    Evaluation the best condition of Fibrinolytic protease production using factorial design by Streptomyces sp DPUA 1573

In the pharmaceutical industry there is a search for new fibrinolytic agent that reduces the production cost and increasing productivity. The use of microorganism for enzyme production, such as the genus Streptomyces has been reported. Streptomyces is a Gram-positive bacteria, responsible for producing many bioactive compounds and extracellular enzymes of pharmaceutical interest. This study aimed to evaluated the production of fibrinolytic protease by Streptomyces sp DPUA 1573. Microbial cells were cultivated in the ISP2 for 48 hours, after this period the strains were inoculated in MS2 (soybean medium) that according with factorial design 24 (concentrations of soybean 0.5; 1.0 and 1.5%, glucose 0; 0.5 and 1.0% and different speeds 150 rpm; 200 rpm and 250 rpm and temperature 28C; 30C and 32C). The factorial design was analyzed by variance analysis (anova) and the glucose concentration showed a positive and significative effect. The results showed that the variable interaction had significant effect. that the best condition was composed 1.5% soybean, 1% glucose, 28 ºC and 150 speed in 48 hours, with production fibrinolytic 1391.66 U/mL. These values were higher than those reported in the literature. However these results show the biggest potencial in production fibrinolytic enzyme by Streptomyces.info:eu-repo/semantics/publishedVersio

    Diluted Magnetic Semiconductors Nanocrystals: Saturation and Modulation

    Diluted Magnetic Semiconductor (DMS) nanocrystals are a new class of materials formed by doping the semiconductor with transition metals (TM), which gives interesting magneto-optical properties. These properties are attributed to the exchange interaction between the pure semiconductor’s sp-electrons and the localized TM d-electrons. This book chapter shows exciting results of new DMS developed by the group, both in powder form and embedded in glassy systems. Depending on the concentration of doping ions, saturation of the incorporation of substitutional and interstitial sites in the nanocrystal structure may occur, forming other nanocrystals. In this context, we investigated the doping saturation limit in nanopowders of DMS Zn1-xMnxO NCs and Zn1-xMnxTe, Zn0.99-xMn0.01CoxTe, and Bi2-xCoxS NCs synthesized in glassy matrices. Thus, the sites’ saturation into the crystalline lattice of nanocrystals is a topic little reported in the literature, and we will comment on this work. Therefore, we will show results from the group about the modulation and saturation in diluted magnetic semiconductors nanocrystals in this work

    Antifungal benzo[b]thiophene 1,1-dioxide IMPDH inhibitors exhibit pan-assay interference (PAINS) profiles

    Fungi cause serious life-threatening infections in immunocompromised individuals and current treatments are now complicated by toxicity issues and the emergence of drug resistant strains. Consequently, there is a need for development of new antifungal drugs. Inosine monophosphate dehydrogenase (IMPDH), a key component of the de novo purine biosynthetic pathway, is essential for growth and virulence of fungi and is a potential drug target. In this study, a high-throughput screen of 114,000 drug-like compounds against Cryptococcus neoformans IMPDH was performed. We identified three 3-((5-substituted)-1,3,4-oxadiazol-2-yl)thio benzo[b]thiophene 1,1-dioxides that inhibited Cryptococcus IMPDH and also possessed whole cell antifungal activity. Analogs were synthesized to explore the SAR of these hits. Modification of the fifth substituent on the 1,3,4-oxadiazole ring yielded compounds with nanomolar in vitro activity, but with associated cytotoxicity. In contrast, two analogs generated by substituting the 1,3,4-oxadiazole ring with imidazole and 1,2,4-triazole gave reduced IMPDH inhibition in vitro, but were not cytotoxic. During enzyme kinetic studies in the presence of DTT, nucleophilic attack of a free thiol occurred with the benzo[b]thiophene 1,1-dioxide. Two representative compounds with substitution at the 5 position of the 1,3,4-oxadiazole ring, showed mixed inhibition in the absence of DTT. Incubation of these compounds with Cryptococcus IMPDH followed by mass spectrometry analysis showed non-specific and covalent binding with IMPDH at multiple cysteine residues.     Motor, linguistic, personal and social aspects of children with Down syndrome

    AbstractA global developmental delay is expected from Down syndrome, affecting motor, cognitive, linguistic and personal-social skills. However, not always these delays are proportional; different conditions occur due to several intrinsic and extrinsic variables that must be controlled to form groups of greater homogeneity.Objective To enhance personal-social, fine motor-adaptive, gross motor and linguistic skills among children with Down syndrome and compare them with typically developing children, matched for gender, socioeconomic status and mental age, while controlling some variables that interfere with the global development.Methods The ethical aspects were fulfilled (Case No. 040/2009). The following inclusion criteria were considered: participants without a history of prematurity, very low birth weight, congenital hypothyroidism, significant hearing and vision problems, and signs of Autism Spectrum Disorder. After the inclusion criteria were considered, 40 children participated in the study, of which 20 had Down syndrome (experimental group - EG), these being of both genders and with chronological ages ranging from 38 to 63 months, and the other 20 being typically developing children (control group - CG), matching the EG in terms of gender, socioeconomic status and mental age, with this age ranging from 13 to 50 months. The evaluation consisted in applying the Denver Developmental Screening Test II, a test that assesses areas such as personal-social, fine motor-adaptive, linguistic and gross motor development. The results were subjected to statistical analysis using Student’s t-test.Results A statistically significant difference was verified between the groups for the language and fine motor-adaptive areas.Conclusion Children with Down syndrome showed lower performance in language and fine motor skills when compared with typically developing children. There was no statistically significant difference in gross motor and personal-social areas. It is worth mentioning the importance of controlling the variables to deal with more homogeneous groups

    Gene-Expression Signatures Can Distinguish Gastric Cancer Grades and Stages

    Microarray gene-expression data of 54 paired gastric cancer and adjacent noncancerous gastric tissues were analyzed, with the aim to establish gene signatures for cancer grades (well-, moderately-, poorly- or un-differentiated) and stages (I, II, III and IV), which have been determined by pathologists. Our statistical analysis led to the identification of a number of gene combinations whose expression patterns serve well as signatures of different grades and different stages of gastric cancer. A 19-gene signature was found to have discerning power between high- and low-grade gastric cancers in general, with overall classification accuracy at 79.6%. An expanded 198-gene panel allows the stratification of cancers into four grades and control, giving rise to an overall classification agreement of 74.2% between each grade designated by the pathologists and our prediction. Two signatures for cancer staging, consisting of 10 genes and 9 genes, respectively, provide high classification accuracies at 90.0% and 84.0%, among early-, advanced-stage cancer and control. Functional and pathway analyses on these signature genes reveal the significant relevance of the derived signatures to cancer grades and progression.     Teatro na educação de crianças e adolescentes participantes de ensaio clínico

    OBJETIVO: Analisar os efeitos de uma intervenção pedagógica na aprendizagem de crianças e adolescentes participantes de pesquisa clínica. MÉTODOS: Estudo quantitativo, quasi-experimental e longitudinal, parte de um conjunto de estudos envolvidos no teste de uma vacina contra ancilostomíase. Amostra por conveniência com 133 estudantes de dez a 17 anos, de ambos os sexos, da Escola Municipal de Maranhão, MG, Brasil, 2009. Utilizou-se um questionário estruturado aplicado pré e pós-intervenção. O dispositivo pedagógico foi o Teatro do Oprimido. As variáveis dependentes foram o conhecimento específico e global sobre pesquisa clínica e sobre verminoses; a variável independente foi a participação na intervenção educativa. RESULTADOS: Houve aumento do conhecimento sobre sinais e sintomas, susceptibilidade à reinfecção e modo de contágio da verminose após a intervenção educativa. Aumentaram acertos relativos à duração da pesquisa clínica, aos procedimentos previstos, à possibilidade de desistência da participação e de ocorrência de eventos adversos. Permaneceu a noção de que o propósito primário da pesquisa é terapêutico, embora tenha reduzido o percentual de participantes que associaram a pesquisa ao tratamento médico. O Teatro do Oprimido possibilitou que as discussões acerca da helmintose e da pesquisa clínica fossem contextualizadas e materializadas. Os sujeitos puderam se despojar ou reduzir suas representações prévias. CONCLUSÕES: A participação de crianças e adolescentes em ensaios clínicos deve ser precedida de intervenção educativa, já que indivíduos dessa faixa etária nem sequer reconhecem que têm direito a decidir por si próprios
