    SEX-BASED DIFFERENCES IN TRUNK ACCELERATION AND LANDING POSTURE DURING THE DROP VERTICAL JUMP TEST

    The purpose of this study was to investigate sex-based differences in trunk acceleration and landing posture during the drop vertical jump test and to determine the relationships among relevant variables. Twenty college students (10 men and 10 women) performed drop vertical jumps from a 30cm high box. Trunk acceleration and the trunk, limb, knee, and ankle angles in the sagittal plane were measured. The unpaired Student t-test and Pearson product-moment correlation coefficients were used for statistical analysis. Female participants demonstrated greater vertical trunk acceleration and smaller trunk angles than male participants did. In addition, vertical acceleration correlated with trunk and limb angles. Therefore, these parameters, which can be estimated by using wearable sensors, may be useful for checking athletes at high risk for injury

    The generation of insulin-producing cells (IPCs) from pluripotent stem cells could be a breakthrough treatment for type 1 diabetes. However, development of new techniques is needed to exclude immature cells for clinical application. Dithizone staining is used to evaluate IPCs by detecting zinc. We hypothesised that zinc ion (Zn2+) dynamics reflect the IPC maturation status. Human adipose-derived stem cells were differentiated into IPCs by our two-step protocol using two-dimensional (2D) or 3D culture. The stimulation indexes of 2D -and 3D-cultured IPCs on day 21 were 1.21 and 3.64 (P < 0.05), respectively. The 3D-cultured IPCs were stained with dithizone during culture, and its intensity calculated by ImageJ reached the peak on day 17 (P < 0.05). Blood glucose levels of streptozotocin-induced diabetic nude mice were normalised (4/4,100%) after transplantation of 96 3D-cultured IPCs. Zn2+ concentration changes in the medium of 3D cultures had a negative value in the early period and a large positive value in the latter period. This study suggests that Zn2+ dynamics based on our observations and staining of zinc transporters have critical roles in the differentiation of IPCs, and that their measurement might be useful to evaluate IPC maturation as a non-destructive method

    ICG fluorescence catheter system in TaTME

    Background. Sometimes intraoperative urethral injury occurs in Transanal total mesorectal excision (TaTME). The aim of this study is to investigate the usefulness of indocyanine green (ICG) fluorescent catheter system for avoiding intraoperative urethral injury in TaTME in experimental model. Methods. A urethral catheter was filled with the mixture of albumin and ICG and raw hams were applied in layers as the surrogate model of rectourethral muscle. The detectability of ICG fluorescence in this catheter was investigated by using laparoscope-type fluorescence camera system. Results. Fluorescence was detected when ICG was mixed with albumin or peripheral blood. ICG fluorescence could be detected within 4 mm depth of layered raw hams as the surrogate model. Quantitative analysis of the picture detected that ICG fluorescence plateaued in lower concentration than that of serum. Conclusion. ICG fluorescent catheter system may be useful for avoiding intraoperative urethral injury in TaTME

    Equivalent circuit model modified for free-standing bilayer lipid membranes beyond 1 TΩ

    A cell is the basic functional unit of living organisms. Bilayer lipid membranes (BLMs), which form cell membranes can be assembled by using artificial methods. The electrochemical characteristics of BLMs are normally investigated using electrochemical impedance spectroscopy (EIS); however, the equivalent circuit need to be modified by the experimental conditions. In this study, we formed plain BLMs to determine the underlying equivalent circuit model of free-standing BLMs, and we measured the electrical characteristics using EIS. To analyze the results of EIS, we proposed equivalent circuit models including electrical double layer (EDL) effects on both sides of a BLM. We also extracted and evaluated the electrochemical parameters; the aperture-suspended BLMs using an Si chip having tapered edge recorded TΩ-order membrane resistances, which were one order higher than those reported in most previous studies.     Capacitance extraction method for a free-standing bilayer lipid membrane formed over an aperture in a nanofabricated silicon chip

    A bilayer lipid membrane (BLM) is the main component of the cell membrane of living organisms, which can be formed artificially. Although the specific capacitance of a BLM is known to be in the range of 0.4–1.0 μF cm^–2, many previous works that formed free-standing BLMs over an aperture in silicon chips reported larger values beyond this typical range, which suggests that equivalent-circuit models are not adequate. In this work, we modified the equivalent-circuit model by adding a resistance element of silicon. To evaluate the validity of the modified model, we applied the model to the results of electrochemical impedance spectroscopy for free-standing BLMs formed over an aperture in nanofabricated silicon chips. The derived specific capacitance values were 0.57 ± 0.08 μF cm^–2, which settles in the typical range

    Several studies have reported that the sagittal vertical axis(SVA)in patients with lumbar spinal stenosis(LSS)is associated with health-related quality of life(QOL)and low back pain. Therefore, it is required to obtain optimal standing spinal alignment. However, the functional factors that increase SVA in LSS patients have not been fully clarified. Therefore, the purpose of this study was to investigate correlations between quadriceps flexibility and spinal sagittal alignment in patients with lumbar spinal stenosis(LSS). We studied 30 LSS patients(16 males, 14 females, age 65.6±12.1 years). The quadriceps flexibility was evaluated by measuring the heel-to-buttock distance(HBD), and the standing spine alignment was evaluated by measuring each parameter from the whole spinal column X-ray sagittal plane image. Each relationship was examined using Spearmanʼs rank correlation coefficient. HBD was positively correlated with SVA(r=0.45, p<0.05)and PT(r=0.39, p<0.05), respectively. It was suggested that quadriceps flexibility was associated with SVA in LSS patients. In conclusion, improving quadriceps flexibility through preoperative physiotherapy intervention may result in good standing spinal alignment in LSS patients. Randomized controlled trials are needed to determine whether increased quadriceps flexibility in LSS patients improves SVA

    Objective: The purpose of this study was to verify the effect on spatial perception in healthy young subjects of an unconscious leftward optical shift created by a head-mounted display (HMD) with an offset camera. Methods: We recruited 40 healthy right-handed adults who were divided into four groups according to the hand used in the tests and the visual direction displayed by the HMD (centered or 10° left). Each of the four groups (n = 10) undertook line bisection tasks across four combinations of variables: using a finger/stick or a mouse to point at a touch panel located 60 or 120 cm away from the subject. Results: According to the results, regardless of the hand used, when the index finger or a stick was used (reaching condition), the line bisection point was displaced significantly to the left of the center. Additionally, a major left-displacement trend was observed in the short-distance reaching task, which did not require the use of a stick. In contrast, the long-distance task required a stick to be used, and the left displacements were all smaller than those for the short-distance tasks that used the index finger. Conclusion: This finding may be explained by the subjects having sufficient experience coordinating hand and eye movements in the condition where they used their dominant hand and reached with their own arms without using a stick.東京都立大学学位論文甲第1160号 副論

    Theranostic Agent Combining Fullerene Nanocrystals and Gold Nanoparticles for Photoacoustic Imaging and Photothermal Therapy

    Developing photoactivatable theranostic platforms with integrated functionalities of biocompatibility, targeting, imaging contrast, and therapy is a promising approach for cancer diagnosis and therapy. Here, we report a theranostic agent based on a hybrid nanoparticle comprising fullerene nanocrystals and gold nanoparticles (FGNPs) for photoacoustic imaging and photothermal therapy. Compared to gold nanoparticles and fullerene crystals, FGNPs exhibited stronger photoacoustic signals and photothermal heating characteristics by irradiating light with an optimal wavelength. Our studies demonstrated that FGNPs could kill cancer cells due to their photothermal heating characteristics in vitro. Moreover, FGNPs that are accumulated in tumor tissue via the enhanced permeation and retention effect can visualize tumor tissue due to their photoacoustic signal in tumor xenograft model mice. The theranostic agent with FGNPs shows promise for cancer therapy
