Quantum-degenerate mixture of fermionic lithium and bosonic rubidium gases
We report on the observation of sympathetic cooling of a cloud of fermionic
6-Li atoms which are thermally coupled to evaporatively cooled bosonic 87-Rb.
Using this technique we obtain a mixture of quantum-degenerate gases, where the
Rb cloud is colder than the critical temperature for Bose-Einstein condensation
and the Li cloud colder than the Fermi temperature. From measurements of the
thermalization velocity we estimate the interspecies s-wave triplet scattering
length |a_s|=20_{-6}^{+9} a_B. We found that the presence of residual rubidium
atoms in the |2,1> and the |1,-1> Zeeman substates gives rise to important
losses due to inelastic collisions.Comment: 4 pages, 3 figure
Notch pathway repression by vestigial is required to promote indirect flight muscle differentiation in Drosophila melanogaster
AbstractDrosophila dorsal longitudinal muscles develop during metamorphosis by fusion of myoblasts with larval templates. It has been shown that both vestigial and Notch are crucial for correct formation of these muscles. We investigated the relationship between vestigial and the Notch pathway during this process. Using Enhancer of Split Region Transcript m6 gene expression as a reporter of Notch pathway activity, we were able to demonstrate that this pathway is only active in myoblasts. Moreover, close examination of the cellular location of several of the main actors of the N pathway (Notch, Delta, neuralized, Serrate, Mind bomb1 and fringe) during dorsal longitudinal muscle development enabled us to find that Notch receptor can play multiple roles in adult myogenesis. We report that the locations of the two Notch ligands (Delta and Serrate) are different. Interestingly, we found that fringe, which encodes a glycosyltransferase that modifies the affinity of the Notch receptor for its ligands, is expressed in muscle fibers and in a subset of myoblasts. In addition, we demonstrate that fringe expression is essential for Notch pathway inhibition and muscle differentiation. Lastly, we report that, in vestigial mutants, fringe expression is lost, and when fringe is overexpressed, a significant rescue of indirect flight muscle degeneration is obtained. Altogether, our data show that a vestigial-differentiating function is achieved through the inhibition of the Notch pathway
Application of magnetically induced hyperthermia on the model protozoan Crithidia fasciculata as a potential therapy against parasitic infections
Magnetic hyperthermia is currently an EU-approved clinical therapy against
tumor cells that uses magnetic nanoparticles under a time varying magnetic
field (TVMF). The same basic principle seems promising against trypanosomatids
causing Chagas disease and sleeping sickness, since therapeutic drugs available
display severe side effects and drug-resistant strains. However, no
applications of this strategy against protozoan-induced diseases have been
reported so far. In the present study, Crithidia fasciculata, a widely used
model for therapeutic strategies against pathogenic trypanosomatids, was
targeted with Fe_{3}O_{4} magnetic nanoparticles (MNPs) in order to remotely
provoke cell death using TVMFs. The MNPs with average sizes of d approx. 30 nm
were synthesized using a precipitation of FeSO_{4}4 in basic medium. The MNPs
were added to Crithidia fasciculata choanomastigotes in exponential phase and
incubated overnight. The amount of uploaded MNPs per cell was determined by
magnetic measurements. Cell viability using the MTT colorimetric assay and flow
cytometry showed that the MNPs were incorporated by the cells with no
noticeable cell-toxicity effects. When a TVMF (f = 249 kHz, H = 13 kA/m) was
applied to MNP-bearing cells, massive cell death was induced via a
non-apoptotic mechanism. No effects were observed by applying a TVMF on control
(without loaded MNPs) cells. No macroscopic rise in temperature was observed in
the extracellular medium during the experiments. Scanning Electron Microscopy
showed morphological changes after TVMF experiments. These data indicate (as a
proof of principle) that intracellular hyperthermia is a suitable technology to
induce the specific death of protozoan parasites bearing MNPs. These findings
expand the possibilities for new therapeutic strategies that combat parasitic
infections.Comment: 9 pages, four supplementary video file
In search of the role’s footprints in client-therapist dialogues
The goal of this research is to identify speaker's role via machine learning of broad acoustic parameters, in order to understand how an occupation, or a role, affects voice characteristics. The examined corpus consists of recordings taken under the same psychological paradigm (Process Work). Four interns were involved in four genuine client-therapist treatment sessions, where each individual had to train her therapeutic skills on her colleague that, in her turn, participated as a client. This uniform setting provided a unique opportunity to examine how role affects speaker's prosody. By a collection of machine learning algorithms, we tested automatic classification of the role across sessions. Results based on the acoustic properties show high classification rates, suggesting that there are discriminative acoustic features of speaker's role, as either a therapist or a client.info:eu-repo/semantics/publishedVersio
