains of the USA. A PTR-MS instrument was used as the VOC sensor. Daily peak emission fluxes of 2-methyl-3-buten-2-ol (MBO), methanol, acetone and acetaldehyde were around 1.5, 1, 0.8 and 0.4 mg m-2 h-1, respectively. High pass filtering due to long sampling lines was investigated in laboratory experiments, and suggested that VOC losses in PTFA lines are generally governed by diffusion laws. Memory effects and surface reactions did not seem to play a dominant role. Model estimates of MBO fluxes compared well with measured fluxes. The results also suggest that latent heat and sensible heat fluxes are reasonably well correlated with VOC fluxes and could be used to predict variations in VOC emissions. The release of MBO, methanol, acetone and acetaldehyde resulted in significant change of tropospheric oxidant levels and a 10--40% increase in ozone levels, as inferred from a photochemical box model. We conclude that vDEC with a PTR-MS instrument is a versatile tool for simultaneous field analysis of multiple VOC fluxes
Starting or Changing Therapy - A Prospective Study Exploring Antiretroviral Decision-Making
Background:: When to start or change antiretroviral treatment against HIV infection is of major importance. Patients' readiness is considered a major factor influencing such treatment decisions, in particular because no objective, absolute time point when to start antiretroviral therapy exists. We aimed at evaluating patients' readiness to start or change antiretroviral therapy (ART). Patients and Methods:: HIV-infected patients starting or changing ART between July 2002 and February 2003, treating physicians and nurses participated in this prospective, observational multicenter study. We assessed shared decision-making including qualitative aspects, expected treatment decisions and treatment status after 3 months. Results:: 75 patients were included. Of 34 patients for whom starting ART was considered, 27 (79%) indicated that they were willing to start treatment. After 3 months, 21 of 27 (78%) actually started therapy, six did not. Patients with depression were less likely to be ready for ART (p < 0.05). Of 41 patients for whom changing ART was considered, 35 (85%) indicated that they were willing to change treatment. Of the latter 35 patients, 33 (94%) finally changed ART within 3 months. Physicians and nurses were too optimistic in predicting the start or change of ART. The main reason to start or change ART was the sole recommendation of the physician (52% in those starting, 61% in those changing ART). Patients mainly judged the decision as shared and were very satisfied (71%) with the process. Qualitative findings revealed the importance of a dialectic decisionmaking, described with two categories: "dealing with oneself and others”‚ and "understanding and being understood.” Conclusion:: Patients mainly shared the decision made during consultation. Although physicians have an essential role concerning ART, patients, physicians, and nurses all contribute to the decision. Modulation of Neutrophil Function by Recombinant Human IgG1 Fc Hexamer in the Endogenous K/BxN Mouse Model of Rheumatoid Arthritis.
INTRODUCTION
Neutrophils are a pivotal cell type in the K/BxN mouse model of rheumatoid arthritis and play an essential role in the progression of the arthritis. They are readily activated by immune complexes (ICs) via their FcγRs to release IL-1β in addition to other cytokines, which are inducing cartilage destruction. Neutrophils also release neutrophil-active chemokines to recruit themselves in an autocrine manner to perpetuate tissue destruction. FcγR-expression on neutrophils is of crucial importance for the recognition of ICs.
METHODS
In this study, due to its high avidity for binding to FcγRs, we investigated the potential anti-inflammatory effect of a recombinant IgG1 Fc hexamer (rFc-µTP-L309C) on neutrophils in the K/BxN mouse model of endogenously generated chronic arthritis. 200 mg/kg rFc-µTP-L309C and human serum albumin (HSA), used as controls, were administered subcutaneously every other day. Mouse ankle joints were monitored daily to generate a clinical score. Immunohistology was used to evaluate neutrophil infiltration and TUNEL to assess apoptosis. ELISA was used to measure IL-1β.
RESULTS
Treatment with rFc-µTP-L309C, but not HSA, was able to significantly ameliorate the arthritis in the K/BxN mice. Significant neutrophil infiltration into the ankle joint was found, but treatment with rFc-µTP-L309C resulted in significantly less neutrophil infiltration. There was no significant influence of rFc-µTP-L309C on neutrophil death or apoptosis. Less neutrophil infiltration could not be correlated to chemokine-mediated migration. Significantly less IL-1β was measured in mice treated with rFc-µTP-L309C.
CONCLUSION
In the endogenous K/BxN mouse model of rheumatoid arthritis, amelioration can be explained in part by inhibition of neutrophil infiltration into the joints as well as inhibition of IL-1β production. Given the observed inhibitory properties on neutrophils, rFc-µTP-L309C may be a potential therapeutic candidate to treat autoimmune and inflammatory conditions in which neutrophils are the predominant cell type involved in pathogenesis
Intraoperative hyperspectral label-free imaging: from system design to first-in-patient translation.
Despite advances in intraoperative surgical imaging, reliable discrimination of critical tissue during surgery remains challenging. As a result, decisions with potentially life-changing consequences for patients are still based on the surgeon's subjective visual assessment. Hyperspectral imaging (HSI) provides a promising solution for objective intraoperative tissue characterisation, with the advantages of being non-contact, non-ionising and non-invasive. However, while its potential to aid surgical decision-making has been investigated for a range of applications, to date no real-time intraoperative HSI (iHSI) system has been presented that follows critical design considerations to ensure a satisfactory integration into the surgical workflow. By establishing functional and technical requirements of an intraoperative system for surgery, we present an iHSI system design that allows for real-time wide-field HSI and responsive surgical guidance in a highly constrained operating theatre. Two systems exploiting state-of-the-art industrial HSI cameras, respectively using linescan and snapshot imaging technology, were designed and investigated by performing assessments against established design criteria and ex vivo tissue experiments. Finally, we report the use of our real-time iHSI system in a clinical feasibility case study as part of a spinal fusion surgery. Our results demonstrate seamless integration into existing surgical workflows
Combination of bevacizumab and 2-weekly pegylated liposomal doxorubicin as first-line therapy for locally recurrent or metastatic breast cancer. A multicenter, single-arm phase II trial (SAKK 24/06)
Background: Pegylated liposomal doxorubicin (PLD) and bevacizumab are active agents in the treatment of metastatic breast cancer (MBC). We carried out a multicenter, single-arm phase II trial to evaluate the toxicity and efficacy of PLD and bevacizumab as first-line treatment in MBC patients. Methods: Bevacizumab (10 mg/kg) and PLD (20 mg/m2) were infused on days 1 and 15 of a 4-week cycle for a maximum of six cycles. Thereafter, bevacizumab monotherapy was continued at the same dose until progression or toxicity. The primary objective was safety and tolerability, and the secondary objective was to evaluate efficacy of the combination. Results: Thirty-nine of 43 patients were assessable for the primary end point. Eighteen of 39 patients (46%, 95% confidence interval 30% to 63%) had a grade 3 toxicity. Sixteen (41%) had grade 3 palmar-plantar erythrodysesthesia, one had grade 3 mucositis, and one severe cardiotoxicity. Secondary end point of overall response rate among 43 assessable patients was 21%. Conclusions: In this nonrandomized single-arm trial, the combination of bimonthly PLD and bevacizumab in locally recurrent and MBC patients demonstrated higher than anticipated toxicity while exhibiting only modest activity. Based on these results, we would not consider this combination for further investigation in this settin
Potential thresholds of critically increased cardiac-related spinal cord motion in degenerative cervical myelopathy.
INTRODUCTION
New diagnostic techniques are a substantial research focus in degenerative cervical myelopathy (DCM). This cross-sectional study determined the significance of cardiac-related spinal cord motion and the extent of spinal stenosis as indicators of mechanical strain on the cord.
METHODS
Eighty-four DCM patients underwent MRI/clinical assessments and were classified as MRI+ [T2-weighted (T2w) hyperintense lesion in MRI] or MRI- (no T2w-hyperintense lesion). Cord motion (displacement assessed by phase-contrast MRI) and spinal stenosis [adapted spinal canal occupation ratio (aSCOR)] were related to neurological (sensory/motor) and neurophysiological readouts [contact heat evoked potentials (CHEPs)] by receiver operating characteristic (ROC) analysis.
RESULTS
MRI+ patients (N = 31; 36.9%) were more impaired compared to MRI- patients (N = 53; 63.1%) based on the modified Japanese Orthopedic Association (mJOA) subscores for upper {MRI+ [median (Interquartile range)]: 4 (4-5); MRI-: 5 (5-5); p < 0.01} and lower extremity [MRI+: 6 (6-7); MRI-: 7 (6-7); p = 0.03] motor dysfunction and the monofilament score [MRI+: 21 (18-23); MRI-: 24 (22-24); p < 0.01]. Both patient groups showed similar extent of cord motion and stenosis. Only in the MRI- group displacement identified patients with pathologic assessments [trunk/lower extremity pin prick score (T/LEPP): AUC = 0.67, p = 0.03; CHEPs: AUC = 0.73, p = 0.01]. Cord motion thresholds: T/LEPP: 1.67 mm (sensitivity 84.6%, specificity 52.5%); CHEPs: 1.96 mm (sensitivity 83.3%, specificity 65.6%). The aSCOR failed to show any relation to the clinical assessments.
DISCUSSION
These findings affirm cord motion measurements as a promising additional biomarker to improve the clinical workup and to enable timely surgical treatment particularly in MRI- DCM patients.
