    Effects of chronic carbon monoxide exposure on fetal growth and development in mice

    <p>Abstract</p> <p>Background</p> <p>Carbon monoxide (CO) is produced endogenously, and can also be acquired from many exogenous sources: ie. cigarette smoking, automobile exhaust. Although toxic at high levels, low level production or exposure lends to normal physiologic functions: smooth muscle cell relaxation, control of vascular tone, platelet aggregation, anti- inflammatory and anti-apoptotic events. In pregnancy, it is unclear at what level maternal CO exposure becomes toxic to the fetus. In this study, we hypothesized that CO would be embryotoxic, and we sought to determine at what level of chronic CO exposure in pregnancy embryo/fetotoxic effects are observed.</p> <p>Methods</p> <p>Pregnant CD1 mice were exposed to continuous levels of CO (0 to 400 ppm) from conception to gestation day 17. The effect on fetal/placental growth and development, and fetal/maternal CO concentrations were determined.</p> <p>Results</p> <p>Maternal and fetal CO blood concentrations ranged from 1.12- 15.6 percent carboxyhemoglobin (%COHb) and 1.0- 28.6%COHb, respectively. No significant difference was observed in placental histological morphology or in placental mass with any CO exposure. At 400 ppm CO vs. control, decreased litter size and fetal mass (p < 0.05), increased fetal early/late gestational deaths (p < 0.05), and increased CO content in the placenta and the maternal spleen, heart, liver, kidney and lung (p < 0.05) were observed.</p> <p>Conclusions</p> <p>Exposure to levels at or below 300 ppm CO throughout pregnancy has little demonstrable effect on fetal growth and development in the mouse.</p

    Could Segue 1 be a destroyed star cluster? - a dynamical perspective

    We attempt to find a progenitor for the ultra-faint object Segue 1 under the assumption that it formed as a dark matter free star cluster in the past. We look for orbits, using the elongation of Segue 1 on the sky as a tracer of its path. Those orbits are followed backwards in time to find the starting points of our N-body simulations. The successful orbit, with which we can reproduce Segue 1 has a proper motion of mu_alpha = -0.19 mas/yr and mu_delta = -1.9 mas/yr, placing Segue 1 near its apo-galacticon today. Our best fitting model has an initial mass of 6224 Msun and an initial scale-length of 5.75 pc.Comment: 9 pages, 5 figures, 3 tables, accepted by MNRA

    The tip of the iceberg: a giant pelvic atypical lipoma presenting as a sciatic hernia

    BACKGROUND: This case report highlights two unusual surgical phenomena: lipoma-like well-differentiated liposarcomas and sciatic hernias. It illustrates the need to be aware that hernias may not always simply contain intra-abdominal viscera. CASE PRESENTATION: A 36 year old woman presented with an expanding, yet reducible, right gluteal mass, indicative of a sciatic hernia. However, magnetic resonance imaging demonstrated a large intra- and extra-pelvic fatty mass traversing the greater sciatic foramen. The tumour was surgically removed through an abdomino-perineal approach. Subsequent pathological examination revealed an atypical lipomatous tumour (synonym: lipoma-like well-differentiated liposarcoma). The patient remains free from recurrence two years following her surgery. CONCLUSION: The presence of a gluteal mass should always suggest the possibility of a sciatic hernia. However, in this case, the hernia consisted of an atypical lipoma spanning the greater sciatic foramen. Although lipoma-like well-differentiated liposarcomas have only a low potential for recurrence, the variable nature of fatty tumours demands that patients require regular clinical and radiological review

    BUDHIES II:A phase-space view of H I gas stripping and star formation quenching in cluster galaxies

    We investigate the effect of ram-pressure from the intracluster medium on the stripping of H I gas in galaxies in a massive, relaxed, X-ray bright, galaxy cluster at z = 0.2 from the Blind Ultra Deep H I Environmental Survey (BUDHIES). We use cosmological simulations, and velocity versus position phase-space diagrams to infer the orbital histories of the cluster galaxies. In particular, we embed a simple analytical description of ram-pressure stripping in the simulations to identify the regions in phase-space where galaxies are more likely to have been sufficiently stripped of their H I gas to fall below the detection limit of our survey. We find a striking agreement between the model predictions and the observed location of H I-detected and non-detected blue (late-type) galaxies in phase-space, strongly implying that ram-pressure plays a key role in the gas removal from galaxies, and that this can happen during their first infall into the cluster. However, we also find a significant number of gas-poor, red (early-type) galaxies in the infall region of the cluster that cannot easily be explained with our model of ram-pressure stripping alone. We discuss different possible additional mechanisms that could be at play, including the pre-processing of galaxies in their previous environment. Our results are strengthened by the distribution of galaxy colours (optical and UV) in phase-space, that suggests that after a (gas-rich) field galaxy falls into the cluster, it will lose its gas via ram-pressure stripping, and as it settles into the cluster, its star formation will decay until it is completely quenched. Finally, this work demonstrates the utility of phase-space diagrams to analyse the physical processes driving the evolution of cluster galaxies, in particular H I gas stripping

    HST Observations of Chromospheres in Metal Deficient Field Giants

    HST high resolution spectra of metal-deficient field giants more than double the stars in previous studies, span about 3 magnitudes on the red giant branch, and sample an abundance range [Fe/H]= -1 to -3. These stars, in spite of their age and low metallicity, possess chromospheric fluxes of Mg II (2800 Angstrom) that are within a factor of 4 of Population I stars, and give signs of a dependence on the metal abundance at the lowest metallicities. The Mg II k-line widths depend on luminosity and correlate with metallicity. Line profile asymmetries reveal outflows that occur at lower luminosities (M_V = -0.8) than detected in Ca K and H-alpha lines in metal-poor giants, suggesting mass outflow occurs over a larger span of the red giant branch than previously thought, and confirming that the Mg II lines are good wind diagnostics. These results do not support a magnetically dominated chromosphere, but appear more consistent with some sort of hydrodynamic, or acoustic heating of the outer atmospheres.Comment: 36 pages, 12 figures, 7 tables, and accepted for publication in The Astronomical Journa

    Targeting the DNA Double Strand Break Repair Machinery in Prostate Cancer

    Regardless of the achievable remissions with first line hormone therapy in patients with prostate cancer (CaP), the disease escapes the hormone dependent stage to a more aggressive status where chemotherapy is the only effective treatment and no treatment is curative. This makes it very important to identify new targets that can improve the outcome of treatment. ATM and DNA-PK are the two kinases responsible for signalling and repairing double strand breaks (DSB). Thus, both kinases are pertinent targets in CaP treatment to enhance the activity of the numerous DNA DSB inducing agents used in CaP treatment such as ionizing radiation (IR). Colony formation assay was used to assess the sensitivity of hormone dependent, p53 wt (LNCaP) and hormone independent p53 mutant (PC3) CaP cell lines to the cytotoxic effect of IR and Doxorubicin in the presence or absence of Ku55933 and NU7441 which are small molecule inhibitors of ATM and DNA-PK, respectively. Flow cytometry based methods were used to assess the effect of the two inhibitors on cell cycle, apoptosis and H2AX foci formation. Neutral comet assay was used to assess the induction of DNA DSBs. Ku55933 or NU7441 alone increased the sensitivity of CaP cell lines to the DNA damaging agents, however combining both inhibitors together resulted in further enhancement of sensitivity. The cell cycle profile of both cell lines was altered with increased cell death, DNA DSBs and H2AX foci formation. This study justifies further evaluation of the ATM and DNA-PK inhibitors for clinical application in CaP patients. Additionally, the augmented effect resulting from combining both inhibitors may have a significant implication for the treatment of CaP patients who have a defect in one of the two DSB repair pathways

Aquesta realitat no tan sols implica que visquem més anys sinó que, també, tinguem més possibilitats de patir tota mena de malalties pròpies de l’edat (com les demències, les degeneratives, etc.) i de patir-les més anys. Per edat, els que cuidaven ahir, són els cuidador d’avui: més grans, més envellits i que, a més, cuiden durant més anys. Un cuidador són dues vides. Així que, com la que s’està cuidant, la vida del cuidador també necessita ser cuidada ja que, sinó, la cura pot esdevenir una càrrega i un factor negatiu en el desenvolupament del mateix cuidador amb conseqüències tant físiques, com psíquiques i psicològiques. Aquesta investigació es centra en la realitat d’aquests cuidadors, en la seva qualitat de vida i en la seva experiència; tenint en compte també, els diversos punts de vista dels professionals que, des de institucions distintes, treballen i actuen en aquests casos.In recent decades, life expectancy has increased dramatically. This does not mean that only live longer but also more likely to suffer from all kinds of diseases typical of the age (such as dementia, degenerative, etc.) and years of suffering the most. For age, who took care of yesterday are the caregiver today: bigger, aging and also care for more years. A caregiver are two lives. So, as being caring, life caregiver also need to be careful because, but care can become a burden and a negative factor in the development of the same caregiver consequences both physical and mental and psychological. This research focuses on the reality of these carers in their quality of life and their experience; taking into account the various views of professionals from different institutions, work and act in such cases
