    Age-Related Olfactory Decline Is Associated With Levels of Exercise and Non-exercise Physical Activities

    Objective: This cross-sectional study evaluates the impact of active or non-active lifestyle in terms of physical, cognitive and social activity on the olfactory function in Elderly Subjects (ES) and aims at looking for a correlation between the time devoted to life activities and the score obtained during the olfactory tests by each individual. Methods: One hundred and twenty-two elderly volunteers were recruited in Sardinia (Italy) and divided into active ES (n = 60; 17 men, 43 women; age 67.8 ± 1.12 years) and inactive ES (n = 62; 21 men, 41 women, age 71.1 ± 1.14 years) based on their daily physical activities. The olfactory function was evaluated using the “Sniffin’s Sticks” battery test, while the assessment of daily activities was made by means of personal interviews. Results: A significant effect of active or inactive lifestyle was found on the olfactory function of ES (F(1,120) > 10.16; p < 0.005). A positive correlation was found between the olfactory scores and the number of hours per week dedicated to physical activities (Pearson’s r > 0.32, p ≤ 0.014) in both active and inactive ES. Conclusions: High levels of exercise and non-exercise physical activity are strongly associated with the olfactory function and, consequently, with the quality of life of the elderly. Given the limited physical exercise of elderly people, they can benefit from a more active lifestyle by increasing non-exercise physical activities

    Association among Olfactory Function, Lifestyle and BMI in Female and Male Elderly Subjects: A Cross-Sectional Study

    Physical activities seem to counteract the age-related physiological decline of the olfactory function which, influencing the food choices and eating behavior, can affect the body weight of individuals. The main purpose of this cross-sectional study was to evaluate the relationships between olfactory function and BMI in female and male Elderly Subjects (ES), according to the level of their lifestyle activities in physical, cognitive, and social terms. Considering weekly physical activities, the adult elderlies who decided to participate in this study were divided into active ES (n = 65) and non-active ES (n = 68). Assessment of weekly activities and olfactory function were performed by means of face-to-face interviews and the "Sniffin' Sticks" battery test, respectively. The results show that ES who are overweight and with a non-active lifestyle achieved lower TDI olfactory scores than normal weight ES and those classified as active. Hyposmic and non-active ES showed a higher BMI than normosmic and active ES. Sex-related differences, with females performing better than males, were evident in the presence of at least one of the following conditions: non-activity, hyposmia, or overweight. Inverse correlations were found between BMI and TDI olfactory score and between BMI and hours/week spent on physical activities, both when subjects were considered all together and when they were divided into females and males. These findings suggest that a higher BMI is related to the olfactory dysfunction linked to active or non-active lifestyle and the sex-related differences, and the condition of hyposmia is related to the increase in body weight due to lifestyle and sex differences. Given that the relationship between BMI and non-exercise physical activities is comparable to that between BMI and exercise physical activities, and this may be of particular importance for ES with limited mobility

    The Human Nose as a Chemical Sensor in the Perception of Coffee Aroma: Individual Variability

    The flavor of foods and beverages is generally composed of a mixture of volatile compounds, however not all the molecules that form an aroma are sensorially relevant. The odor-active compounds present in a mixture are different for each subject, both in quantitative and qualitative terms. This means that the ability of the human nose to act as a chemical sensor varies among individuals. In this study, we used the headspace of roasted coffee beans as a complex olfactory stimulus and, by means of the coupled Gas Chromatography-Olfactometry (GC-O) technique, the single components of coffee flavor were separated. Each subject, previously classified for his/her olfactory status (normosmic, hyposmic or anosmic) by means of the Sniffin' Sticks battery (composed of Threshold, Discrimination and Identification subtests), had to identify and evaluate each smelled molecule. The results show that the individual ability to detect individual compounds during the GC-O experiments and the odor intensity reported during the sniffing of pen #10 (the pen of the identification test) containing coffee aroma were related to TDI olfactory status (based on the score obtained from the sum composed of Threshold, Discrimination and Identification scores). We also found that the number of total molecules and of molecules smelling of coffee is linearly related to the TDI olfactory score. Finally, the odor intensity reported when sniffing pen #10 containing coffee aroma is positively correlated with the number of molecules detected and the average intensity reported. In conclusion, our findings show that the human perception of both individual compounds and complex odors is strongly conditioned by the olfactory function of subjects

    A polymorphism in the human gene encoding OBPIIa affects the perceived intensity of smelled odors

    Among the factors that contribute to the physiological variability of the olfactory function of individuals, an important role seems to be played by the OBPs present in the mucus that bathes the ciliated terminals of the olfactory sensory neurons, facilitating the access of odorants to the olfactory receptors. It was recently highlighted that the rs2590498 polymorphism in the odor binding-protein (OBPIIa) gene it is associated with the olfactory threshold in healthy individuals. Aim of this study was to evaluate: 1) the presence of a relationship between the threshold olfactory performance of healthy subjects and the intensity with which they perceive the smelled odorants, and 2) the effect of the rs2590498 polymorphism of the OBPIIa gene on perceived intensity. We found a positive correlation between threshold olfactory and perceived intensity, and that AA homozygous subjects reported a perceived intensity higher than heterozygous and GG homozygous subjects. By showing a positive effect of the rs2590498 polymorphism of the hOBPIIa gene on the intensity perceived, these results suggest that it allows a larger number of molecules in an odorous mixture to reach the olfactory receptors

    Role of inflammation and oxidative stress in post-menopausal oestrogen-dependent breast cancer

    Weight gain and obesity are among the most important risk factors for post-menopausal oestrogen-dependent breast cancer (EDBC). Weight gain is associated with oxidative stress, which in turn promotes breast cancer progression. We carried out a prospective study in 216 consecutive post-menopausal breast cancer patients aiming to examine the correlations between traditional prognostic factors (tumour size, T, nodal, N, grading, G, and metastasis status, M), and body mass index (BMI), leptin, pro-inflammatory cytokines (Interleukin, IL,-6 and tumour necrosis factor-alpha, TNF-α), and oxidative stress (reactive oxygen species, ROS, glutathione peroxidase, GPx, superoxide dismutase, SOD) among patients with oestrogen receptor (ER)+ and ER- breast cancers. Distribution of T, N and M categories did not differ between ER+ and ER- breast cancer patients. ER- patients showed a higher incidence of G3 tumours. Weight, BMI, leptin, IL-6 and ROS were higher in ER+ compared with ER- patients. Among ER+ patients, BMI, leptin, IL-6 and ROS correlated with T and M. Leptin, IL-6 and ROS were positively correlated also with N. Among ER- patients, BMI and leptin did not correlate with any of prognostic parameters, whereas a positive correlation between IL-6, ROS and M was found. Multivariate regression analysis showed that BMI, leptin, IL-6 and ROS were predictive for T, N and M in ER+ patients. Weight gain, inflammation and oxidative stress are involved in EDBC prognosis. Their modulation through antidiabetic, anti-inflammatory and antioxidants drugs combined with endocrine therapy may constitute a targeted approach in post-menopausal EDBC

    Gustatory Sensitivity and Food Acceptance in Two Phylogenetically Closely Related Papilionid Species: Papilio hospiton and Papilio machaon.

    In herbivorous insects, food selection depends on sensitivity to specific chemical stimuli from host-plants as well as to secondary metabolites (bitter) and to sugars (phagostimulatory). Bitter compounds are noxious, unpalatable or both and evoke an aversive feeding response. Instead, sugars and sugar alcohols play a critical role in determining and enhancing the palatability of foods. We assumed that peripheral taste sensitivity may be related to the width of the host selection. Our model consists of two closely phylogenetically related Papilionid species exhibiting a difference in host plant choice: Papilio hospiton and Papilio machaon. The spike activity of the lateral and medial maxillary styloconic taste sensilla was recorded following stimulation with several carbohydrates, nicotine and NaCl, with the aim of characterizing their gustatory receptor neurons and of comparing their response patterns in the light of their different acceptability in feeding behaviour. The results show that: a) each sensillum houses phagostimulant and phagodeterrent cells; b) the spike activity of the gustatory neurons in response to different taste stimuli is higher in P. hospiton than in P. machaon; c) sugar solutions inhibit the spike activity of the deterrent and salt cells, and the suppression is higher in P. machaon than in P. hospiton. In conclusion, we propose that the different balance between the phagostimulant and phagodeterrent inputs from GRNs of maxillary sensilla may contribute in determining the difference in food choice and host range

    Immune infiltrating cells in duodenal cancers

    Duodenal adenocarcinoma (DA) is a rare yet aggressive malignancy, with increasing incidence in the last decades. Its low frequency has hampered a thorough understanding of the pathogenesis of the disease and of its biology, limiting the identification of tailored therapeutic options. A large body of evidence has clearly shown the clinical relevance of immune cells in solid tumors, correlating immune features with post-surgical prognosis. The aim of this study was to analyze the immune contexture in a cohort of duodenal adenocarcinomas surgically resected at our Institution and define its correlation with clinical variables. Methods Tissue slides from paraffin-embedded tumor specimens of 15 consecutive DA and 3 adenomas that underwent a pancreaticoduodenectomy in our center between 2010 to 2018 were immunohistochemically stained. The density (percentage of immune reactive area, IRA%) of immune markers CD45RO, CD8, CD20, IL-17, PD-1, CD68 was quantified by computer-assisted image analysis. Demographic, clinical, histopathological data were collected. Results In our population, median IRA % (IQR) of immune subsets was respectively CD45RO-TILs 2.19 (2.14), CD8-TIL 0.42 (0.81), CD20-TILs 0.22 (0.51), CD20-TLT 2.84 (4.64), CD68-TAM 2.19 (1.56), IL17+ cells 0.39 (0.39), PD1-TILs 0.19 (0.41). The median follow-up was 47.5 (22.4\u201363.3) months. At statistical analysis, the density of CD8-TILs inversely correlated with lymph node ratio (p\u2009=\u20090.013), number of metastatic lymph nodes (p\u2009=\u20090.019), and was lower in N+\u2009adenocarcinomas compared to N0 (1.07 vs 0.29; p\u2009=\u20090.093), albeit not significantly. Stratifying patients for the N status, the density of CD8-TILs decreased with the increasing of the N stage (p\u2009=\u20090.065) and was lower in patients who experienced recurrence and died for the disease (0.276 vs 0.641; p\u2009=\u20090.044). Notably, also CD68-TAM distribution was different in patients who had recurrence versus patients who did not (1.028 vs 2.276; p\u2009=\u20090.036). Conclusions Immune cells showed variable expression in correlation with common prognostic factors, suggesting T cell infiltration may play a protective role towards lymphatic spread of disease and nodal metastatization. Furthermore, T cell density and macrophage infiltration were associated to a lower risk of recurrence and disease related death. A multicentric approach may be indicated to allow analysis of larger cohorts of patients, potentially increasing the power of our observations

    An automated system for the objective evaluation of human gustatory sensitivity using tongue biopotential recordings

    The goal of this work is to develop an automatic system for the evaluation of the gustatory sensitivity of patients using an electrophysiological recording of the response of bud cells to taste stimuli. In particular, the study aims to evaluate the effectiveness and limitations of supervised classifiers in the discrimination between subjects belonging to the three 6-n-pro-pylthiouracil (PROP) taster categories (supertasters, medium tasters, and non-tasters), exploiting features extracted from electrophysiological recordings of the tongue. Thirty-nine subjects (equally divided into the three PROP status classes by standard non-objective scaling methods) underwent a non-invasive, differential, biopotential recording of their tongues during stimulation with PROP by using a custom-made, flexible, silver electrode. Two different classifiers were trained to recognize up to seven different features extracted from the recorded depolarization signal. The classification results indicate that the identified set of features allows to distinguish between PROP tasters and non-tasters (average accuracy of 80% ± 18% and up to 94% ± 15% when only supertasters and non-tasters are considered), but medium tasters were difficult to identify. However, these apparent classification errors are related to uncertainty in the labeling procedures, which are based on non-objective tests, in which the subjects provided borderline evaluations. Thus, using the proposed method, it is possible, for the first time, to automatically achieve objective PROP taster status identification with high accuracy. The simplicity of the recording technique allows for easy reproduction of the experimental setting; thus the technique can be used in future studies to evaluate other gustatory stimuli. The proposed approach represents the first objective and automatic method to directly measure human gustatory responses and a milestone for physiological taste studies, with applications ranging from basic science to food tasting evaluations

    Archaeal Sources of Intact Membrane Lipid Biomarkers in the Oxygen Deficient Zone of the Eastern Tropical South Pacific

    Archaea are ubiquitous in the modern ocean where they are involved in the carbon and nitrogen biogeochemical cycles. However, the majority of Archaea remain uncultured. Archaeal specific membrane intact polar lipids (IPLs) are biomarkers of the presence and abundance of living cells. They comprise archaeol and glycerol dibiphytanyl glycerol tetraethers (GDGTs) attached to various polar headgroups. However, little is known of the IPLs of uncultured marine Archaea, complicating their use as biomarkers. Here, we analyzed suspended particulate matter (SPM) obtained in high depth resolution from a coastal and open ocean site in the eastern tropical South Pacific (ETSP) oxygen deficient zone (ODZ) with the aim of determining possible biological sources of archaeal IPL by comparing their composition by Ultra High Pressure Liquid Chromatography coupled to high resolution mass spectrometry with the archaeal diversity by 16S rRNA gene amplicon sequencing and their abundance by quantitative PCR. Thaumarchaeotal Marine Group I (MGI) closely related to Ca. Nitrosopelagicus and Nitrosopumilus dominated the oxic surface and upper ODZ water together with Marine Euryarchaeota Group II (MGII). High relative abundance of hexose phosphohexose- (HPH) crenarchaeol, the specific biomarker for living Thaumarchaeota, and HPH-GDGT-0, dihexose- (DH) GDGT-3 and -4 were detected in these water masses. Within the ODZ, DPANN (Diapherotrites, Parvarchaeota, Aenigmarchaeota, Nanoarchaeota, and Nanohaloarchaea) of the Woesearchaeota DHVE-6 group and Marine Euryarchaeota Group III (MGIII) were present together with a higher proportion of archaeol-based IPLs, which were likely made by MGIII, since DPANN archaea are supposedly unable to synthesize their own IPLs and possibly have a symbiotic or parasitic partnership with MGIII. Finally, in deep suboxic/oxic waters a different MGI population occurred with HPH-GDGT-1, -2 and DH-GDGT-0 and -crenarchaeol, indicating that here MGI synthesize membranes with IPLs in a different relative abundance which could be attributed to the different detected population or to an environmental adaptation. Our study sheds light on the complex archaeal community of one of the most prominent ODZs and on the IPL biomarkers they potentially synthesize
