    Segregation of a microsporidian parasite during host cell mitosis

    We investigated the segregation of an intracellular microsporidian parasite during host cell division. A time-course experiment was carried out to examine the distribution of parasites relative to host chromosomal DNA via light and electron microscopy. Fluorescent light microscopy and EM studies showed that the parasite lay in the perinuclear zone of the host cell during interphase and segregated to daughter cells at mitosis. At metaphase, the parasite was frequently closely associated with host microtubules and mitochondria. Electron-dense bridges were observed between the parasites and the host microtubules and also between host mitochondria and microtubules. The study suggests that both the parasite and the host cell organelles segregate in association with spindle microtubules

    Cellular distribution of a feminizing microsporidian parasite: a strategy for transovarial transmission

    The cellular distribution of a vertically transmitted, feminizing microsporidian was followed in its host Gammarus duebeni. In adult females the parasite was restricted to gonadal tissue, in particular primary and secondary follicle cells. Spores were diplokaryotic with a thin spore wall and a short polar filament, characteristics typical of ‘early’ spores involved in autoinfection. The diplokaryotic life-cycle, absence of spore groupings and of a pansporoblast membrane typify the genus Nosema. However, the unusual globular polaroplast of the spore and restriction of this stage to host ovarian tissue have not previously been described in Nosema. Sporogony occurred only in follicle cells adjacent to developing oocytes and was in synchrony with the process of vitellogenesis. Oocytes were infected after formation of intracellular connections with follicle cells but harboured only vegetative stages of the parasite. Parasites were associated with the perinuclear cytoplasm and, in developing embryos, segregated to daughter cells along the axis of the spindle. In juvenile animals there was no evidence of pathology linked with feminization and the parasite was found at low density in cells under the cuticle. The parasite is highly adapted to transovarial transmission with an efficient mechanism of oocyte infection and no evidence of pathology

    A spatial analysis of physiological changes associated with infection of cotyledons of marrow plants with cucumber mosaic virus

    Changes in host primary metabolism associated with the compatible interaction between cucumber mosaic virus and cotyledons of the marrow plant (Cucurbita pepo L.) have been localized, first by measuring activities of key enzymes in infected and uninfected regions of the cotyledon, and second by histochemical techniques applied to tissue prints of the infected region. A series of progressive metabolic changes occurs within the expanding infected lesion. Virus replication and the synthesis of viral protein at the periphery creates a strong sink demand associated with increased activities of anaplerotic enzymes, increased photosynthesis, and starch accumulation. Inside the lesion, when the synthesis of virus has declined, photosynthesis is reduced, starch is mobilized, and the emphasis of metabolism is shifted toward glycolysis and mitochondrial respiration. These changes are associated spatially with the onset of chlorosis. A decrease in total protein synthesis in this inner zone could be instrumental in some or all of these changes, leading to symptoms of viral infection

    Has Kelantan grown faster than other states in Malaysia? A panel data analysis

    Kelantan has been the poorest state in Malaysia for the past five decades. Despite the various Malaysian Development Plans for the past several decades, regional disparity between states remains in Malaysia. Thus, the objective of the present paper is to address the question whether Kelantan has been narrowing their income gap with other states in Malaysia. Using annual data for the period 1961 to 2003, our panel unit root test result suggest that (i) Kelantan converges towards Kedah, Negeri Sembilan, Perak, Pahang, Perlis and Selangor.; (ii) Kelantan is catching-up to Johor, Melaka, Penang, Sabah, Terengganu and Wilayah Persekutuan; and (iii) Kelantan show divergence with Sarawak. In this respect, the government has an important role to play in enhancing growth by continuously providing stable economic environment for investment and other productive economic activities.     Infection of Gammarus duebeni populations by two vertically transmitted microsporidia; parasite detection and discrimination by PCR–RFLP

    We screened a population of the brackish water crustacean Gammarus duebeni from the Isle of Cumbrae for the presence of vertically transmitted microsporidia. We compared 2 screening techniques; light microscopy and PCR-based detection using generic 16S rDNA microsporidian primers. Fifty percent of females from this population tested positive for vertically transmitted microsporidia. The PCR screen was 100% efficient in comparison with existing LM based screening. In addition, the PCR screen produced bands of 2 sizes suggesting that more than 1 species of microsporidian was present. Sequencing revealed 2 distinct species of vertically transmitted microsporidia; 33% of females were infected with the feminizer Nosema granulosis and 17% were infected with a new species which we provisionally designate Microsporidium sp. On the basis of sequence information, we developed a discriminatory PCR–RFLP test based on MspI and HaeIII digests. This screen allows rapid detection and discrimination of vertically transmitted microsporidia in natural field populations. We applied the PCR–RFLP screen to a second G. duebeni population from the Isle of Man. This population also hosted these 2 parasite species. In total 45% of females harboured N. granulosis and 10% harboured Microsporidium sp. No dual-infected individuals were found in either population. The occurrence of 2 vertically transmitted parasites within a population has implications for our understanding of parasite–host relationships in the field and we discuss factors affecting the dynamics of parasite–parasite competition and coexistence

    The Upper Critical Field in Disordered Two-Dimensional Superconductors

    We present calculations of the upper critical field in superconducting films as a function of increasing disorder (as measured by the normal state resistance per square). In contradiction to previous work, we find that there is no anomalous low-temperature positive curvature in the upper critical field as disorder is increased. We show that the previous prediction of this effect is due to an unjustified analytical approximation of sums occuring in the perturbative calculation. Our treatment includes both a careful analysis of first-order perturbation theory, and a non-perturbative resummation technique. No anomalous curvature is found in either case.     First results from the VIMOS-IFU survey of gravitationally lensing clusters at z~0.2

    We present the on-going observational program of a VIMOS Integral Field Unit survey of the central regions of massive, gravitational lensing galaxy clusters at redshift z~0.2. We have observed six clusters using the low-resolution blue grism (R about 200), and the spectroscopic survey is complemented by a wealth of photometric data, including Hubble Space Telescope optical data and near infrared VLT data. The principal scientific aims of this project are: the study of the high-z lensed galaxies, the transformation and evolution of galaxies in cluster cores and the use of multiple images to constrain cosmography.     Tidal influence on Rutford Ice Stream, West Antarctica: observations of surface flow and basal processes from closely-spaced GPS and passive seismic stations

    High-resolution surface velocity measurements and passive seismic observations from Rutford Ice Stream, West Antarctica, 40 km upstream from the grounding line are presented. These measurements indicate a complex relationship between the ocean tides and currents, basal conditions and ice-stream flow. Both the mean basal seismicity and the velocity of the ice stream are modulated by the tides. Seismic activity increases twice during each semi-diurnal tidal cycle. The tidal analysis shows the largest velocity variation is at the fortnightly period, with smaller variations superimposed at diurnal and semi-diurnal frequencies. The general pattern of the observed velocity is two velocity peaks during each semi-diurnal tidal cycle, but sometimes three peaks are observed. This pattern of two or three peaks is more regular during spring tides, when the largest-amplitude velocity variations are observed, than during neap tides. This is the first time that velocity and level of seismicity are shown to correlate and respond to tidal forcing as far as 40 km upstream from the grounding line of a large ice stream
