    N-Acetylcysteine an Allium Plant Compound Improves High-Sucrose Diet-Induced Obesity and Related Effects

    This study was designed to determine whether N-acetylcysteine (NAC, C5H9–NO3S), a compound from Allium species may be used as a complementary therapeutic agent, to inhibit high-sucrose induced-obesity and its effects on glucose tolerance, in vivo low-density lipoprotein (LDL)-oxidation and serum oxidative stress in rats. Initially, 24 male Wistar rats were divided into two groups: controls receiving standard chow (C, n = 6) and those receiving high-sucrose diet (HS, n = 18). After 22 days, (HS) group was divided into three groups (n = 6/group); (HS-HS) continued to eat high-sucrose diet and water; (HS-N) continued to eat high-sucrose diet and received 2 mg l−1-NAC in its drinking water; (HS-CN) changing high-sucrose to standard chow and receiving 2 mg l­1-NAC in its drinking water. After 22 days of the HS-group division (44 days of experimental period) body weight, body mass index and surface area were enhanced in HS-HS rats (P < .001). HS-HS rats had glucose intolerance, increased serum triacylglycerol (TG), very low-density lipoprotein (VLDL), oxidized-LDL (ox-LDL) and lipid-hydroperoxide (LH) than the others (P < .01). NAC in HS-N and HS-CN rats reduced the obesity markers, feed efficiency, LH and ox-LDL, as well normalized glucose response, TG and VLDL (P < .01) in these groups compared with HS-HS. Total antioxidant substances, GSH/GSSG ratio and glutathione-reductase, were higher in HS-N than in HS-HS (P < .01). In conclusion, NAC improved high-sucrose diet-induced obesity and its effects on glucose tolerance, lipid profile, in vivo LDL-oxidation and serum oxidative stress, enhancing antioxidant defences. The application of this agent may be feasible and beneficial for high-sucrose diet-induced obesity, which certainly would bring new insights on obesity-related adverse effects control

    Identification of Cell-Free Circulating MicroRNAs for the Detection of Early Breast Cancer and Molecular Subtyping

    Early detection is crucial for achieving a reduction in breast cancer mortality. Analysis of circulating cell-free microRNAs present in the serum of cancer patients has emerged as a promising new noninvasive biomarker for early detection of tumors and for predicting their molecular classifications. The rationale for this study was to identify subtype-specific molecular profiles of cell-free microRNAs for early detection of breast cancer in serum. Fifty-four early-stage breast cancers with 27 age-matched controls were selected for circulating microRNAs evaluation in the serum. The 54 cases were molecularly classified (luminal A, luminal B, luminal B Her2 positive, Her-2, triple negative). NanoString platform was used for digital detection and quantitation of 800 tagged microRNA probes and comparing the overall differences in serum microRNA expression from breast cancer cases with controls. We identified the 42 most significant (P ≤ 0.05, 1.5-fold) differentially expressed circulating microRNAs in each molecular subtype for further study. Of these microRNAs, 19 were significantly differentially expressed in patients presenting with luminal A, eight in the luminal B, ten in luminal B HER 2 positive, and four in the HER2 enriched subtype. AUC is high with suitable sensitivity and specificity. For the triple negative subtype miR-25-3p had the best accuracy. Predictive analysis of the mRNA targets suggests they encode proteins involved in molecular pathways such as cell adhesion, migration, and proliferation. This study identified subtype-specific molecular profiles of cell-free microRNAs suitable for early detection of breast cancer selected by comparison to the microRNA profile in serum for female controls without apparent risk of breast cancer. This molecular profile should be validated using larger cohort studies to confirm the potential of these miRNA for future use as early detection biomarkers that could avoid unnecessary biopsy in patients with a suspicion of breast cancer.Foundation for Research Support of the State of São Paulo (FAPESP process 2015/21082-0) and Public Ministry of Labor Campinas (Research, Prevention, and Education of Occupational Cancer)

    U?Pb ages and Hf-isotope data of detrital zircons from the late Neoarchean-Paleoproterozoic Minas Basin, SE Brazil.

    Because of its world-class iron ore deposits and promising Au and U mineralizations, the late Neoarchean to Paleoproterozoic Minas Basin (Minas Supergroup, SE of Brazil) is one of the best-studied basins in South America. However, the lack of datable interlayered volcanic rocks prevented discourse over ages of the strata, the sources and the nature of its ore deposits. In this paper, we present detrital zircon U? Pb age patterns coupled with Lu?Hf data for 18 samples, representing different stages of the Minas Basin evolution ( 2000 analyzed zircons). Age spectra for the main basal unit (Moeda Formation) show a classic rift-related detrital zircon pattern, characterized by multiple autochthonous sources, which in turn are much older than the age of deposition. Maximum age for the rifting event is constrained at ca. 2600 Ma. Detritus accumulated at the base of the Minas Supergroup were derived from Archean source rocks and their sedimentation was marked by differential uplift of the Archean crust, shortly after the 2730?2600 Ma high-K calc-alkaline magmatism (Mamona Event). The age of the BIF deposits is younger than 2600 Ma, most likely coinciding with the great oxygenation event between 2400 and 2200 Ma and the precipitation of banded iron deposits worldwide. Detrital zircons from the topmost units of the Minas strata suggest that tectonic inversion and closure of the basin took place at ca. 2120 Ma with the deposition of the synorogenic Sabar? Group. Rhyacian zircon supply showing juvenile Hf signatures gives evidence of a late Rhyacian amalgamation between the Mineiro Belt and the craton. The eHf signatures support the hypothesis that the Archean crystalline crust of the craton was mostly built by crust?mantle mixing processes, with a successive decrease of eHf values in zircons crystallized after 3250 Ma and minor mantle-like additions after Paleoarchean times.     Regras de associação aplicadas ao problema de evasão de alunos de cursos de pós-graduação (Lato Sensu) na modalidade EaD de uma instituição de ensino privada

    Um dos grandes desafios enfrentados pelo Ensino a Distância (EaD) é a desistência do aluno antes da conclusão do curso. Neste sentido, este problema tem sido abordado sobre os mais variados aspectos. Contudo, estudos sobre a evasão de cursos de pós-graduação EaD ainda são mais restritos quando comparados à cursos de graduação nesta mesma modalidade, notadamente devido a menor disponibilidade de dados. Este trabalho realiza a análise dos fatores que levaram à evasão de cursos de pós-graduação EaD de uma Instituição de Ensino privada que detém mais de 300 mil alunos. Para tanto, utilizaram-se ferramentas de estatística descritiva como também de Mineração de Dados, mais especificamente de Regras de Associação. Resultados apontam que fatores institucionais relacionados ao material didático e dificuldade com recursos tecnológicos foram alguns dos principais agentes causadores de evasão nos cursos

    Viabilidade de ovos de Ascaris lumbricoides após tratamento humano com medicamentos específicos: I— Levamisole e Pamoato de pirantel

    Com o objetivo de testar a viabilidade do ciclo biológico, 15 pacientes (masculinos, de quatro a 14 anos) com Ascaris lumbricoides, foram selecionados ao acaso. Após tratamento clássico com sais básicos de levamisole (7 pacientes) e de pamoato de pirantel (8), os ovos retirados das fêmeas expelidas ficaram incubados por 18 dias em de H2SO4 N/10. A seguir foram administrados per os a grupos de 5 camundongos por pacientes. Decorridos 8 dias da infecção, os animais foram sacrificados para pesquisas microscópica de larvas nos fragmentos pulmonares. Dos 75 animais, somente 1, pertencente ao grupo de tratados com levamisole, não apresentou larvas nos fragmentos pulmonares. Concluiu-se que as drogas, nas doses utilizadas, não possuem ação deletéria sobre os ovos de A. lumbricoides, mas promovem a eliminação de material infectivo, com possibilidade de incrementar a poluição onde vivem comunidades sem adequadas condições de saneamento básico
