    The interaction of large scale and mesoscale environment leading to formation of intense thunderstorms over Kolkata part I: Doppler radar and satellite observations

    The weather systems that predominantly affect the eastern and northeastern parts of India during the pre-monsoon summer months (March, April and May) are severe thunderstorms, known as Nor'westers. The storms derive their names from the fact that they frequently strike cities and towns in the southern part of West Bengal in the afternoon from the north-west direction while traveling far from its place of genesis over the Bihar plateau. The storms are devastating in nature particularly due to strong (gusty) winds, heavy rains and hails associated with it. Although these storms are well known for its power of causing damages, studies on them are relatively few due to their small size and sparse network of observations. To address this important issue, the evolution of two Nor'westers of 12 March and 22 May 2003 over Kolkata is studied in detail in this paper using hourly Doppler weather radar (DWR) observations and high resolution Meteosat-5 imageries. In addition, supporting meteorological reports are used to find the large scale conditions that influence the moisture convergence and vertical wind shear. The genesis of both the storms is found to be over Bihar-Jharkhand region and beyond the range of the DWR. The satellite observations are found to be useful in identifying the location and initiation of the storms. The movements of the storms are captured by the DWR estimated vertical cross-section of reflectivities. The Doppler estimate shows that the 12 March storm had a vertical extent of about 10-12 km at the time of maturity and that of 22 May reaching up to 18 km signifying deep convection associated with these events. The genesis, maturity and dissipation are well brought out by the hourly DWR and satellite imageries. The DWR observations suggest that the systems move at a speed of 20-25m/s. The DWR estimated precipitation shows a detailed spatial distribution around Kolkata with several localized zones of heavy rain and this is found to be well supported by the nearby station observations. This study establishes that DWR observations along with hourly satellite imageries are able to capture the evolution of Nor'westers. The study also shows that the composite DWR-satellite information is a reliable tool for nowcasting the location, time and path of movement of Nor'westers. Based on these observations, a conceptual model of the Nor'wester is proposed. © Printed in India

    Bio-inspired Tensegrity Soft Modular Robots

    In this paper, we introduce a design principle to develop novel soft modular robots based on tensegrity structures and inspired by the cytoskeleton of living cells. We describe a novel strategy to realize tensegrity structures using planar manufacturing techniques, such as 3D printing. We use this strategy to develop icosahedron tensegrity structures with programmable variable stiffness that can deform in a three-dimensional space. We also describe a tendon-driven contraction mechanism to actively control the deformation of the tensegrity mod-ules.     Human M1 macrophages express unique innate immune response genes after mycobacterial infection to defend against tuberculosis

    Mycobacterium tuberculosis (Mtb) is responsible for approximately 1.5 million deaths each year. Though 10% of patients develop tuberculosis (TB) after infection, 90% of these infections are latent. Further, mice are nearly uniformly susceptible to Mtb but their M1-polarized macrophages (M1-MΦs) can inhibit Mtb in vitro, suggesting that M1-MΦs may be able to regulate anti-TB immunity. We sought to determine whether human MΦ heterogeneity contributes to TB immunity. Here we show that IFN-γ-programmed M1-MΦs degrade Mtb through increased expression of innate immunity regulatory genes (Inregs). In contrast, IL-4-programmed M2-polarized MΦs (M2-MΦs) are permissive for Mtb proliferation and exhibit reduced Inregs expression. M1-MΦs and M2-MΦs express pro- and anti-inflammatory cytokine-chemokines, respectively, and M1-MΦs show nitric oxide and autophagy-dependent degradation of Mtb, leading to increased antigen presentation to T cells through an ATG-RAB7-cathepsin pathway. Despite Mtb infection, M1-MΦs show increased histone acetylation at the ATG5 promoter and pro-autophagy phenotypes, while increased histone deacetylases lead to decreased autophagy in M2-MΦs. Finally, Mtb-infected neonatal macaques express human Inregs in their lymph nodes and macrophages, suggesting that M1 and M2 phenotypes can mediate immunity to TB in both humans and macaques. We conclude that human MФ subsets show unique patterns of gene expression that enable differential control of TB after infection. These genes could serve as targets for diagnosis and immunotherapy of TB

    Empirical Constraints on Convective Core Overshoot

    In stellar evolution calculations, the local pressure scale height is often used to empirically constrain the amount of convective core overshoot. However, this method brings unsatisfactory results for low-mass stars (< 1.1 -1.2 Mo for Z= \Zs) which have very small cores or no convective core at all. Following Roxburgh's integral constraint,we implemented an upper limit of overshoot within the conventional method of alpha parameterization in order to remove an overly large overshoot effect on low-mass stars. The erroneously large effect of core overshoot due to the failure of alpha parameterization can be effectively corrected by limiting the amount of overshoot to < 15 % of the core radius. 15 % of the core radius would be a proper limit of overshoot which can be implemented in a stellar evolution code for intermediate to low mass stars. The temperature structure of the overshoot region does not play a crucial role in stellar evolution since this transition region is very thin.Comment: 15 pages, 5 figures, accepted for A

    Primary Sebaceous Carcinoma of the Corneaoscleral Limbus With Pagetoid Recurrence

    We report a sebaceous carcinoma confined to the corneoscleral limbus without involvement of the eyelid. A 60-year-old man, who showed multiple masses on the corneaoscleral limbus and limbal ulceration but with normal eyelids, underwent surgical en-bloc excision of the masses. Histopathologic examination revealed a sebaceous carcinoma. Three weeks after excision, multiple pagetoid recurrences were found along the bulbar conjunctiva 2 mm away from the limbus. After the application of topical mitomycin C, the pagetoid spread regressed completely. After a 2 year follow-up, no other local or systemic recurrences were observed. This report shows that the ulcerative mass which is confined to only the corneoscleral limbus may be a sebaceous carcinoma even without eyelid involvement. Topical mitomycin C may be effective for treating pagetoid spread of sebaceous carcinoma of limbal origin

    A Useful Predictor of Early Atherosclerosis in Obese Children: Serum High-sensitivity C-reactive Protein

    Childhood obesity seems to contribute to the development of vascular inflammation and the progression of arterial wall changes. High-sensitivity C-reactive protein (hs-CRP) has recently emerged as a useful biomarker for vascular inflammation associated with atherosclerosis. The objectives of this study were to evaluate the association of the serum hs-CRP level with ultrasonic findings of early atherosclerosis, carotid intima-media wall thickness (IMT) and brachial flow-mediated dilation (FMD), in obese children. Thirty eight obese children and 45 sex/age-matched healthy control children were recruited. Serum CRP levels were measured by the high-sensitive latex turbidimetric immunoassay, and we measured carotid IMT and brachial FMD using high-resolution B-mode ultrasonography. Obese children had significantly higher hs-CRP levels (1.40±0.74 mg/L vs. 0.55±0.49 mg/L, p<0.01), as well as increased IMT (0.52±0.09 mm vs. 0.41±0.07 mm, p<0.01) and impaired FMD (7.35±7.78% vs. 20.34±16.81%, p<0.01) compared to healthy controls. Serum hs-CRP correlated positively with IMT (r=0.413, p<0.05) and inversely with FMD (r=-0.350, p<0.05) in the obesity group. Measurement of the serum hs-CRP level is a simple, cheap, and highly reproducible assay and correlates with IMT and FMD in obese children. Thus, it would be a useful marker for evaluating and estimating the degree of atherosclerosis in children

    FOXD1-ALDH1A3 signaling is a determinant for the self-renewal and tumorigenicity of mesenchymal glioma stem cells

    Glioma stem-like cells (GSC) with tumor-initiating activity orchestrate the cellular hierarchy in glioblastoma and engender therapeutic resistance. Recent work has divided GSC into two subtypes with a mesenchymal (MES) GSC population as the more malignant subtype. In this study, we identify the FOXD1-ALDH1A3 signaling axis as a determinant of the MES GSC phenotype. The transcription factor FOXD1 is expressed predominantly in patient-derived cultures enriched with MES, but not with the proneural GSC subtype. shRNA-mediated attenuation of FOXD1 in MES GSC ablates their clonogenicity in vitro and in vivo. Mechanistically, FOXD1 regulates the transcriptional activity of ALDH1A3, an established functional marker for MES GSC. Indeed, the functional roles of FOXD1 and ALDH1A3 are likely evolutionally conserved, insofar as RNAi-mediated attenuation of their orthologous genes in Drosophila blocks formation of brain tumors engineered in that species. In clinical specimens of high-grade glioma, the levels of expression of both FOXD1 and ALDH1A3 are inversely correlated with patient prognosis. Finally, a novel small-molecule inhibitor of ALDH we developed, termed GA11, displays potent in vivo efficacy when administered systemically in a murine GSC-derived xenograft model of glioblastoma. Collectively, our findings define a FOXD1-ALDH1A3 pathway in controling the clonogenic and tumorigenic potential of MES GSC in glioblastoma tumors

    Reproductive Factors and Serum Uric Acid Levels in Females from the General Population: The KORA F4 Study

    Hyperuricemia is associated with an increased risk of metabolic and cardiovascular diseases. There are pronounced sex differences in the levels of uric acid. It is largely unknown whether or not reproductive parameters which induce hormonal changes are responsible for this. We examined if there are associations between reproductive parameters and uric acid levels in a female population-based sample. In this cross-sectional analysis, data of 1530 women aged 32 to 81 years participating in the KORA F4 study, conducted between 2006 and 2008 in Southern Germany were used. Reproductive parameters were obtained by standardized interviews. Uric acid levels were tested by the uricase method. The whole study sample and stratified in pre- and postmenopausal women was analyzed. Menopausal status and earlier age at menarche were associated with higher serum uric acid levels (age-adjusted: p-values 0.003, <0.001 respectively; after multivariable adjustment, including BMI: p-values 0.002, 0.036). A history of oral contraceptive use showed an association with uric acid levels only after multivariable adjustment (p-value 0.009). Hot flushes showed an association with uric acid levels only after age-adjustment (p-value 0.038), but lost significance after adding other confounders. Other reproductive factors, including parity, current or ever use of hormone replacement therapy, current use of oral contraceptives, hysterectomy, bilateral oophorectomy, or depressive mood related to menopausal transition were not associated with uric acid levels. Postmenopausal status, earlier age at menarche and a history of oral contraceptive use were independently associated with higher serum uric acid concentrations in women from the general population. Further studies, especially longitudinal population-based studies investigating the relationship of female reproductive parameters with uric acid levels are necessary to confirm our findings
