Novel treatment approaches to combat trichomoniasis, a neglected and sexually transmitted infection caused by trichomonas vaginalis : translational perspectives

    Get PDF
The multistep translational science behind new drugs comprehends the entire process through laboratory, clinical, and community observations turned into health interventions. The development of new drug options from discovering targets and leading compounds in basic research for implementing therapeutic guidelines contributes to the emergence of health policies essential for infection control. This review updates the translational research in the scenario of the most common non-viral sexually transmitted infection (STI), trichomoniasis. Paradoxically to its high occurrence, it is considered neglected since notification is not mandatory. It turns into a stable disease with health complications, and receives little emphasis from public health programs to control STI. Although related to curable STIs, the current drugs, metronidazole and tinidazole, present therapeutic failures. The need for new options to treat trichomoniasis is established by basic research studies and patents revealing novel synthetic compounds and natural products presenting anti-Trichomonas vaginalis activities, mainly based on in vitro findings. Clinical trials are still focused on new routes of administration for conventional drugs. In addition, nanotechnology approaches are in their infancy, shedding light on potential possibilities for creating more effective, targeted, and safe delivery systems. Overall, the novel proposed approaches need, in addition to pharmaceutical development and efficacy assessments, to ensure that the quality requirements for their use as medicines are met. It is essential to overcome these issues to cross the “Death Valley” of drug discovery and to advance in the translational science criteria in the trichomoniasis drug development field

The Study of Heritage and Memory - NEP - the Federal University of Santa Maria develops research project about the japanese immigration in Santa Maria. From interviews with immigrants, realized the invisibility of this community in the state of Rio Grande do Sul. Aware of the undissociability between research and extension, the NEP has made the first exposition in honor of Japanese Immigration in Santa Maria, Rio Grande do Sul, celebrating 50 years of the arrival of the japaneses in the city and 100 years in Brazil. The exposition, temporary, and conducted in a space provided by the Secretariat of Culture of the city related the history of a group of people who dream with the quickly enrichment, but that ended up working on a semi- slave way in the city of Uruguaiana, west border of the State. The success of the exposition made the NEP to contact with other institutions of Japanese culture in the state to take the initiative of viabilization of a memorial to Japanese culture, once the actual state of Rio Grande do Sul does not have a museum or institution equivalent to preserving the history of the Japanese immigration.O Núcleo de Estudos do Patrimônio e Memória - NEP - da Universidade Federal de Santa Maria desenvolve um projeto de pesquisa sobre a imigração japonesa na cidade.

The potential therapeutic effect of guanosine after cortical focal ischemia in rats

Background and Purpose: Stroke is a devastating disease. Both excitotoxicity and oxidative stress play important roles in ischemic brain injury, along with harmful impacts on ischemic cerebral tissue. As guanosine plays an important neuroprotective role in the central nervous system, the purpose of this study was to evaluate the neuroprotective effects of guanosine and putative cerebral events following the onset of permanent focal cerebral ischemia. Methods: Permanent focal cerebral ischemia was induced in rats by thermocoagulation. Guanosine was administered immediately, 1 h, 3 h and 6 h after surgery. Behavioral performance was evaluated by cylinder testing for a period of 15 days after surgery. Brain oxidative stress parameters, including levels of ROS/RNS, lipid peroxidation, antioxidant nonenzymatic levels (GSH, vitamin C) and enzymatic parameters (SOD expression and activity and CAT activity), as well as glutamatergic parameters (EAAC1, GLAST and GLT1, glutamine synthetase) were analyzed. Results: After 24 h, ischemic injury resulted in impaired function of the forelimb, caused brain infarct and increased lipid peroxidation. Treatment with guanosine restored these parameters. Oxidative stress markers were affected by ischemic insult, demonstrated by increased ROS/RNS levels, increased SOD expression with reduced SOD activity and decreased nonenzymatic (GSH and vitamin C) antioxidant defenses. Guanosine prevented increased ROS/RNS levels, decreased SOD activity, further increased SOD expression, increased CAT activity and restored vitamin C levels. Ischemia also affected glutamatergic parameters, illustrated by increased EAAC1 levels and decreased GLT1 levels; guanosine reversed the decreased GLT1 levels and did not affect the EAAC1 levels. Conclusion: The effects of brain ischemia were strongly attenuated by guanosine administration. The cellular mechanisms involved in redox and glutamatergic homeostasis, which were both affected by the ischemic insult, were also modulated by guanosine. These observations reveal that guanosine may represent a potential therapeutic agent in cerebral ischemia by preventing oxidative stress and excitotoxicity

The present study aimed to compare the composition on the label of dry complete food for adult cats with the nutritional composition determined by chemical analysis, to determine the occurrence of aflatoxins, and to list the most used ingredients. The determined parameters were: crude protein (CP), ether extract in acid hydrolysis (EEA), mineral matter (MM), moisture (MO), crude fiber (CF), calcium (Ca), phosphorus (P), sodium (Na), calculated metabolizable energy (ME) and aflatoxins. In assessing the levels guaranteed on the label, MO, CP, CF, MM and P remained within the range, but with a variation on the limits. The metabolizable energy varied from 3,712 to 4,136 kcal/kg. There was a divergence in the levels of two diets, with Ca above and EE below the values declared on the label in relation to composition obtained by chemical analysis, and both non-compliant in 20% foods tested. Regarding sodium, the diets showed levels 130 - 384% higher than stated on the label. Aflatoxin was found in 90% samples from zero to 4.37 ppb. The wide variation observed in the composition within the same super premium segment can cause losses due to the lack or excess of essential nutrients for the animal
