In situ analysis of pH gradients in mosquito larvae using non-invasive, self-referencing, pH-sensitive microelectrodes
The alkaline environment, pH approximately 11, in the anterior midgut lumen of mosquito larvae is essential for normal nutrition and development. The mechanism of alkalization is, however, unknown. Although evidence from immunohistochemistry, electron microscopy and electrophysiology suggests that a V-ATPase is present in the basal membranes of the epithelial cells, its physiological role in the alkalization process has not been demonstrated. To investigate a possible role of the V-ATPase in lumen alkalization, pH gradients emanating from the hemolymph side of the midgut in semi-intact mosquito larvae were measured using non-invasive, self-referencing, ion-selective microelectrodes (SERIS). Large H+ concentration gradients, with highest concentrations close to the basal membrane (outward [H+] gradients), were found in the anterior midgut, whereas much smaller gradients, with concentrations lowest close to this membrane (inward [H+] gradients), were found in the gastric caeca and posterior midgut. Similar region-specific pH gradients, with consistent anterior-to-posterior profiles, were observed in individuals of two Aedes species, Aedes aegypti from semi-tropical Florida and Aedes canadensis from north-temperate Massachusetts. The gradients remained in a steady state for up to 6 h, the maximum duration of the recordings. Bafilomycin A1 (10(-5), 10(-7 )mol x l(-1)) on the hemolymph side greatly diminished the [H+] gradients in the anterior midgut but had no effect on the gradients in the gastric caecum and posterior midgut. These physiological data are consistent with the previous findings noted above. Together, they support the hypothesis that a basal, electrogenic H+ V-ATPase energizes luminal alkalization in the anterior midgut of larval mosquitoes
Force-Extension Relations for Polymers with Sliding Links
Topological entanglements in polymers are mimicked by sliding rings
(slip-links) which enforce pair contacts between monomers. We study the
force-extension curve for linear polymers in which slip-links create additional
loops of variable size. For a single loop in a phantom chain, we obtain exact
expressions for the average end-to-end separation: The linear response to a
small force is related to the properties of the unstressed chain, while for a
large force the polymer backbone can be treated as a sequence of Pincus--de
Gennes blobs, the constraint effecting only a single blob. Generalizing this

Radiative charge transfer lifetime of the excited state of (NaCa)
New experiments were proposed recently to investigate the regime of cold
atomic and molecular ion-atom collision processes in a special hybrid
neutral-atom--ion trap under high vacuum conditions. The collisional cooling of
laser pre-cooled Ca ions by ultracold Na atoms is being studied. Modeling
this process requires knowledge of the radiative lifetime of the excited
singlet A state of the (NaCa) molecular system. We calculate
the rate coefficient for radiative charge transfer using a semiclassical
approach. The dipole radial matrix elements between the ground and the excited
states, and the potential curves were calculated using Complete Active Space
Self-Consistent field and M\"oller-Plesset second order perturbation theory
(CASSCF/MP2) with an extended Gaussian basis, 6-311+G(3df). The semiclassical
charge transfer rate coefficient was averaged over a thermal Maxwellian
distribution. In addition we also present elastic collision cross sections and
the spin-exchange cross section. The rate coefficient for charge transfer was
found to be cm/sec, while those for the elastic and
spin-exchange cross sections were found to be several orders of magnitude
higher ( cm/sec and cm/sec,
respectively). This confirms our assumption that the milli-Kelvin regime of
collisional cooling of calcium ions by sodium atoms is favorable with the
respect to low loss of calcium ions due to the charge transfer.Comment: 4 pages, 5 figures; v.2 - conceptual change
The Value of Information for Populations in Varying Environments
The notion of information pervades informal descriptions of biological
systems, but formal treatments face the problem of defining a quantitative
measure of information rooted in a concept of fitness, which is itself an
elusive notion. Here, we present a model of population dynamics where this
problem is amenable to a mathematical analysis. In the limit where any
information about future environmental variations is common to the members of
the population, our model is equivalent to known models of financial
investment. In this case, the population can be interpreted as a portfolio of
financial assets and previous analyses have shown that a key quantity of
Shannon's communication theory, the mutual information, sets a fundamental
limit on the value of information. We show that this bound can be violated when
accounting for features that are irrelevant in finance but inherent to
biological systems, such as the stochasticity present at the individual level.
This leads us to generalize the measures of uncertainty and information usually
encountered in information theory

This document is based on the deliberations and outputs of two ILCA in-house workshops held in 1992, and of numerous individual contributions by ILCA scientists. This work has similarities with the ILCA project on smallholder market-oriented dairying. Factors influencing development of market-oriented smallholder dairying in SSA, and factors affecting farm productivity in dairying are listed. The objectives and methods of the four phases, viz, typification of dairy system, characterization of a specific dairy system, seeking ways to enhance the development of specific dairy systems and cross-site synthesis are described
