    Marker-Less Stage Drift Correction in Super-Resolution Microscopy Using the Single-Cluster PHD Filter

    Fluorescence microscopy is a technique which allows the imaging of cellular and intracellular dynamics through the activation of fluorescent molecules attached to them. It is a very important technique because it can be used to analyze the behavior of intracellular processes in vivo in contrast to methods like electron microscopy. There are several challenges related to the extraction of meaningful information from images acquired from optical microscopes due to the low contrast between objects and background and the fact that point-like objects are observed as blurred spots due to the diffraction limit of the optical system. Another consideration is that for the study of intracellular dynamics, multiple particles must be tracked at the same time, which is a challenging task due to problems such as the presence of false positives and missed detections in the acquired data. Additionally, the objective of the microscope is not completely static with respect to the cover slip due to mechanical vibrations or thermal expansions which introduces bias in the measurements. In this paper, a Bayesian approach is used to simultaneously track the locations of objects with different motion behaviors and the stage drift using image data obtained from fluorescence microscopy experiments. Namely, detections are extracted from the acquired frames using image processing techniques, and then these detections are used to accurately estimate the particle positions and simultaneously correct the drift introduced by the motion of the sample stage. A single cluster Probability Hypothesis Density (PHD) filter with object classification is used for the estimation of the multiple target state assuming different motion behaviors. The detection and tracking methods are tested and their performance is evaluated on both simulated and real data

    A Scheme for the Detection and Tracking of People Tuned for Aerial Image Sequences

    Abstract. This paper addresses the problem of detecting and tracking a large number of individuals in aerial image sequences that have been taken from high altitude. We propose a method which can handle the numerous challenges that are associated with this task and demonstrate its quality on several test sequences. Moreover this paper contains several contributions to improve object detection and tracking in other domains, too. We show how to build an effective object detector in a flexible way which incorporates the shadow of an object and enhanced features for shape and color. Furthermore the performance of the detector is boosted by an improved way to collect background samples for the classifier train-ing. At last we describe a tracking-by-detection method that can handle frequent misses and a very large number of similar objects

    Absence of differential predation on rats by Malaysian Barn Owls in oil palm plantations

    Barn Owls (Tyto alba javanica) have been widely introduced in Malaysian oil palm plantations to control rodent pests. However, their effectiveness in regulating rodent populations is unknown. We investigated whether Barn Owls selected prey with respect to size and sex classes based on data from 128 pellets of Barn Owls compared to 1292 live-trapped rats in an oil palm plantation in Malaysia. The birds mostly fed on Rattus rail as diardii, the most commonly trapped species. Body mass of prey consumed was predicted based on models derived from measurements from trapped rats. Sex of prey was determined by pelvic measurements with reference to those taken from specimens of known gender. There was no clear selection of prey by Barn Owls in relation to size or sex of prey, and no difference in the body mass of prey between the owls' breeding and nonbreeding seasons. The absence of differential predation in Barn Owls may partly explain the lack of dear evidence that they regulate rodent populations and thus act as successful biological control agents

    BRCA2 diffuses as oligomeric clusters with RAD51 and changes mobility after DNA damage in live cells

    Genome maintenance by homologous recombination depends on coordinating many proteins in time and space to assemble at DNA break sites. To understand this process, we followed the mobility of BRCA2, a critical recombination mediator, in live cells at the single-molecule level using both single-particle tracking and fluorescence correlation spectroscopy. BRCA2-GFP and -YFP were compared to distinguish diffusion from fluorophore behavior. Diffusive behavior of fluorescent RAD51 and RAD54 was determined for comparison. All fluorescent proteins were expressed from endogenous loci. We found that nuclear BRCA2 existed in oligomeric clusters, and exhibited heterogeneous mobility. DNA damage increased BRCA2 transient binding, presumably including binding to damaged sites. Despite its very different size, RAD51 displayed mobility similar to BRCA2, which indicates physical interaction between these proteins both before and after induction of DNA damage. We propose that BRCA2-mediated sequestration of nuclear RAD51 serves to prevent inappropriate DNA interactions and that all RAD51 is delivered to DNA damage sites in association with BRCA2

    Concerted action of kinesins kif5b and kif13b promotes efficient secretory vesicle transport to microtubule plus ends

    Intracellular transport relies on multiple kinesins, but it is poorly understood which kinesins are present on particular cargos, what their contributions are and whether they act simultaneously on the same cargo. Here, we show that Rab6-positive secretory vesicles are transported from the Golgi apparatus to the cell periphery by kinesin-1 KIF5B and kinesin-3 KIF13B, which determine the location of secretion events. KIF5B plays a dominant role, whereas KIF13B helps Rab6 vesicles to reach freshly polymerized microtubule ends, to which KIF5B binds poorly, likely because its cofactors, MAP7-family proteins, are slow in populating these ends. Sub-pixel localization demonstrated that during microtubule plus-end directed transport, both kinesins localize to the vesicle front and can be engaged on the same vesicle. When vesicles reverse direction, KIF13B relocates to the middle of the vesicle, while KIF5B shifts to the back, suggesting that KIF5B but not KIF13B undergoes a tug-of-war with a minus-end directed motor.info:eu-repo/semantics/publishe
