On the application of radio frequency voltages to ion traps via helical resonators
Ions confined using a Paul trap require a stable, high voltage and low noise
radio frequency (RF) potential. We present a guide for the design and
construction of a helical coil resonator for a desired frequency that maximises
the quality factor for a set of experimental constraints. We provide an
in-depth analysis of the system formed from a shielded helical coil and an ion
trap by treating the system as a lumped element model. This allows us to
predict the resonant frequency and quality factor in terms of the physical
parameters of the resonator and the properties of the ion trap. We also compare
theoretical predictions with experimental data for different resonators, and
predict the voltage applied to the ion trap as a function of the Q-factor,
input power and the properties of the resonant circuit

Revealing the former bed of Thwaites Glacier using sea-floor bathymetry: Implications for warm-water routing and bed controls on ice flow and buttressing
Abstract. The geometry of the sea floor immediately beyond
Antarctica's marine-terminating glaciers is a fundamental control on
warm-water routing, but it also describes former topographic pinning points
that have been important for ice-shelf buttressing. Unfortunately, this
information is often lacking due to the inaccessibility of these areas for
survey, leading to modelled or interpolated bathymetries being used as
boundary conditions in numerical modelling simulations. At Thwaites Glacier
(TG) this critical data gap was addressed in 2019 during the first cruise of
the International Thwaites Glacier Collaboration (ITGC) project. We present more than 2000 km2 of new multibeam
echo-sounder (MBES) data acquired in exceptional sea-ice conditions
immediately offshore TG, and we update existing bathymetric compilations.
The cross-sectional areas of sea-floor troughs are under-predicted by up to
40 % or are not resolved at all where MBES data are missing, suggesting that
calculations of trough capacity, and thus oceanic heat flux, may be
significantly underestimated. Spatial variations in the morphology of
topographic highs, known to be former pinning points for the floating ice
shelf of TG, indicate differences in bed composition that are supported by
landform evidence. We discuss links to ice dynamics for an overriding ice
mass including a potential positive feedback mechanism where erosion of
soft erodible highs may lead to ice-shelf ungrounding even with little
or no ice thinning. Analyses of bed roughnesses and basal drag contributions
show that the sea-floor bathymetry in front of TG is an analogue for extant
bed areas. Ice flow over the sea-floor troughs and ridges would have been
affected by similarly high basal drag to that acting at the grounding zone
today. We conclude that more can certainly be gleaned from these 3D
bathymetric datasets regarding the likely spatial variability of bed
roughness and bed composition types underneath TG. This work also addresses
the requirements of recent numerical ice-sheet and ocean modelling studies
that have recognised the need for accurate and high-resolution bathymetry to
determine warm-water routing to the grounding zone and, ultimately, for
predicting glacier retreat behaviour.

Learn before Lecture: A Strategy That Improves Learning Outcomes in a Large Introductory Biology Class
Actively engaging students in lecture has been shown to increase learning gains. To create time for active learning without displacing content we used two strategies for introducing material before class in a large introductory biology course. Four to five slides from 2007/8 were removed from each of three lectures in 2009 and the information introduced in preclass worksheets or narrated PowerPoint videos. In class, time created by shifting lecture material to learn before lecture (LBL) assignments was used to engage students in application of their new knowledge. Learning was evaluated by comparing student performance in 2009 versus 2007/8 on LBL-related question pairs, matched by level and format. The percentage of students who correctly answered five of six LBL-related exam questions was significantly higher (p < 0.001) in 2009 versus 2007/8. The mean increase in performance was 21% across the six LBL-related questions compared with <3% on all non-LBL exam questions. The worksheet and video LBL formats were equally effective based on a cross-over experimental design. These results demonstrate that LBLs combined with interactive exercises can be implemented incrementally and result in significant increases in learning gains in large introductory biology classes
Molecular structure and biodegradation kinetics of Linear Alkylbenzene Sulphonates in sea water.
The present paper describes the results of the application of the biodegradation test proposed by the United States Environmental Protection Agency (USEPA) “Biodegradability in sea water” Office of Prevention, Pesticides, and Toxic Substances (OPPTS) 835.3160, to Linear Alkylbenzene Sulphonate (LAS), the synthetic surfactant with the highest consumption volume on a world-wide basis. High performance liquid chromatography (HPLC) has been employed for the separation and quantification of the different homologues and isomers of the surfactant. Water from the Bay of Cádiz (South–West of the Iberian peninsula) has been used as test medium. The results indicate how both lag and t50 time shows a significant linear relationship with the length of the alkyl chain of the homologue; the effect of this is that the homologues of longer chain length not only begin to degrade first but also degrade at a faster rate. Regarding the isomeric composition, it is observed that as the percentage of biodegradation increases, there is an increase in the proportion of internal isomers, in comparison with the isomeric relationships of the original test substanc
Exploiting the Role of Endogenous Lymphoid-Resident Dendritic Cells in the Priming of NKT Cells and CD8+ T Cells to Dendritic Cell-Based Vaccines
Transfer of antigen between antigen-presenting cells (APCs) is potentially a physiologically relevant mechanism to spread antigen to cells with specialized stimulatory functions. Here we show that specific CD8+ T cell responses induced in response to intravenous administration of antigen-loaded bone marrow-derived dendritic cells (BM-DCs), were ablated in mice selectively depleted of endogenous lymphoid-resident langerin+ CD8α+ dendritic cells (DCs), suggesting that the antigen is transferred from the injected cells to resident APCs. In contrast, antigen-specific CD4+ T cells were primed predominantly by the injected BM-DCs, with only very weak contribution of resident APCs. Crucially, resident langerin+ CD8α+ DCs only contributed to the priming of CD8+ T cells in the presence of maturation stimuli such as intravenous injection of TLR ligands, or by loading the BM-DCs with the glycolipid α-galactosylceramide (α-GalCer) to recruit the adjuvant activity of activated invariant natural killer-like T (iNKT) cells. In fact, injection of α-GalCer-loaded CD1d−/− BM-DCs resulted in potent iNKT cell activation, suggesting that this glycolipid antigen can also be transferred to resident CD1d+ APCs. While iNKT cell activation per se was independent of langerin+ CD8α+ DCs, some iNKT cell-mediated activities were reduced, notably release of IL-12p70 and transactivation of NK cells. We conclude that both protein and glycolipid antigens can be exchanged between distinct DC species. These data suggest that the efficacy of DC-based vaccination strategies may be improved by the incorporation of a systemic maturation signal aimed to engage resident APCs in CD8+ T cell priming, and α-GalCer may be particularly well suited to this purpose
Level models of continuing professional development evaluation: a grounded review and critique
Continuing professional development (CPD) evaluation in education has been heavily influenced by ‘level models’, deriving from the work of Kirkpatrick and Guskey in particular, which attempt to trace the processes through which CPD interventions achieve outcomes. This paper considers the strengths and limitations of such models, and in particular the degree to which they are able to do justice to the complexity of CPD and its effects. After placing level models within the broader context of debates about CPD evaluation, the paper reports our experience of developing such models heuristically for our own evaluation practice. It then draws on positivist, realist and constructivist traditions to consider some more fundamental ontological and epistemological questions to which they give rise. The paper concludes that level models can be used in a number of ways and with differing emphases, and that choices made about their use will need to reflect both theoretical choices and practical considerations
The positions of primary and secondary schools in the English school field: a case of durable inequality
In interviews as part of a research study of structural reform in England, some tension between primary head teachers and their secondary peers was evident. This was symptomatic of a long-standing difference in status between the two phases. At a time when relations between stakeholders in local systems are subject to change, we seek to understand anew why that might be the case and how the tension we found was evidence of a current difference of power within interactions between representatives of the phases. We analyse differences of size, resources, workforce, pedagogy and history, and how they have resulted in different, and differently valued, practices and professional identities. We explore how attributes of the two phases have been counterposed and how, in complex interaction with wider discourses of politics, gender and age, this process has invested the differences with meanings and values that tend to relegate attributes associated with primary school. By focusing on the activation of cumulative inequality in interactions, we contribute a complementary perspective to studies of perceived relative status and highlight the implications for understanding school positioning in local arenas as the role of local authorities is reduced
Spleen-Resident CD4+ and CD4− CD8α− Dendritic Cell Subsets Differ in Their Ability to Prime Invariant Natural Killer T Lymphocytes
One important function of conventional dendritic cells (cDC) is their high capacity to capture, process and present Ag to T lymphocytes. Mouse splenic cDC subtypes, including CD8α+ and CD8α− cDC, are not identical in their Ag presenting and T cell priming functions. Surprisingly, few studies have reported functional differences between CD4− and CD4+ CD8α− cDC subsets. We show that, when loaded in vitro with OVA peptide or whole protein, and in steady-state conditions, splenic CD4− and CD4+ cDC are equivalent in their capacity to prime and direct CD4+ and CD8+ T cell differentiation. In contrast, in response to α-galactosylceramide (α-GalCer), CD4− and CD4+ cDC differentially activate invariant Natural Killer T (iNKT) cells, a population of lipid-reactive non-conventional T lymphocytes. Both cDC subsets equally take up α-GalCer in vitro and in vivo to stimulate the iNKT hybridoma DN32.D3, the activation of which depends solely on TCR triggering. On the other hand, and relative to their CD4+ counterparts, CD4− cDC more efficiently stimulate primary iNKT cells, a phenomenon likely due to differential production of co-factors (including IL-12) by cDC. Our data reveal a novel functional difference between splenic CD4+ and CD4− cDC subsets that may be important in immune responses
Restorative urban open space: Exploring the spatial configuration of human emotional fulfilment in urban open space
The capacity of outdoor settings to benefit human well being is well established by research. Examples of restorative settings can be found throughout history and are still applied today in health-care facilities, as healing or restorative gardens for the sick, but their wider significance in the urban public realm remains insufficiently explored. A conceptual framework for restorative urban open space based on mosaics of linked and nested spaces woven into the urban fabric is presented. The concept synthesizes the theory of centres, pioneered in the 1970s and refined in recent work by architectural theorist Christopher Alexander, with material relating to social and ecological dimensions of outdoor spatial configuration. The concept argues for fundamental properties of order, as integrations of locational, directional and transitional spatial experience, which are present in the natural and cultural world and associated with human psychological benefit. This spatial arrangement may offer potential to resurrect people's connection with intuitively preferred forms and strengthen beneficial relations between human functioning and the spatial environment. © 2005 Landscape Research Group Ltd
