    Inhibition of proliferation and differentiation during early T cell development by anti-transferrin receptor antibody

    Proliferating cells require iron and, therefore, express the transferrin receptor (CD71) that mediates cellular iron uptake. Cycling thymocytes, which have the CD4−8−3−, CD4−8+3−, or CD4+8+3− phenotypes, also express CD71. The importance of CD71-mediated iron uptake for proliferation and maturation of thymocytes was studied using fetal thymus organ cultures at day 14 of gestation and treating them for 7 days with a CD71 monoclonal antibody (mAb). The intracellular iron deficiency caused by this treatment, inhibits both proliferation and maturation of the thymocytes. Cell recovery was reduced by 60%, but cells still expanded tenfold during the culture. Remarkably, the final maturation of αβ T cells was completely blocked as no thymocytes with low or high CD3/αβTcR expression developed. Moreover, only few cells reached the CD4+8+3− stage of T cell development. CD4−8−3− thymocytes, however, as well as its CD44−25+ subset developed in normal numbers, suggesting that CD44−25+ CD4−8−3− cells, or their immediate progeny, were most vulnerable to CD71 mAb treatment. The development of γδ T cells, which also express CD71, was not affected in these cultures. This suggests that γδ T cells are either less iron-dependent or possess alternative iron-uptake mechanisms.     Dividing cells require iron and, therefore, express the transferrin receptor (CD71) on the cell surface to enable internalization of transferrin-bound iron. Since early T cell development is marked by intense proliferation, we questioned whether CD71 might serve as a marker of immature T cells. Therefore, we analyzed the expression of CD71 on fetal, neonatal, and adult thymocytes in correlation with cell size, cell cycle status, and expression of CD3, CD4, CD8, αβTcR, and γδTcR. Phenotypic analysis showed that only the large, immature CD4-8-3-, CD4-8+3-, and CD4+8+3- cells in fetal, neonatal, and adult thymus expressed CD71. In addition, DNA analysis showed that all CD71+ large adult thymocytes were cycling. Downregulation of CD71 occurs when proliferation ceases, i.e., within the CD4+8+3- thymocyte subpopulation. The gradual changes in size and CD71 expression suggest a sequential development within this CD4+8+3- subpopulation from large CD71+ via small CD71± to small CD71- cells. As a consequence, CD71 expression is downregulated, in adult T cell development as well as in ontogeny, before the αβTcR appears on the cell surface of the thymocyte.     Studies on digestive physiology and feed digestibilities in captive Indian rhinoceros (Rhinoceros unicornis)

    We performed intake, digestibility and ingesta passage studies in 11 Indian rhinoceroses (Rhinoceros unicornis) from four zoological institutions, using total faecal collection for the quantification of faecal output. The regularly fed zoo ration of roughage and concentrates (ration RC) and a roughage-only ration (ration R) were used; the roughage source differed between the facilities and comprised grass hay, grass silage, straw and lucerne hay. Dry matter intake ranged between 0.8 and 1.3% of body weight on ration RC and 0.5-1.2% on ration R. Digestibility coefficients achieved were similar to those reported for horses on diets of comparable composition. Endogenous losses as determined by linear regression analysis were within the range reported for horses. Measurements of faecal volatile fatty acids, faecal lactate and faecal pH also showed similarity to similar measurements in horses. The mean retention times of fluids (Co-EDTA) and particles (Cr-mordanted fibre < 2 mm) in the whole gastrointestinal tract averaged 42 and 61 h, respectively, and were the longest ever recorded in a monogastric ungulate with this marker system. The results suggest that the horse is a useful model animal for designing diets for Indian rhinoceroses. Why digestive parameters are similar between these species in spite of enormous differences in body weight and retention times remains to be answered

    Development and evaluation of a tropical feed library for the Cornell Net Carbohydrate and Rrotein System model Desenvolvimento e avaliação de uma biblioteca de alimentos tropicais para o modelo "Sistema de Carboidrato e Proteína Líquidos" da Universidade de Cornell

    The Cornell Net Carbohydrate and Protein System (CNCPS) model has been increasingly used in tropical regions for dairy and beef production. However, the lack of appropriate characterization of the feeds has restricted its application. The objective of this study was to develop and evaluate a feed library containing feeds commonly used in tropical regions with characteristics needed as inputs for the CNCPS. Feed composition data collected from laboratory databases and from experiments published in scientific journals were used to develop this tropical feed library. The total digestible nutrients (TDN) predicted at 1x intake of maintenance requirement with the CNCPS model agreed with those predicted by the Weiss et al. (1992) equation (r² of 92.7%, MSE of 13, and bias of 0.8%) over all feeds. However, the regression r² of the tabular TDN values and the TDN predicted by the CNCPS model or with the Weiss equation were much lower (58.1 and 67.5%, respectively). A thorough comparison between observed and predicted TDN was not possible because of insufficient data to characterize the feeds as required by our models. When we used the mean chemical composition values from the literature data, the TDN predicted by our models did not agree with the measured values. We conclude using the TDN values calculated using the Weiss equation and the CNCPS model that are based on the actual chemical composition of the feeds result in energy values that more accurately represent the feeds being used in specific production situations than do the tabular values. Few papers published in Latin America journals that were used in this study reported information need by models such as the CNCPS.<br>O uso do Sistema de Carboidrato e Proteina Líquidos da Universidade de Cornell (CNCPS) tanto para produção de leite como carne tem aumentado durante o últimos anos nas regiões tropicais. Entretanto, a falta de uma caracterização adequada de alimentos tem restringido o seu uso corretamente. Esse trabalho teve como objetivo principal o desenvolvimento e a avaliação de uma tabela de composição de alimentos utilizados nas condições tropicais. Os dados da composição desses alimentos foram baseados nas informações necessárias para o uso do modelo CNCPS desenvolvido pela Universidade de Cornell, USA. A composição desses alimentos foi obtida através de análises realizadas em laboratórios e de experimentos publicados em revistas científicas. Os nutrientes digestíveis totais (NDT) estimados através da composição de carboidratos e proteina dos alimentos pela equação de Weiss et al. (1992) e pelo modelo CNCPS foram comparados com os valores da tabela. O NDT estimado ao nível de mantença (1x) com o modelo CNCPS obteve valores próximos aos estimados pela equação de Weiss et al. (1992) (r² = 92.7% e bias = 0.8%). Entretanto, o r² da regressão entre os valores de NDT da tabela e o estimado pelo CNCPS e por Weiss foram menores (58.1 e 67.5%, respectivamente). Uma comparação completa entre os valores observados e preditos não foi possível devido a falta de caracterização dos alimentos conforme necessário pelos modelos testados. Quando os valores médios de literatura foram utilizados, a correlação entre o NDT estimado e o observado foi muito baixa. Concluímos que os valores de NDT estimados por Weiss e modelo CNCPS fornecem melhores estimativas de NDT do que os valores de tabela. A maioria dos trabalhos publicados que foram avaliados nesse estudo raramente incluíam informações necessárias para modelos como o CNCPS

    Rumen fermentation and nutrient flow to the omasum in Holstein cows fed extruded canola seeds treated with or without lignosulfonate

    Four multiparous Holstein cows averaging 548 kg of body weight and 74 d in lactation were used in a Latin square design with four 21-d experimental periods to determine effects of feeding extruded versus non-extruded canola seed, with or without 50 g/kg lignosulfonate on rumen fermentation, nutrient flow to the omasum, and degradability of dry matter (DM) and N of each diet. The DM effective degradability increased with extrusion and lignosulfonate treatment had no effect. The effective degradability of N was similar between diets. Lignosulfonate treatment of extruded versus non-extruded canola seeds decreased ruminal and total tract apparent digestibility of organic matter. The lowest apparent ruminal and highest intestinal digestibilities of protein, expressed as a percentage of N intake were observed for cows fed extruded canola seeds without lignosulfonate. Lignosulfonate treatment and extrusion had no effect on pH and concentrations of ammonia N and volatile fatty acids in the rumen. Results suggest that extruded canola seed untreated with formaldehyde may stimulate efficiency of microbial protein synthesis and is an effective means of increasing the availability of protein in the small intestine without affecting the total tract apparent digestibility of protein
