    Dynamic variation of supernatant quality in a dairy shed waste stabilisation pond system

    An intensive monitoring program of a standard two-stage dairy shed waste stabilisation pond system was undertaken to determine the incidence and extent of spatial and temporal variation of basic physio-chemical parameters, and to shed light on the longer term dynamic nature of in-pond conditions. The anaerobicâfacultative pond system, located in a remote rural area, treats wastewater from the hosing down and hydraulic flushing of the milking parlour and holding yard at the farm dairy shed. A number of multi-parameter water quality field monitoring probes were permanently deployed at various locations within the two ponds to enable continuous measurement of temperature, pH, conductivity and dissolved oxygen. In addition, profiling of the supernatant of both ponds was undertaken at different times of the year to examine vertical variation of the same parameters. Continuous monitoring revealed spatial homogeneity in EC and pH levels in the upper metre of both ponds. Physio-chemical parameters also appear to change uniformly across the ponds in response to external stimuli such as rainfall. Neither pond, however, exhibits homogeneity down the profile of the supernatant. Seasonal stratification is prevalent in the facultative pond suggesting poor vertical mixing, while the anaerobic pond is notably affected by sludge accumulation. A long-term pattern of rising conductivity in both ponds indicated accumulation of dissolved salt species in the system due to recirculation of reclaimed effluent for hydraulic flushing of the dairy shed. In the facultative pond, diurnal fluctuations in dissolved oxygen, oxidation-reduction potential and turbidity during warmer months of the year closely followed temperature swings.     Physical and hydrodynamic characteristics of a dairy shed waste stabilisation pond system

    Waste stabilization pond systems are widely used to treat animal wastes under highly variable hydraulic loading regimes. These systems have received limited research attention with regard to their hydrodynamic behaviour and the potential impact of shock hydraulic loading on their performance. In this study a two-stage dairy shed waste stabilisation pond system was topographically surveyed to determine the physical shape and the theoretical hydraulic retention time (HRT) of each pond, as well as the extent of sludge accumulation in the primary pond. The primary pond was then subjected to a series of drogue tracking runs whereby weighted floating survey targets with submerged âsailsâ were tracked during their movement through the pond at times of peak flow in order to characterise the hydrodynamic behaviour of the pond. The full capacity volumes of the primary and secondary ponds were calculated to be 1285m3 and 2391m3, respectively. Sludge had been accumulating in the primary pond at a rate of 0.73m3/d over a period of 2.4 years and this has reduced the active treatment volume of the pond to 657m3. Based on mean outflow, the HRTs of the ponds were 40 d and 137 d, respectively. The drogue runs revealed a vortexlike mixing pattern within the pond with higher velocities around the perimeter of the pond between the inlet and outlet, and lower velocities in the centre of the pond. In-pond velocities seemed relatively high in comparison with those from other drogue studies of larger ponds and the surging inflow caused the formation of a flow âjetâ that potentially contributed to significant short-circuiting. The range of influence of this flow jet, however, was limited to within 15m of the inlet, suggesting that short-circuiting would be likely to occur only under certain high inflow conditions

    Antidiabetic potential of Moringa oleifera Lam. leaf extract in type 2 diabetic rats, and its mechanism of action

    Purpose: To explore the antidiabetic potential of Moringa oleifera leaf extract in type 2 diabetic rats, and the underlying mechanisms.Methods: Streptozotocin (STZ) at a dose of 40 mg/kg was given to high fat diet (HFD)- fed rats to induce type 2 diabetes. M. oleifera leaf extract at doses 100, 200 and 400 mg/kg were given to 3 groups of type 2 diabetic rats. The area under curve (AUC) of glucose and homeostasis model assessment of insulin resistance (HOMA-R) were calculated using appropriate formulas, whereas levels of glucose,insulin, peroxisome proliferator activated receptor-γ (PPARγ, dipeptidyl peptidase-IV (DPP-IV) and inflammatory cytokines (IL-6, IL-1β and TNFα) were assayed using ELISA kits.Results: The leaf extract of M. oleifera significantly reduced the levels of glucose, insulin and cytokines in treated type 2 diabetic groups (p < 0.05). DC group had significantly increased AUC for glucose, whereas the extract-treated groups showed significant  decrease in glucose AUC. There was significant decrease in insulin sensitivity parameters, as indicated by increase in HOMA-R and decrease in PPARγ levels in the DC group (p < 0.05). However, treatment with the M. oleifera extract reversed this trend via marked decrease in HOMA-R level and significant rise in PPARγ level. In contrast, the extract had no effect on DPP-IV concentration in diabetic treated groups (p < 0.05).Conclusion: These results indicate that M. oleifera leaf extract mitigates hyperglycemia in type 2 DM by modulating hyperinsulinemia, PPARγ and inflammatory cytokines. Thus, the extract is a potential source of drug for the management of type 2 DM.     Effect of Garlic (Allium Sativum) Supplementation on the Growth Performance of Crossbred Calves

    . Trial was conducted to assess the effects of different garlic supplementation methods on the performance of crossbred calves. Jersey crossbred calves of both sexes were randomly separated into three treatments (T1, T2 and T3) of six calves in each group. Calves in T1 group were supplemented with garlic powder at the dose rate of 250 mg/kg BW in water whereas calves in the T2 group were supplemented with 250 mg/kg BW in concentrate feed and the T3 group was treated as control. The data on growth rate in terms of body weight, average daily gain, feed intake, faecal consistency score, faecal egg count and disease incidence were analysed as per standard procedures. It was found that the calves in T1 and T2 group gained significantly higher overall body weight and average daily gain compared with calves in T3 group. No significant difference was observed in feed intake and feed conversion efficiency between the treatment groups. The faecal consistency score and faecal egg count remained the same in all the treatment groups

    A self-report comorbidity questionnaire for haemodialysis patients

Obtaining comorbidity data from medical records is cumbersome. A self-report comorbidity questionnaire is a useful alternative. Our aim in this study was to examine the predictive value of a self-report comorbidity questionnaire in terms of survival in ESRD patients. Methods. We studied a prospective cross-sectional cohort of 282 haemodialysis (HD) patients in a single centre. Participants were administered the self-report questionnaire during an HD session. Information on their comorbidities was subsequently obtained from an examination of the patient's medical records. Levels of agreement between parameters derived from the questionnaire, and from the medical records, were examined. Participants were followed-up for 18 months to collect survival data. The influence on survival of comorbidity scores derived from the self-report data (the Composite Self-report Comorbidity Score [CSCS]) and from medical records data - the Charlson Comorbidity Index [CCI] were compared. Results: The level of agreement between the self-report items and those obtained from medical records was almost perfect with respect the presence of diabetes (Kappa score κ 0.97), substantial for heart disease and cancer (κ 0.62 and κ 0.72 respectively), moderate for liver disease (κ 0.51), only fair for lung disease, arthritis, cerebrovascular disease, and depression (κ 0.34, 0.35, 0.34 and 0.29 respectively). The CSCS was strongly predictive of survival in regression models (Nagelkerke R2value 0.202), with a predictive power similar to that of the CCI (Nagelkerke R2value 0.211). The influences of these two parameters were additive in the models - suggesting that these parameters make different contributions to the assessment of comorbidity. Conclusion: This self-report comorbidity questionnaire is a viable tool to collect comorbidity data and may have a role in the prediction of short-term survival in patients with end-stage renal disease on haemodialysis.     Mosaic RAS/MAPK variants cause sporadic vascular malformations which respond to targeted therapy.

    BACKGROUND: Sporadic vascular malformations (VMs) are complex congenital anomalies of blood vessels that lead to stroke, life-threatening bleeds, disfigurement, overgrowth, and/or pain. Therapeutic options are severely limited, and multidisciplinary management remains challenging, particularly for high-flow arteriovenous malformations (AVM). METHODS: To investigate the pathogenesis of sporadic intracranial and extracranial VMs in 160 children in which known genetic causes had been excluded, we sequenced DNA from affected tissue and optimized analysis for detection of low mutant allele frequency. RESULTS: We discovered multiple mosaic-activating variants in 4 genes of the RAS/MAPK pathway, KRAS, NRAS, BRAF, and MAP2K1, a pathway commonly activated in cancer and responsible for the germline RAS-opathies. These variants were more frequent in high-flow than low-flow VMs. In vitro characterization and 2 transgenic zebrafish AVM models that recapitulated the human phenotype validated the pathogenesis of the mutant alleles. Importantly, treatment of AVM-BRAF mutant zebrafish with the BRAF inhibitor vemurafinib restored blood flow in AVM. CONCLUSION: Our findings uncover a major cause of sporadic VMs of different clinical types and thereby offer the potential of personalized medical treatment by repurposing existing licensed cancer therapies.     COVID-19 prevalence and mortality in patients with cancer and the effect of primary tumour subtype and patient demographics: a prospective cohort study

    BACKGROUND: Patients with cancer are purported to have poor COVID-19 outcomes. However, cancer is a heterogeneous group of diseases, encompassing a spectrum of tumour subtypes. The aim of this study was to investigate COVID-19 risk according to tumour subtype and patient demographics in patients with cancer in the UK. METHODS: We compared adult patients with cancer enrolled in the UK Coronavirus Cancer Monitoring Project (UKCCMP) cohort between March 18 and May 8, 2020, with a parallel non-COVID-19 UK cancer control population from the UK Office for National Statistics (2017 data). The primary outcome of the study was the effect of primary tumour subtype, age, and sex and on severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) prevalence and the case-fatality rate during hospital admission. We analysed the effect of tumour subtype and patient demographics (age and sex) on prevalence and mortality from COVID-19 using univariable and multivariable models. FINDINGS: 319 (30·6%) of 1044 patients in the UKCCMP cohort died, 295 (92·5%) of whom had a cause of death recorded as due to COVID-19. The all-cause case-fatality rate in patients with cancer after SARS-CoV-2 infection was significantly associated with increasing age, rising from 0·10 in patients aged 40-49 years to 0·48 in those aged 80 years and older. Patients with haematological malignancies (leukaemia, lymphoma, and myeloma) had a more severe COVID-19 trajectory compared with patients with solid organ tumours (odds ratio [OR] 1·57, 95% CI 1·15-2·15; p<0·0043). Compared with the rest of the UKCCMP cohort, patients with leukaemia showed a significantly increased case-fatality rate (2·25, 1·13-4·57; p=0·023). After correction for age and sex, patients with haematological malignancies who had recent chemotherapy had an increased risk of death during COVID-19-associated hospital admission (OR 2·09, 95% CI 1·09-4·08; p=0·028). INTERPRETATION: Patients with cancer with different tumour types have differing susceptibility to SARS-CoV-2 infection and COVID-19 phenotypes. We generated individualised risk tables for patients with cancer, considering age, sex, and tumour subtype. Our results could be useful to assist physicians in informed risk-benefit discussions to explain COVID-19 risk and enable an evidenced-based approach to national social isolation policies.     Development, characterization, and stability of O/W pepper nanoemulsions produced by high-pressure homogenization

    Interest in the utilization of bioactive plant compounds in foods has increased due to their biochemical activities (antioxidant, antimicrobial, etc.), and as alternatives in the reduction of the use of high concentrations of chemical substances. However, some of these additives are hydrophobic, thus being harder to disperse into the food matrix, which is generally water-based. A good alternative is the use of low concentrations of these compounds as nanoemulsions. The objective of the present study was to develop oil-in-water nanoemulsions containing dedo-de-moça pepper extract for food applications. Research in the development of these nanoemulsions was carried out using a high-speed homogenizer, followed by a high-pressure homogenizer. The influence of the following parameters was assessed: type and concentration of surfactants, hidrophilic-lipophilic balance, lipid/aqueous phase ratio, surfactant/oil ratio, pepper extract composition in nanoemulsion, and processing conditions. Nanoemulsions were evaluated by environmental (centrifugal and thermal) and storage stabilities, characterized by average droplet size and -potential measurements, color, interfacial tension, atomic force, and cryo-scanning electron microscopy. Those with average droplet size between 132 ± 2.0 and 145 ± 1.0 nm were developed depending on working pressure and number of cycles; -potential was around 36.71 ± 0.62 mV and the best nanoemulsion was stable to centrifugation and most of the thermal stresses. Droplets were characterized with cryo-scanning electron microscopy as being spherical, homogeneous, and stable, and remained stable when stored at 4 °C and room temperature for over 120 days.     Treatment of synthetic textile wastewater containing dye mixtures with microcosms

    The aim was to assess the ability of microcosms (laboratory-scale shallow ponds) as a post polishing stage for the remediation of artificial textile wastewater comprising two commercial dyes (basic red 46 (BR46) and reactive blue 198 (RB198)) as a mixture. The objectives were to evaluate the impact of Lemna minor L. (common duckweed) on the water quality outflows; the elimination of dye mixtures, organic matter, and nutrients; and the impact of synthetic textile wastewater comprising dye mixtures on the L. minor plant growth. Three mixtures were prepared providing a total dye concentration of 10 mg/l. Findings showed that the planted simulated ponds possess a significant (p &lt; 0.05) potential for improving the outflow characteristics and eliminate dyes, ammonium-nitrogen (NH4-N), and nitrate-nitrogen (NO3-N) in all mixtures compared with the corresponding unplanted ponds. The removal of mixed dyes in planted ponds was mainly due to phyto-transformation and adsorption of BR46 with complete aromatic amine mineralisation. For ponds containing 2 mg/l of RB198 and 8 mg/l of BR46, removals were around 53%, which was significantly higher than those for other mixtures: 5 mg/l of RB198 and 5 mg/l of BR46 and 8 mg/l of RB198 and 2 mg/l of BR46 achieved only 41 and 26% removals, respectively.
