Unequal Perylene Diimide Twins in a Quadruple Assembly
Natural light-harvesting (LH) systems can divide identical dyes into unequal aggregate states, thereby achieving intelligent "allocation of labor". From a synthetic point of view, the construction of such kinds of unequal and integrated systems without the help of proteinaceous scaffolding is challenging. Here, we show that four octatetrayne-bridged ortho-perylene diimide (PDI) dyads (POPs) self-assemble into a quadruple assembly (POP)4 both in solution and in the solid state. The two identical PDI units in each POP are compartmentalized into weakly coupled PDIs (P520) and closely stacked PDIs (P550) in (POP)4 . The two extreme pools of PDI chromophores were unambiguously confirmed by single-crystal X-ray crystallography and NMR spectroscopy. To interpret the formation of the discrete quadruple assembly, we also developed a two-step cooperative model. Quantum-chemical calculations indicate the existence of multiple couplings within and across P520 and P550, which can satisfactorily describe the photophysical properties of the unequal quadruple assembly. This finding is expected to help advance the rational design of dye stacks to emulate functions of natural LH systems.</p
Impact of old age on resectable colorectal cancer outcomes
Objective This study was performed to identify a reasonable cutoff age for defining older patients with colorectal cancer (CRC) and to examine whether old age was related with increased colorectal cancer-specific death (CSD) and poor colorectal cancer-specific survival (CSS). Methods A total of 76,858 eligible patients from the surveillance, epidemiology, and end results (SEER) database were included in this study. The Cox proportional hazard regression model and the Chow test were used to determine a suitable cutoff age for defining the older group. Furthermore, a propensity score matching analysis was performed to adjust for heterogeneity between groups. A competing risk regression model was used to explore the impact of age on CSD and non-colorectal cancer-specific death (non-CSD). Kaplan–Meier survival curves were plotted to compare CSS between groups. Also, a Cox regression model was used to validate the results. External validation was performed on data from 1998 to 2003 retrieved from the SEER database. Results Based on a cutoff age of 70 years, the examined cohort of patients was classified into a younger group (n = 51,915, <70 years of old) and an older group (n = 24,943, ≥70 years of old). Compared with younger patients, older patients were more likely to have fewer lymph nodes sampled and were less likely to receive chemotherapy and radiotherapy. When adjusted for other covariates, age-dependent differences of 5-year CSD and 5-year non-CSD were significant in the younger and older groups (15.84% and 22.42%, P < 0.001; 5.21% and 14.21%, P < 0.001). Also an age of ≥70 years remained associated with worse CSS comparing with younger group (subdistribution hazard ratio, 1.51 95% confidence interval (CI) [1.45–1.57], P < 0.001). The Cox regression model as a sensitivity analysis had a similar result. External validation also supported an age of 70 years as a suitable cutoff, and this older group was associated with having reduced CSS and increased CSD. Conclusions A total of 70 is a suitable cutoff age to define those considered as having elderly CRC. Elderly CRC was associated with not only increased non-CSD but also with increased CSD. Further research is needed to provide evidence of whether cases of elderly CRC should receive stronger treatment if possible
High Glucose and Lipopolysaccharide Prime NLRP3 Inflammasome via ROS/TXNIP Pathway in Mesangial Cells
While inflammation is considered a central component in the development in diabetic nephropathy, the mechanism remains unclear. The NLRP3 inflammasome acts as both a sensor and a regulator of the inflammatory response. The NLRP3 inflammasome responds to exogenous and endogenous danger signals, resulting in cleavage of procaspase-1 and activation of cytokines IL-1β, IL-18, and IL-33, ultimately triggering an inflammatory cascade reaction. This study observed the expression of NLRP3 inflammasome signaling stimulated by high glucose, lipopolysaccharide, and reactive oxygen species (ROS) inhibitor N-acetyl-L-cysteine in glomerular mesangial cells, aiming to elucidate the mechanism by which the NLRP3 inflammasome signaling pathway may contribute to diabetic nephropathy. We found that the expression of thioredoxin-interacting protein (TXNIP), NLRP3, and IL-1β was observed by immunohistochemistry in vivo. Simultaneously, the mRNA and protein levels of TXNIP, NLRP3, procaspase-1, and IL-1β were significantly induced by high glucose concentration and lipopolysaccharide in a dose-dependent and time-dependent manner in vitro. This induction by both high glucose and lipopolysaccharide was significantly inhibited by N-acetyl-L-cysteine. Our results firstly reveal that high glucose and lipopolysaccharide activate ROS/TXNIP/ NLRP3/IL-1β inflammasome signaling in glomerular mesangial cells, suggesting a mechanism by which inflammation may contribute to the development of diabetic nephropathy
Social media and sensemaking patterns in new product development: demystifying the customer sentiment
Artificial intelligence by principle is developed to assist but also support decision making processes. In our study, we explore how information retrieved from social media can assist decision-making processes for new product development (NPD). We focus on consumers’ emotions that are expressed through social media and analyse the variations of their sentiments in all the stages of NPD. We collect data from Twitter that reveal consumers’ appreciation of aspects of the design of a newly launched model of an innovative automotive company. We adopt the sensemaking approach coupled with the use of fuzzy logic for text mining. This combinatory methodological approach enables us to retrieve consensus from the data and to explore the variations of sentiments of the customers about the product and define the polarity of these emotions for each of the NPD stages. The analysis identifies sensemaking patterns in Twitter data and explains the NPD process and the associated steps where the social interactions from customers can have an iterative role. We conclude the paper by outlining an agenda for future research in the NPD process and the role of the customer opinion through sensemaking mechanisms
Pavement structure assessment from the multi-source data of a moving vehicle
This paper proposed a novel assessment method for pavement structural status based on the multi-source data of a moving vehicle. The classification of pavement on subgrade (POS) and pavement on bridge deck (PBD) was taken as an example. A mobile acquisition system was designed to collect multi-source data. Vehicle accelerations on PBD and POS were analyzed, and some features were selected to build 15 Support Vector Machine (SVM) classifiers. Results show that some features are helpful, including vehicle speed, maximum value, minimum value, standard deviation of accelerations at time domain, and three peak frequencies at frequency domain. The accuracy was 91.67%. Then some sections were tested by a falling weight deflectometer (FWD) to verify the structural differences between POS and PBD. Accordingly, this paper initially generated the method for pavement structural analysis by vehicle vibration. A Characterization Method for Pavement Structural Condition Assessment Based on the Distribution Parameter of the Vehicle Vibration Signal
A pavement structural survey plays a vital role in road maintenance and management. This study was intended to explore the feasibility of a non-stop pavement structure assessment method by analyzing the vibration data from a vehicle sensor. In this study, three falling weight deflectometer (FWD) tests and four vehicle vibration tests were conducted on five pavement structures. The FWD test results show that the continuously reinforced composite pavement has a higher structural stiffness than the semi-rigid base asphalt pavement. According to the statistical distribution of vehicle acceleration, a distribution parameter, the peak probability density (PPD), was proposed. The correlation coefficient (−0.722) of the center deflection (D1) and PPD indicates a strong correlation between the two variables. Therefore, PPD is strongly correlated with pavement structural stiffness. This study proposed a novel characterization method for pavement structural conditions based on the distribution parameter of the vehicle vibration signal
Mechanism of innovation and standardization driving company competitiveness in the digital economy
Data have changed the characteristics of the global value chain in the context of digital economy. Standards and innovation are the key facilitators of digital economy. Considering the collaborative development of innovation and standardization, this paper constructs a theoretical model of the impact of innovation and standardization levels on company competitiveness. Furthermore, the moderating effect of the coupling coordination of innovation and standardization was analyzed for both factors. In total, 171 listed companies in the field of digital economy were selected as the research. The results indicated that a company’s level of innovation and standardization influences company competitiveness. Moreover, the relationship between innovation and standardization was found to be a coordinating development relationship and not a conventional promotion or hindrance. This article also proposed some pertinent suggestions for companies involved in the global value chain competition
Extracting subpixel vegetation NDVI time series for evaluating the mixed pixel effect on GPP estimation in urban areas
The normalized difference vegetation index (NDVI) is the most widely used vegetation index for monitoring vegetation vigor and cover. As NDVI time series are usually derived at coarse or medium spatial resolutions, pixel size often represents a mixture of vegetated and non-vegetated surfaces. In heterogeneous urban areas, mixed pixels impede the accurate estimation of gross primary productivity (GPP). To address the mixed pixel effect on NDVI time series and GPP estimation, we proposed a framework to extract subpixel vegetation NDVI (NDVIvege) from Landsat OLI images in urban areas, using endmember fractions, mixed NDVI (NDVImix), and NDVI of non-vegetation endmembers. Results demonstrated that the NDVIvege extracted by this framework agreed well with the true NDVIvege cross seasons and vegetation fractions, with R2 ranging from 0.74 to 0.82 and RMSE ranging from 0.03 to 0.04. The NDVIvege time series was applied to evaluate vegetation GPP in Wuhan, China. The total annual GPP estimated with NDVIvege was 28-35% higher than the total annual GPP estimated with NDVImix, implying uncertainty in the GPP estimations caused by mixed pixels. This study showed the potential of the proposed framework to resolve NDVIvege for characterizing vegetation dynamics in heterogeneous areas
Methyl 3,4-dihydroxybenzoate alleviates oxidative damage in granulosa cells by activating Nrf2 antioxidant pathway
Abstract Oxidative damage induced granulosa cells (GCs) apoptosis was considered as a significant cause of compromised follicle quality, antioxidants therapy has emerged as a potential method for improving endometriosis pregnancy outcomes. Here, we found that GCs from endometriosis patients show increased oxidative stress level. Methyl 3,4-dihydroxybenzoate (MDHB), a small molecule compound that is extracted from natural plants, reversed tert-butyl hydroperoxide (TBHP) induced GCs oxidative damage. Therefore, the aim of this study was to assess the protective effect of MDHB for GCs and its potential mechanisms. TUNEL staining and immunoblotting of cleaved caspase-3/7/9 showed MDHB attenuated TBHP induced GCs apoptosis. Mechanistically, MDHB treatment decreased cellular and mitochondria ROS production, improved the mitochondrial function by rescuing the mitochondrial membrane potential (MMP) and ATP production. Meanwhile, MDHB protein upregulated the expression of vital antioxidant transcriptional factor Nrf2 and antioxidant enzymes SOD1, NQO1 and GCLC to inhibited oxidative stress state, further beneficial to oocytes and embryos quality. Therefore, MDHB may represent a potential drug candidate in protecting granulosa cells in endometriosis, which can improve pregnancy outcomes for endometriosis-associated infertility
