    EXPRESSION OF USP22 AND CPC IN ORAL CANCER

    Oral cancer is a common cancer of the head and neck. Oral squamous cell carcinoma (OSCC) represents almost 90% of the total cases of head and neck cancer. Ubiquitin‑specific protease 22 (USP22) is a deubiquitinating hydrolase, and it is highly expressed in various types of cancer, which also typically have a poor prognosis. Aurora‑B and Survivin, which belong to the chromosomal passenger complex, are also highly expressed in a number of types of cancer. In the present study, USP22 expression and its associations with Aurora‑B and Survivin, and the clinicopathological features in OSCC were explored. USP22 is highly expressed in OSCC. Overexpression of USP22 is associated with lymph node metastasis and histological grade (P<0.01). Additionally, the expression of USP22 was positively associated with Aurora‑B (P<0.01), Survivin (P<0.01), and Ki‑67 (P<0.01). Furthermore, USP22 small interfering RNA inhibited cell growth and reduced the expression levels of Aurora‑B, Survivin and Cyclin B, together with the upregulation of cyclin‑dependent kinase inhibitor 1A (p21). These data suggest that USP22, Aurora‑B and Survivin promote the OSCC development and may represent novel targets for OSCC diagnosis and treatment in the future

    Understanding finite size effects in quasi-long-range orders for exactly solvable chain models

    In this paper, we investigate how much of the numerical artefacts introduced by finite system size and choice of boundary conditions can be removed by finite size scaling, for strongly-correlated systems with quasi-long-range order. Starting from the exact ground-state wave functions of hardcore bosons and spinless fermions with infinite nearest-neighbor repulsion on finite periodic chains and finite open chains, we compute the two-point, density-density, and pair-pair correlation functions, and fit these to various asymptotic power laws. Comparing the finite-periodic-chain and finite-openchain correlations with their infinite-chain counterparts, we find reasonable agreement among them for the power-law amplitudes and exponents, but poor agreement for the phase shifts. More importantly, for chain lengths on the order of 100, we find our finite-open-chain calculation overestimates some infinite-chain exponents (as did a recent density-matrix renormalization-group (DMRG) calculation on finite smooth chains), whereas our finite-periodic-chain calculation underestimates these exponents. We attribute this systematic difference to the different choice of boundary conditions. Eventually, both finite-chain exponents approach the infinite-chain limit: by a chain length of 1000 for periodic chains, and > 2000 for open chains. There is, howwever, a misleading apparent finite size scaling convergence at shorter chain lengths, for both our finite-chain exponents, as well as the finite-smooth-chain exponents. In recent years, with the increase in Aurora kinase-related research, the important role of Aurora kinases in tumorigenesis has been gradually recognized. Aurora kinases have been regarded as a new target for cancer therapy, resulting in the development of Aurora kinase inhibitors. The study and application of these small-molecule inhibitors, especially in combination with chemotherapy drugs, represents a new direction in cancer treatment.     The Broad Host Range Phage vB_CpeS_BG3P Is Able to Inhibit Clostridium perfringens Growth

    Clostridium perfringens is an important pathogen for both humans and animals, causing human foodborne disease and necrotic enteritis in poultry. In the present study, a C. perfringens-specific phage, vB_CpeS_BG3P (designated as BG3P hereafter), was isolated from chicken farm sewage. Both electron microscopy and phylogenetic analysis suggested that phage BG3P is a novel phage belonging to Siphoviridae family. Phage BG3P exhibited a broad host range against different C. perfringens isolates (90.63% of strains were infected). Sequencing of the complete genome revealed a linear double-stranded DNA (43,528 bp) with 28.65% GC content. After sequence analysis, 73 open reading frames (orf s) were predicted, of which only 13 were annotated with known functions. No tRNA and virulence encoding genes were detected. It should be noted that the protein of orf 15 has 97.92% homology to C. perfringens-specific chloramphenicol resistance protein, which has not been reported for any C. perfringens phage. Phylogenetic analysis of the ssDNA binding protein demonstrated that this phage is closely related to C. perfringens phages phiSM101 and phi3626. In considering future use as an antimicrobial agent, some biological characteristics were observed, such as a good pH (3–11) stability and moderate temperature tolerance (<60 C). Moreover, bacteriophage BG3P showed a good antimicrobial effect against C. perfringens liquid cultures. Thus, phage treatment with MOI ≥ 100 completely inhibited bacterial growth compared to untreated cultures. Although phage BG3P shows good lytic efficiency and broad host range in vitro, future development and application may need to consider removal of the chloramphenicol-like resistance gene or exploring its lysin for future antibacterial applications.This work was supported by the National Key Research and Development Program of China (No. 2018YFE0101900) and the Jiangsu Agricultural Science and Technology Foundation (No. cx(21)1004)

    A Forager Adjustment Strategy Used by the Bees Algorithm for Solving Optimization Problems in Cloud Manufacturing

    Intelligent technologies have become increasingly important in manufacturing nowadays. Optimal service management and allocation in current cloud manufacturing model are impossible without applications of appropriate intelligent tools. The Bees Algorithm (BA) is a swarm-based intelligent optimizer that provides support for smart decision-making process in manufacturing models. A novel forager adjustment strategy (FAS) is proposed in this paper to manage the forager division in the algorithm, so as to make the entire colony perform with higher efficiency. The proposed FAS based Bees Algorithm (FAS-BA) is able to realize flexible allocation of its forager resources between different roles in accordance with the solution fitness sampled by current scout population. The proposed algorithm is presented in detail. Experiments are conducted based on a set of well-known benchmark functions and a case study. Comparisons between FAS-BA and an improved Bees Algorithm are made to highlight the effectiveness of FAS. The results demonstrate that the proposed algorithm requires less function evaluation cost than the improved version but is capable of obtaining at least the same optimal solution to a problem.</jats:p

    Classification method of surrounding rock of plateau tunnel based on BP neural network

    Due to the unique high-altitude geological conditions of the railway in the cold region, the problem of high ground stress in the construction process is very prominent. In constructing high ground stress tunnels, accurately evaluating the surrounding rock grades is important in rock mass engineering. Based on this, based on a plateau tunnel under construction, this paper selects the classification index of the surrounding rock, which can accurately reflect the geological characteristics of high ground stress tunnel around the geological environment elements of the surrounding rock of high ground stress tunnel. Based on the rapid classification method of surrounding rock of the BP neural network, the classification method of the surrounding rock suitable for high ground stress tunnel is constructed, and the tunnel engineering data is introduced into the BP neural network classification method of surrounding rock for training and testing. It is found that the classification results of surrounding rock obtained by the classification method of surrounding rock of high ground stress tunnel are in good agreement with the actual situation, which provides an important guarantee for the accurate and rapid determination of the surrounding rock grade of high ground stress tunnel and the safe and efficient construction of the tunnel

    PARP6 acts as a tumor suppressor via downregulating Survivin expression in colorectal cancer

    Poly (ADP-ribose) polymerases (PARPs) are enzymes that transfer ADP-ribose groups to target proteins and are involved in a variety of biological processes. PARP6 is a novel member, and our previous findings suggest that PARP6 may act as a tumor suppressor via suppressing cell cycle progression. However, it is still unclear that PARP6 function besides growth suppression in colorectal cancer (CRC). In this study, we examined tumor suppressive roles of PAPR6 in CRC cells both in vitro and in vivo. We found that PARP6 inhibited colony formation, invasion and migration as well as cell proliferation. Moreover, ectopic overexpression of PARP6 decreased Survivin expression, which acts as an oncogene and is involved in apoptosis and mitosis. We confirmed the inverse correlation between PARP6 and Survivin expression in CRC cases by immunohistochemistry. Importantly, CRC cases with downregulation of PARP6 and upregulation of Survivin showed poor prognosis. In summary, PARP6 acts as a tumor suppressor via downregulating Survivin expression in CRC. PARP6 can be a novel diagnostic and therapeutic target together with Survivin for CRC
