    A growing share of the United States population uses e-cigarettes but the optimal regulation of these controversial products remains an open question. We conduct a discrete choice experiment to investigate how adult tobacco cigarette smokers' demand for e-cigarettes and tobacco cigarettes varies by four attributes: (i) whether e-cigarettes are considered healthier than tobacco cigarettes, (ii) the effectiveness of e-cigarettes as a cessation device, (iii) bans on use in public places, and (iv) price. We find that adult smokers' demand for e-cigarettes is motivated more by health concerns than by the desire to avoid smoking bans or higher prices

    Validation of lethality processes for products with slow come up time: Bacon and bone-in ham

    Pork bellies and boneless hams were smoked or cooked using unusually long processes to determine the impact of extended come-up times on the populations of Clostridium perfringens, Salmonella enterica, Staphylococcus aureus and Listeria monocytogenes. The products were formulated using brine formulations representative of what might be used in commercial production, and the thermal processes were more than doubled in length. Pork bellies and boneless hams were inoculated on the surface as well as 1 cm below the surface, and samples were collected every 3 h. The populations of C. perfringens (spores and vegetative cells) at internal locations of pork bellies increased by less than 1 log10 and declined significantly (approximately 3 log10/cm2) on the surface of the bellies during an extended bacon process. The populations of S. enterica, L. monocytogenes and S. aureusdid not increase during the extended bacon process. The populations of C. perfringens (spores and vegetative cells), S. aureus, S. enterica and L. monocytogenesdeclined significantly over an extended ham process. There were significant population reductions (\u3e2 log10/cm2) at 7 h (surface) and 12 h (\u3e5 log10/g; internal) for the hams. Populations of both surface and internal locations of the hams declined to a point approaching the limit of detection of the assays within 17 h

    Template-free 13-protofilament microtubule–MAP assembly visualized at 8 A resolution

    Microtubule-associated proteins (MAPs) are essential for regulating and organizing cellular microtubules (MTs). However, our mechanistic understanding of MAP function is limited by a lack of detailed structural information. Using cryo-electron microscopy and single particle algorithms, we solved the 8 Å structure of doublecortin (DCX)-stabilized MTs. Because of DCX’s unusual ability to specifically nucleate and stabilize 13-protofilament MTs, our reconstruction provides unprecedented insight into the structure of MTs with an in vivo architecture, and in the absence of a stabilizing drug. DCX specifically recognizes the corner of four tubulin dimers, a binding mode ideally suited to stabilizing both lateral and longitudinal lattice contacts. A striking consequence of this is that DCX does not bind the MT seam. DCX binding on the MT surface indirectly stabilizes conserved tubulin–tubulin lateral contacts in the MT lumen, operating independently of the nucleotide bound to tubulin. DCX’s exquisite binding selectivity uncovers important insights into regulation of cellular MTs

    Ring-like Oligomers of Synaptotagmins and Related C2 Domain Proteins

    We recently reported that the C2AB portion of Synaptotagmin 1 (Syt1) could self-assemble into Ca2+-sensitive ring-like oligomers on membranes, which could potentially regulate neurotransmitter release. Here we report that analogous ring-like oligomers assemble from the C2AB domains of other Syt isoforms (Syt2, Syt7, Syt9) as well as related C2 domain containing protein, Doc2B and extended Synaptotagmins (E-Syts). Evidently, circular oligomerization is a general and conserved structural aspect of many C2 domain proteins, including Synaptotagmins. Further, using electron microscopy combined with targeted mutations, we show that under physiologically relevant conditions, both the Syt1 ring assembly and its rapid disruption by Ca2+ involve the well-established functional surfaces on the C2B domain that are important for synaptic transmission. Our data suggests that ring formation may be triggered at an early step in synaptic vesicle docking and positions Syt1 to synchronize neurotransmitter release to Ca2+ influx

    Long-term no-till and stover retention each decrease the global warming potential of irrigated continuous corn

    Over the last 50 years, the most increase in cultivated land area globally has been due to a doubling of irrigated land. Long-term agronomic management impacts on soil organic carbon (SOC) stocks, soil greenhouse gas (GHG) emissions, and global warming potential (GWP) in irrigated systems, however, remain relatively unknown. Here, residue and tillage management effects were quantified by measuring soil nitrous oxide (N2O) and methane (CH4) fluxes and SOC changes (ΔSOC) at a long-term, irrigated continuous corn (Zea mays L.) system in eastern Nebraska, USA. Management treatments began in 2002, and measured treatments included no or high stover removal (0 or 6.8 Mg DM ha-1 yr-1, respectively) under no-till (NT) or conventional disk tillage (CT) with full irrigation (n = 4). Soil N2O and CH4 fluxes were measured for five crop-years (2011 to 2015), and ΔSOC was determined on an equivalent-mass basis to ~30 cm soil depth. Both area- and yield-scaled soil N2O emissions were greater with stover retention compared to removal and for CT compared to NT, with no interaction between stover and tillage practices.     Pulmonary stretch receptor activity during partial liquid ventilation in cats with healthy lungs

    Aim: To study whether pulmonary stretch receptor (PSR) activity in mechanically ventilated young cats with healthy lungs during partial liquid ventilation (PLV) is different from that during gas ventilation (GV). Methods: In 10 young cats (4.4 +/- 0.4 months, 2.3 +/- 0.3 kg; mean B SD), PSR instantaneous impulse frequency (PSR f(imp)) was recorded from single fibres in the vagal nerve during GV and PLV with perfluorocarbon (30 ml/kg) at increasing positive inspiratory pressures (PIP; 1.2, 1.8, 2.2 and 2.7 kPa), and at a positive end-expiratory pressure of 0.5 kPa. Results: All PSRs studied during GV maintained their phasic character with increased impulse frequency during inspiration during PLV. Peak PSR fimp was lower at PIP 1.2 kPa (p < 0.05) and at PIP 2.7 kPa (p = 0.10) during PLV than during GV, giving a lower number of PSR impulses at these two settings during PLV (p < 0.05). Conclusion: The phasic character of PSR activity is similar during GV and PLV.     Structural basis for the clamping and Ca²⁺ activation of SNARE-mediated fusion by synaptotagmin

    Synapotagmin-1 (Syt1) interacts with both SNARE proteins and lipid membranes to synchronize neurotransmitter release to calcium (Ca2+) influx. Here we report the cryo-electron microscopy structure of the Syt1–SNARE complex on anionic-lipid containing membranes. Under resting conditions, the Syt1 C2 domains bind the membrane with a magnesium (Mg2+)-mediated partial insertion of the aliphatic loops, alongside weak interactions with the anionic lipid headgroups. The C2B domain concurrently interacts the SNARE bundle via the ‘primary’ interface and is positioned between the SNAREpins and the membrane. In this configuration, Syt1 is projected to sterically delay the complete assembly of the associated SNAREpins and thus, contribute to clamping fusion. This Syt1–SNARE organization is disrupted upon Ca2+-influx as Syt1 reorients into the membrane, likely displacing the attached SNAREpins and reversing the fusion clamp. We thus conclude that the cation (Mg2+/Ca2+) dependent membrane interaction is a key determinant of the dual clamp/activator function of Synaptotagmin-1

    Twelve Years of Stover Removal Increases Soil Erosion Potential without Impacting Yield

    Corn (Zea mays L.) stover (non-grain aboveground biomass) in the US Corn Belt is used increasingly for livestock grazing and co-feed and for cellulosic bioenergy production. Continuous stover removal, however, could alter long-term agricultural productivity by affecting soil organic C (SOC) and soil physical properties, indicators of soil fertility and erosion potential. In this study, we showed that 12 consecutive yr of 55% stover removal did not affect mean grain yields at any N fertilizer rate (4.5, 6.3, and 6.0 Mg ha−1 for 60, 120, and 180 kg N ha−1 yr−1, respectively) in a marginally productive, rainfed continuous corn system under no-till (NT). Although SOC increased in the top 30 cm of all soils since 1998 (0.54–0.79 Mg C ha−1 yr−1), stover removal tended to limit SOC gains compared with no removal. Near-surface soils (0–5-cm depth) were more sensitive to stover removal and showed a 41% decrease in particulate organic matter stocks, smaller mean weight diameter of dry soil aggregates, and lower abundance of water-stable soil aggregates compared with soils with no stover removal. Increasing N fertilizer rate mitigated losses in total water-stable aggregates in near-surface soils related to stover removal. Collectively, however, our results indicated soil structure losses in surface soils due to lower C inputs. Despite no effect on crop yields and overall SOC gains with time using NT management, annually removing stover for 12 yr resulted in a higher risk of wind and water erosion at this NT continuous corn site in the western Corn Belt
