Identification of a cDNA encoding Ascorbate peroxidase from Pinus Pinaster Ait.
Poster apresentado no X Congresso Nacional de Biotecnologia "BIOTEC' 2003", Lisboa, Portugal em 2003.The maritime or wild pine (Pinus pinaster Ait.) is the conifer with the widest distribution in Portugal, being of an extreme economic importance to wood and paper industries and also as a large source of pitch, turpentine and resin. Pine forest decay is largely associated with abiotic and biotic stresses, which ultimately leads to excessive production of reactive oxygen species (ROS), thus resulting in damage at the cellular level. Antioxidants and antioxidative enzymes are referred to have a role in the interruption of uncontrolled oxidation cascades occurring in different cell compartments. Ascorbate peroxidase (APX) exists as several isoforms that play an important role in the scavenging of H2O2 in higher plants, preventing not only cellular damage but also the inhibition of cytosolic and chloroplastic enzyme activity.
The identification of cDNAs encoding ascorbate peroxidase isoenzymes was attempted by screening a Pinus pinaster cDNA library with homologous probes previously obtained by PCR amplification of cDNA using degenerated primers. A full cDNA sequence was obtained and the sequence analysis revealed high similarity with cytosolic soluble APX from higher plants. Petrographical and geochemical signatures of the Granja paragneisses (NW Ceará, Brasil)
The Granja Granulitic Complex
(GCC) constitutes a NE–SW
trending high-grade
metamorphic belt occupying the
western sector of the Médio
Coreaú Domain (NW Ceará,
Brasil) (Figs. 1 and 2).
According to the published data,
the GCC represents a segment
of the Paleoproterozoic
basement intensely reworked
during the Brasiliano Orogeny.
Lithologically, the GCC is mainly
composed of garnet-sillimanite
migmatitic paragneisses,
enclosing discontinuous lenses
of mafic granulites and
enderbites.
At outcrop scale, the GCC
gneisses are well-foliated fineto
medium-grained grey rocks
showing a stromatic layering
parallel to the main regional
fabric (Fig. 3). The occurrence of
lenses of leucocratic quartzfeldspathic
material interlayered
with the host paragneisses
indicates that these rocks
reached partial melting
conditions during the Brasiliano
orogenic event (≈ 600 Ma).
Petrographically, the gneisses
are characterized by the
presence of mineral
assemblages dominated by
garnet, sillimanite and biotite
with moderate amounts of
quartz, plagioclase, cordierite
and K-feldspar (Figs. 4a-b).
Twelve whole-rock samples
were analyzed for major and
trace elements. As shown in
Graph 1, all the analyzed
samples plot in the sedimentary
field. Their REE patterns closely
resemble those of PAAS (PostArchean
average Australian
Shale) confirming the
sedimentary origin of the
paragneiss protoliths (Graph 2).
Geochemical data also suggest
that these sediments derived
from source areas of felsic to
intermediate composition and
were deposited in a tectonically
active continental margin
(Graphs 3 and 4)
Restraints to Anticoagulation Prescription in Atrial Fibrillation and Attitude Towards the New Oral Anticoagulants
Introdução e Objetivos: Avaliar a taxa de prescrição de anticoagulantes orais na fibrilhação auricular, os fatores associados à não prescrição, os motivos referidos pelos clínicos para não prescrição de anticoagulantes incluindo os de nova geração e realizar estudo evolutivo a médio prazo.
Material e Métodos: Estudo prospetivo sobre casos consecutivos de doentes com fibrilhação auricular com alta hospitalar. Registaram- se os scores CHA2DS2VASc e HASBLED, comorbilidades associadas e a medicação prévia e à data de alta. Na alta hospitalar, o médico assistente indicou em questionário o motivo de não prescrição de anticoagulantes orais e dos novos anticoagulantes orais.
Exclusão: contra-indicação absoluta para anticoagulação, CHA2DS2VASc ≤ 1 e doença valvular. Os doentes foram reavaliados um ano
após o recrutamento do primeiro doente.
Resultados: Identificaram-se 103 candidatos a anticoagulação oral (79,6 ± 8,0 anos; CHA2DS2VASc 5,8 ± 1,4; HASBLED 2,6 ± 1,0;
HASBLED ≥ 3 em 55,3%); os anticoagulantes foram prescritos em 34,0%. Fatores associados à não prescrição por ordem decrescente de relevância: uso prévio de antiagregantes, doente acamado e/ou demente, ausência de insuficiência cardíaca e número de fatores de risco hemorrágico. Razões invocadas para não prescrição por ordem decrescente de frequência: risco hemorrágico elevado, pequeno benefício, incapacidade de seguir o esquema terapêutico e dificuldade na monitorização da razão normalizada internacional(INR). Os novos anticoagulantes não foram prescritos e as razões invocadas foram, por ordem decrescente de frequência: informação insuficiente sobre estes fármacos, risco hemorrágico elevado, custo elevado e pequeno benefício. Aos 8,2 ± 2,5 meses de estudo evolutivo 33,3% dos doentes encontravam-se sob anticoagulação sem que os novos anticoagulantes tivessem sido prescritos.
Conclusões: Nesta amostra, a taxa de prescrição de anticoagulação oral foi baixa e o fator mais associado à não prescrição foi o uso prévio de antiagregantes. O impedimento à prescrição mais referido foi o risco hemorrágico, seguido do pequeno benefício reconhecido.
Os principais impedimentos referidos à prescrição dos novos anticoagulantes foram a informação insuficiente e o alto risco
hemorrágico. A médio prazo, a proporção de doentes sob anticoagulação mantinha-se baixa e os novos anticoagulantes não tinham
sido prescritos

Desenvolvimento de espécies florestais nativas em sistema de integração lavoura-pecuária-floresta.
Este experimento foi implantado na Fazenda Canchim, São Carlos, SP. A área experimental estava formada por Brachiaria decumbens. As árvores foram plantadas em três linhas, acompanhando o nível do terreno e com distancia entre árvores 2,5 m x 2,5m, resultando em cerca de 600 árvores/ha. As espécies florestais plantadas na linha central, ao acaso, foram: angico-branco (Anadenanlhera colubrina); canafistula (Pellophorum dubium); ipê-felpudo (Zeyheria tuberculosa); jequitibá-branco (Cariniana eslre/lensis) e pau-jacaré (Pipladenia gonoacanlha). Para o tutoramento destas espécies e para disponibilizar recursos para a fauna, foram plantadas duas linhas marginais de mutambo (Guazuma ulmifolia) e de capixingui (Crolon jloribundus) alternados. As árvores avaliadas, correspondeRAM A 10% do total do experimento. Foram analisados alatura (cm), diâmetro do colo e diâmetro a altura do peito (mm). O diâmetro da base do caule foi medido com auxilio de paquímetro entre dois e cinco cm do colo; o diâmetro a altura do peito (DAP) corresponde ao diâmetro do caule a 1,30 m de altura. A altura foi medida até a gema apical em plantas até 1,30 m de altura, a partir de 1,30 m, altura total passou a ser medida como o comprimento desde a base até o final da copa. Para os resultados foram consideradas as médias obtidas por quadrados mínimos, sendo o teste de hipótese entre médias realizad as por meio do teste de Tukey. Houve diferença significativa (P<0,05) entre as espécies em relação a todas as variáveis estudadas. As espécies que apresentaram maior altura média ao final da estação chuvosa ( maio/2009, aos 16 meses de idade) foram capixingui (128,95 cm) e mutambo (129,72 cm), seguidas por pau-jacaré (90,10 cm). Canatlstula (73,00 cm) e angico-branco (60,55 cm) ficaram numa posição intermediária, enquanto ipê-felpudo (17,19 cm) e jequitibá-branco (52,24 cm) tiveram menor desempenho. As espécies com bom desempenho cresceram principalmente na estação chuvosa. O desenvolvimento em diâmetro do colo acompanhou padrão semelhante ao de crescimento em altura, com destaques para capixingui (26,76 mm) e mutambo (29,67 mm), com valores intermediários para pau-jacaré (11,69 mm) e canafistula (18,44 mm), e menores valores para angico-branco (9,26 mm), ipê-felpudo (7,88 mm) e jequitibá-branco (9,12 mm). Conclui-se que os melhores desempenhos foram obtidos por capixingui e mutambo visto que são arvores de crescimento pioneiro e utilizadas como tutoras das demais espécies, dentre as demais espécies destaca-se o pau-jacaré. leguminosa fixadora de nitrogênio, Jequitibá-branco e, ipê-felpudo obtiveram os piores resultados nesse primeiro ano de implanatação do sistema
The Portuguese observatory on occupational psychosocial factors: contribution for public health
To achieve the goal of sustainable employment, considering
the profile of the Portuguese working population (PWP), is
needed a range of strategies to ensure long, productive, and
sustainable careers allied with a better quality of working life,
health, and wellbeing, but also with public health policies
grounded on scientifically validated and reliable data. This is
possible through a comprehensive working system approach
that ensures workers will be mentally and physically able to
remain at work by the balance between work demands and
individual resources allied with public health policies transfer
into the workplaces by organizations’ leadership and policy
makers. The Portuguese Observatory on Occupational Factors
(Popsy@Work) aims at addressing this global challenge by: i)
digitally collecting psychosocial data on the PWP; ii)
implementing and strengthening of a psychosocial occupational
health surveillance digital system; iii) providing
reference values for the PWP concerning Psychosocial
Health; iv) Transferring to society knowledge and best
practices; v) Raising awareness on the importance of
psychosocial management in occupational settings based on
science. Popsy@work is a digital platform that collects and
aggregates psychosocial data analytically and creates a
visualization hub adding value to data on the PWP and
giving science back to society in a usable way, empowering
workers, strengthening organizations and grounding public
policies. Pospy@Work considers the development of strategic
intelligence on levels and inequalities of psychosocial health
and well-being in occupational settings by robust metrics and
reference data. Creating opportunities for national policy
dialogue on inequalities, including the psychosocial health of
the PWP through collaboration with diverse sectors identifying
and mapping subgroups of populations whose unmet needs
require specific outreach measures
