    Application of diagnostics for mosquito vector incrimination and innovative biotechnology-based methods for disease control

    Τα κουνούπια, ειδικότερα είδη των γενών Aedes, Anopheles και Culex, είναι οι πιο κοινοί φορείς ασθενειώνπαγκοσμίως, προκαλώντας περισσότερα από 500 εκατομμύρια ανθρώπινα κρούσματα ετησίως, εξαιτίας της μετάδοσης παθογόνων, όπως το παράσιτο της ελονοσίας, ο Δάγκειος ιός, ο ιός του Δυτικού Νείλου, ο ιός Zika, κ.λπ. Η πλειοψηφία των κρουσμάτων συμβαίνει στην Αφρική, την Κεντρική και Λατινική Αμερική και την νοτιοανατολική Ασία, ωστόσο, τις τελευταίες 2 δεκαετίες, ορισμένες ασθένειες έχουν (επαν)εμφανιστεί και στην Ευρώπη. Η καταπολέμηση των εντομομεταδιδόμενων ασθενειών βασίζεται σε μεγάλο βαθμό στον έλεγχο του εντόμου-φορέα, κυρίως με τη χρήση εντομοκτόνων (π.χ. πυρεθροειδή, οργανοφωσφορικά, ρυθμιστές ανάπτυξης). Όμως, ο περιορισμένος αριθμός εγκεκριμένων εντομοκτόνων σε συνδυασμό με τηναυξανόμενη εμφάνιση ανθεκτικότητας σ’αυτά τονίζουν τη σημασία της συνεχούς εντομολογικής επιτήρησης. Ο αριθμός τέτοιων μελετών για την Ελλάδα είναι μικρός. Το είδος Aedesalbopictus, γνωστό ως Ασιατικό κουνούπι τίγρης, αποτελεί σημαντικό κίνδυνο για τη δημόσια υγεία, με παγκόσμια διασπορά. Η παρουσία του στην Ελλάδα είναι ευρεία και, πιθανώς, συνδεδεμένη με τις πρόσφατες επιδημίες του ιού του Δυτικού Νείλου. Το πρώτο μέρος της παρούσας μεταπτυχιακής μελέτης αφορά στην ανάλυση της ανθεκτικότητας σε εντομοκτόνα πληθυσμών Ae. albopictusαπό διαφορετικές περιοχές της Ελλάδας, με τη χρήση μοριακών διαγνωστικών εργαλείων. Η μεταλλαγή F1534C στο γονίδιο που κωδικοποιεί για το κανάλι νατρίου, που προσδίδει ανθεκτικότητα στα πυρεθροειδή, βρέθηκε σε όλες τις δειγματοληπτικές περιοχές, με υψηλότερη συχνότητα στην Αθήνα, ενώ η Ι1532Τ στο ίδιο γονίδιο εντοπίστηκε για πρώτη φορά στη χώρα μας, με σχετικά χαμηλή συχνότητα. Ανθεκτικότητα στο temephos, λόγω υπερέκφρασης των καρβοξυλεστερασών 3 και 6, βρέθηκε επίσης διαδεδομένη. Τέλος, δεν ανιχνεύθηκε κάποια μεταλλαγή σχετιζόμενη με ανθεκτικότητα στο προνυμφοκτόνο diflubenzuron. Οι περιορισμοί που προκύπτουν από την εκτενή χρήση εντομοκτόνων υποδεικνύουν την ανάγκη εφαρμογής ολοκληρωμένης στρατηγικής ελέγχου των κουνουπιών-φορέων. Στο δεύτερο τμήμα της συγκεκριμένης εργασίας, μελετήθηκεμια μέθοδος γενετικού ελέγχου μεταδιδόμενων ασθενειών: πολυ-αμιδο-αμίνες (PAAs) αξιολογήθηκαν ως πολυμερικοί φορείς δίκλωνου RNA (dsRNA) στο κουνούπι An. gambiae, με σκοπό τη σίγηση γονιδίων (RNAi) του φορέα που υποβοηθούν τη διέλευση του παρασίτου της ελονοσίας από το έντερό του. dsRNA εναντίον των γονιδίων CACTUS και ESP του κουνουπιού συζεύκτηκαν με PAAs και εξετάστηκε η σταθερότητα των νανοσωματιδίων σε αναγωγικό περιβάλλον. Εν συνεχεία, τα νανοσωματίδια χορηγήθηκαν σε ενήλικα κουνούπια μέσω τροφής ή εμβολίου. Η σίγηση των γονιδίων-στόχων στο έντερο, ήταν ικανοποιητική, ως πρώτη ένδειξη. Αυτά τα προκαταρκτικά αποτελέσματα θα αξιοποιηθούν ως θεμέλια στα επόμενα πειραματικά βήματα με ορισμένες κάποιες βελτιώσεις/ τροποποιήσεις.Mosquitoes, especially species of the genera Aedes, Anopheles and Culex, are the most common disease vectorsworldwide, responsible for more than 500 million human cases annually, due to malaria, Dengue virus, West Nile virus, Zika virus, etc., transmission. Mosquito-borne diseases occur mainly in Africa, Central and Latin America and southeastern Asia, but during the last two decades they have (re-)emerged also in Europe. Prevention of mosquito-borne diseases depends to a great extent on vector control; insecticides (e.g. pyrethroids, organophosphates, growth regulators, etc) are the most powerful tool. However, the limited number of available formulations along with the increasing occurence of insecticide resistance highlight the importance of continuous surveillance and evidence-based control. There is a small number of such recent studies in Greece. Aedes albopictus, commonly known as the Asian tiger mosquito, is a major risk for public health, being globally dispersed. Its presence in Greece is also wide and potentially associated with West Nile virus circulation, over the last years. Therefore, we conducted an extended analysis on the insecticide resistance status of Ae. albopictus populations throughout Greece, using molecular diagnostic techniques. Target-site resistance mutation F1534C in the vssc gene leading to pyrethroid resistance was spread across the country, with the highest frequency in Athens, while mutation vssc I1532T was reported for the first time in Greece, in low frequencies. Metabolic resistance to temephos attributed to carboxylesterases 3 and 6 overexpression was also recorded extensively in many sampling areas. No mutation correlated with resistance to the larvicide diflubenzuron was detected. Limitations arisen by the extended use of insecticides determine the necessity of integrated vector control methods. In the second part of the present study, RNAi was used as a gene silencing mechanism to block malaria parasite transmission to humans; polyamidoamines (PAAs) were evaluated as polymeric carriers of dsRNA molecules in Anopheles gambiae, against mosquito genes that promote Plasmodium invasion of the midgut. dsRNAs against CACTUS and ESP genes were conjugated to PAAs. Nanoparticle stability was estimated, and then they were administered to adult mosquitoes both by injecting and feeding them. A moderate knock-down efficiency of the nanoparticles against the genes expression in the mosquito midgut was measured. Many experiment modifications are required and our findings are still preliminary, but of fundamental importance for the next steps

    A Practical Insecticide Resistance Monitoring Bioassay for Orally Ingested Dinotefuran in Anopheles Malaria Vectors

    Attractive Toxic Sugar Baits (ATSB) deployed outdoors are likely to be particularly effective against outdoor biting mosquitoes and, if they contain insecticides with a different mode of action, mosquitoes resistant to pyrethroids. One such ATSB based on the neonicotinoid dinotefuran is currently under evaluation in Africa. As with any insecticide-based intervention, it will be important to monitor for the possible emergence of vector resistance. While methods for detecting resistance to insecticides via tarsal contact are recommended by the World Health Organization (WHO), these may not be applicable for orally ingested insecticides. Here, a new ingestion assay, appropriate for a controlled laboratory setting, is described using fluorescein sodium salt (uranine) as a feeding marker. Conventional topical application bioassays, more appropriate for routine deployment, have also been used to apply dinotefuran to the thorax of adult Anopheles mosquitoes with an organic carrier to bypass lipid cuticle barriers. The two methods were compared by establishing lethal doses (LD) in several Anopheles strains. The similarity of the ratios of susceptibility to dinotefuran between pairs of pyrethroid susceptible and resistant strains validates topical application as a suitable, more practical and field applicable method for monitoring for the emergence of resistance to orally ingested dinotefuran. A discriminating dose is proposed, which will be further validated against field populations and used to routinely monitor for the emergence of resistance alongside ATSB trial

    Bioassay and molecular monitoring of insecticide resistance status in Aedes albopictus populations from Greece, to support evidence-based vector control

    Background Aedes albopictus has a well-established presence in southern European countries, associated with recent disease outbreaks (e.g. chikungunya). Development of insecticide resistance in the vector is a major concern as its control mainly relies on the use of biocides. Data on the species’ resistance status are essential for efficient and sustainable control. To date the insecticide resistance status of Ae. albopictus populations from Greece against major insecticides used in vector control remains largely unknown. Methods We investigated the insecticide resistance status of 19 Ae. albopictus populations from 11 regions of Greece. Bioassays were performed against diflubenzuron (DFB), Bacillus thuringiensis var. israelensis (Bti), deltamethrin and malathion. Known insecticide resistance loci were molecularly analysed, i.e. voltage-gated sodium channel (VGSC) mutations associated with pyrethroid resistance; presence and frequency of carboxylesterases 3 (CCEae3a) and 6 (CCEae6a) gene amplification associated with organophosphate (OP) resistance and; chitin synthase-1 (CHS-1) for the possible presence of DFB resistance mutations. Results Bioassays showed full susceptibility to DFB, Bti and deltamethrin, but resistance against the OP malathion (range of mortality: 55.30–91.40%). VGSC analysis revealed a widespread distribution of the mutations F1534C (in all populations, with allelic frequencies between 6.6–68.3%), and I1532T (in 6 populations; allelic frequencies below 22.70%), but absence of V1016G. CCE gene amplifications were recorded in 8 out of 11 populations (overall frequency: 33%). Co-presence of the F1534C mutation and CCEae3a amplification was reported in 39 of the 156 samples analysed by both assays. No mutations at the CHS-1 I1043 locus were detected. Conclusions The results indicate: (i) the suitability of larvicides DFB and Bti for Ae. albopictus control in Greece; (ii) possible incipient pyrethroid resistance due to the presence of kdr mutations; and (iii) possible reduced efficacy of OPs, in a scenario of re-introducing them for vector control. The study highlights the need for systematic resistance monitoring for developing and implementing appropriate evidence-based control programmes

    Monitoring of Insecticide Resistance Mutations and Pathogen Circulation in Sand Flies from Emilia-Romagna, a Leishmaniasis Endemic Region of Northern Italy

    The continuously expanding distribution of sand flies, proven vectors of Leishmania and of several phleboviruses, is a growing public health issue in Europe. Especially in Italy, visceral leishmaniasis (VL) is occurring with increasing incidence northward, in previously non-endemic provinces. Around the globe, disease elimination efforts largely focus on sand fly vector insecticidal control, often leading to the development of resistance. In Emilia-Romagna (ER), northern Italy, insecticides are heavily applied for agricultural and mosquito control, but not specifically against sand flies. Here, we investigated the sand fly species composition in certain environmental settings in ER provinces and monitored the presence of pyrethroid resistance mutations and pathogen circulation. Phlebotomus perfiliewi, a dominant vector of Leishmania infantum, was detected almost exclusively in the region. No mutations in the voltage-gated sodium channel gene, e.g., knock-down resistance mutations I1011M, L1014F/S, V1016G, or F1020S, were recorded. Pathogen monitoring revealed that almost 40% of the tested sand fly pools were positive for Leishmania, while the presence of Toscana and Fermo phleboviruses was also observed in much lower frequencies (≤3% positive pools). Regular epidemiological and entomological monitoring, alongside resistance surveillance, is highly recommended to ensure the sustainability and efficiency of vector control interventions

    Molecular monitoring of insecticide resistance in major disease vectors in Armenia

    Abstract Background Armenia is considered particularly vulnerable to life-threatening vector-borne diseases (VBDs) including malaria, West Nile virus disease and leishmaniasis. However, information relevant for the control of the vectors of these diseases, such as their insecticide resistance profile, is scarce. The present study was conducted to provide the first evidence on insecticide resistance mechanisms circulating in major mosquito and sand fly populations in Armenia. Methods Sampling sites were targeted based mainly on previous historical records of VBD occurrences in humans and vertebrate hosts. Initially, molecular species identification on the collected vector samples was performed. Subsequently, molecular diagnostic assays [polymerase chain reaction (PCR), Sanger sequencing, PCR-restriction fragment length polymorphism (RFLP), quantitative PCR (qPCR)] were performed to profile for major insecticide resistance mechanisms, i.e. target site insensitivity in voltage-gated sodium channel (vgsc) associated with pyrethroid resistance, acetylcholinesterase (ace-1) target site mutations linked to organophosphate (OP) and carbamate (CRB) resistance, chitin synthase (chs-1) target site mutations associated with diflubenzuron (DFB) resistance and gene amplification of carboxylesterases (CCEs) associated with resistance to the OP temephos. Results Anopheles mosquitoes were principally represented by Anopheles sacharovi, a well-known malaria vector in Armenia, which showed no signs of resistance mechanisms. Contrarily, the knockdown resistance (kdr) mutations V1016G and L1014F/C in the vgsc gene were detected in the arboviral mosquito vectors Aedes albopictus and Culex pipiens, respectively. The kdr mutation L1014S was also detected in the sand fly, vectors of leishmaniasis, Phlebotomus papatasi and P. tobbi, whereas no mutations were found in the remaining collected sand fly species, P. sergenti, P. perfiliewi and P. caucasicus. Conclusions This is the first study to report on molecular mechanisms of insecticide resistance circulating in major mosquito and sand fly disease vectors in Armenia and highlights the need for the establishment of systematic resistance monitoring practices for the implementation of evidence-based control applications.     The Implementation of a Health Care Worker Screening Program Based on the Advanta RT-qPCR Saliva Assay in a Tertiary Care Referral Hospital in Northern Greece

    Health care workers are at increased risk of acquiring SARS-CoV-2 infection due to different exposures in the community and in hospital settings. Interventions implemented to avoid nosocomial outbreaks include preventive testing strategies. In this report, we present results from the mass screening program applied in our hospital to all professionals, irrespective of symptoms or risk of exposure. We processed saliva specimens with real-time reverse transcription polymerase chain reaction. The total number of samples received was 43,726. Positive results were 672 and average positivity rate was 1.21%. The average positivity rate was similar to the positivity rate in the community in Greece and EU. More specifically, 80.5% of the positive participants care for patients in their daily activities, 31% experienced no symptoms before receiving the positive result, 46.1% reported a close contact with a patient or infected coworkers and 32.8% reported a close contact with infected family members. We believe that the identification of asymptomatic carriers has proved the effectiveness of the screening program by preventing the putative nosocomial spread of the virus and the depletion of workforce. In conclusion, in times of high incidence in the community, the periodic testing of health care personnel is wise and relevant for implementation costs

    A Novel Real-Time RT-PCR-Based Methodology for the Preliminary Typing of SARS-CoV-2 Variants, Employing Non-Extendable LNA Oligonucleotides and Three Signature Mutations at the Spike Protein Receptor-Binding Domain

    Mutations resulting in amino-acid substitutions of the SARS-CoV-2 spike protein receptor-binding domain (RBD) have been associated with enhanced transmissibility and immune escape of the respective variants, namely Alpha, Beta, Gamma or Delta. Rapid identification of the aforementioned variants of concern and their discrimination of other variants is thus of importance for public health interventions. For this reason, a one-step real-time RT-PCR assay employing four locked nucleic acid (LNA) modified TaqMan probes was developed, to target signature mutations associated with amino-acid substitutions at positions 478, 484 and 501 present in the receptor-binding motif (RBM) of the spike protein RBD. This region contains most contacting residues of SARS-CoV-2 that bind to ACE2. A novel strategy employing the use of non-extendable LNA oligonucleotide blockers that can reduce non-specific hybridization of probes increased the number of different mutated sites examined in a multiplex PCR. The combinatory analysis of the different fluorescence signals obtained enabled the preliminary differentiation of SARS-CoV-2 variants of concern. The assay is sensitive with a LOD of 263 copies/reaction for the Delta variant, 170 copies/reaction for the Beta variant, amplification efficiencies > 91% and a linear range of >5 log10 copies/reaction against all targets. Validation of the assay using known SARS-CoV-2-positive and negative samples from humans and animals revealed its ability to correctly identify the targeted mutations and preliminary characterize the SARS-CoV-2 variants. The novel approach for mutation typing using LNA oligonucleotide blockers can be modified to target signature mutations at four different sites in the RBM and further expand the range of variants detected

    A Toxoplasma gondii oxopurine transporter binds nucleobases and nucleosides using different binding modes

    Toxoplasma gondii is unable to synthesize purines de novo, instead salvages them from its environment, inside the host cell, for which they need high affinity carriers. Here, we report the expression of a T. gondii Equilibrative Nucleoside Transporter, Tg244440, in a Trypanosoma brucei strain from which nucleobase transporters have been deleted. Tg244440 transported hypoxanthine and guanine with similar affinity (Km ~1 µM), while inosine and guanosine displayed Ki values of 4.05 and 3.30 µM, respectively. Low affinity was observed for adenosine, adenine, and pyrimidines, classifying Tg244440 as a high affinity oxopurine transporter. Purine analogues were used to probe the substrate-transporter binding interactions, culminating in quantitative models showing different binding modes for oxopurine bases, oxopurine nucleosides, and adenosine. Hypoxanthine and guanine interacted through protonated N1 and N9, and through unprotonated N3 and N7 of the purine ring, whereas inosine and guanosine mostly employed the ribose hydroxy groups for binding, in addition to N1H of the nucleobase. Conversely, the ribose moiety of adenosine barely made any contribution to binding. Tg244440 is the first gene identified to encode a high affinity oxopurine transporter in T. gondii and, to the best of our knowledge, the first purine transporter to employ different binding modes for nucleosides and nucleobases

    High insecticide resistance mediated by different mechanisms in Culex quinquefasciatus populations from the city of Yaoundé, Cameroon

    Culex mosquitoes particularly Culex quinquefasciatus are important arboviral and filariasis vectors, however despite this important epidemiological role, there is still a paucity of data on their bionomics. The present study was undertaken to assess the insecticide resistance status of Cx. quinquefasciatus populations from four districts of Yaoundé (Cameroon). All Culex quinquefasciatus populations except one displayed high resistance to bendiocarb and malathion with mortalities ranging from 0-89% while high resistance intensity against both permethrin and deltamethrin was recorded. Molecular analyses revealed high frequencies of the ACE-1 G119S mutation (ranging from 0-33%) and kdr L1014F allele (ranging from 55-74%) in all Cx. quinquefasciatus populations. Significant overexpression was detected for cytochrome P450s genes CYP6AA7 and CYP6Z10, as well as for Esterase A and Esterase B genes. The total cuticular hydrocarbon content, a proxy of cuticular resistance, was significantly increased (compared to the S-lab strain) in one population. The study confirms strong insecticide resistance mediated by different mechanisms in Cx. quinquefasciatus populations from the city of Yaoundé. The expansion of insecticide resistance in Culex populations could affect the effectiveness of current vector control measures and stress the need for the implementation of integrated vector control strategies in urban settings.
