    Modeling and identification of the dynamic behavior of stranded wire helical springs

    A stranded wire helical spring is a cylindrical helical spring wound by a wire strand. Owing to its unique structure, the spring features special dynamic behavior such as nonlinear stiffness, hysteresis and hardening overlap. The dynamic response model, which gives an accurate description of the dynamic behavior, of the spring is a very important tool for designing systems using the spring as well as evaluating the responses of such systems. However, no accurate model has been reported. In the present study, a modified normalized Bouc-Wen model is proposed to model the dynamic behavior of the spring. A simple yet effective identification method is developed for identifying the model parameters using experimental data. Numerical simulations and periodic loading experiments were carried out to validate the proposed model and identification method. The results verify that the proposed model and method are effective for modeling and identifying the dynamic behavior of stranded wire helical springs

    Investigation of the interaction between the MIR-503 and CD40 genes in irradiated U937 cells

    <p>Abstract</p> <p>Background</p> <p>MicroRNAs (miRNAs) are a group of small noncoding RNAs that take part in diverse biological processes by suppressing target gene expression. Relatively few miRNAs have been studied in detail, especially miR-503, and hence the biological relevance of majority remains to be uncovered. Whether altered expression of miRNA-503 affects the immunity response to radiotherapy has yet to be addressed.</p> <p>Results</p> <p>In the present study, we applied ionizing radiation with a dose of either 0.1 Gy or 5 Gy to irradiate U937 cells to confirm CD40 as a miR-503 target, which was identified using a bioimformatics tool. In high dose (5 Gy) ionizing-irradiated U937 cells, expression of miR-503 was up regulated while the expression of CD40 gene was down regulated. Using the transfection of the miR-503 gene into U937 cells and Luciferase assay, we confirmed that miR-503 suppressed the expression of CD40, and was a negtive regulator of CD40.</p> <p>Conclusions</p> <p>To our knowledge, we are the first to describe involvement of miR-503 in radiobiological effect at a molecular level. This initial finding suggested the evidence that ionizing radiation could alter the expression of miR-503 and its target gene CD40, and may be very important to shed light on a possible mechanism regarding regulation of immune responses to irradiation.</p

    Recent increases in terrestrial carbon uptake at little cost to the water cycle

    Quantifying the responses of the coupled carbon and water cycles to current global warming and rising atmospheric CO2 concentration is crucial for predicting and adapting to climate changes. Here we show that terrestrial carbon uptake (i.e. gross primary production) increased significantly from 1982 to 2011 using a combination of ground-based and remotely sensed land and atmospheric observations. Importantly, we find that the terrestrial carbon uptake increase is not accompanied by a proportional increase in water use (i.e. evapotranspiration) but is largely (about 90%) driven by increased carbon uptake per unit of water use, i.e. water use efficiency. The increased water use efficiency is positively related to rising CO2 concentration and increased canopy leaf area index, and negatively influenced by increased vapour pressure deficits. Our findings suggest that rising atmospheric CO2 concentration has caused a shift in terrestrial water economics of carbon uptake

    Comparative proteomics of human milk casein fraction collected from women of Korean and Han ethnic groups in China

    IntroductionHuman breast milk provides neonates with indispensable nutrition and function. Milk protein is one of the main constituents of breast milk. Human milk profiles can be influenced by many factors.MethodsThe present study aimed to investigate the difference in casein isolated from mature milk of healthy mothers of Korean and Han ethnic groups in China using data-independent acquisition (DIA) proteomics.ResultsA total of 535 proteins were identified and quantified in casein fraction samples from both groups. A total of 528 proteins were annotated to 52 Gene Ontology (GO) terms, the majority (94.13%) of which were distributed in the cell and cell parts of the cellular component. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that 106 proteins were involved in 23 pathways, the greatest (36.79%) in carbohydrate metabolism. There were 39 differentially expressed proteins (DEPs)–10 upregulated and 29 downregulated–between Korean and Han milk. The GO function of blood microparticles and KEGG pathway of Staphylococcus aureus infection for DEPs were the most significantly enriched (p &lt; 0.05). Protein-protein interaction analysis revealed a network with 23 DEPs in 47 interactions, and the fibrinogen alpha chain ranked first as the hub protein.DiscussionThese data may provide useful technical guidance for the development of specific infant foods for certain populations

    Serum metabolomics analysis in patients with alcohol dependence

    ObjectiveAlcohol dependence (AD) is a chronic recurrent mental disease caused by long-term drinking. It is one of the most prevalent public health problems. However, AD diagnosis lacks objective biomarkers. This study was aimed to shed some light on potential biomarkers of AD patients by investigating the serum metabolomics profiles of AD patients and the controls.MethodsLiquid chromatography-mass spectrometry (LC–MS) was used to detect the serum metabolites of 29 AD patients (AD) and 28 controls. Six samples were set aside as the validation set (Control: n = 3; AD group: n = 3), and the remaining were used as the training set (Control: n = 26; AD group: n = 25). Principal component analysis (PCA) and partial least squares discriminant analysis (PCA-DA) were performed to analyze the training set samples. The metabolic pathways were analyzed using the MetPA database. The signal pathways with pathway impact &gt;0.2, value of p &lt;0.05, and FDR &lt; 0.05 were selected. From the screened pathways, the metabolites whose levels changed by at least 3-fold were screened. The metabolites with no numerical overlap in their concentrations in the AD and the control groups were screened out and verified with the validation set.ResultsThe serum metabolomic profiles of the control and the AD groups were significantly different. We identified six significantly altered metabolic signal pathways, including protein digestion and absorption; alanine, aspartate, and glutamate metabolism; arginine biosynthesis; linoleic acid metabolism; butanoate metabolism; and GABAergic synapse. In these six signal pathways, the levels of 28 metabolites were found to be significantly altered. Of these, the alterations of 11 metabolites changed by at least 3-fold compared to the control group. Of these 11 metabolites, those with no numerical overlap in their concentrations between the AD and the control groups were GABA, 4-hydroxybutanoic acid, L-glutamic acid, citric acid and L-glutamine.ConclusionThe metabolite profile of the AD group was significantly different from that of the control group. GABA, 4-hydroxybutanoic acid, L-glutamic acid, citric acid, and L-glutamine could be used as potential diagnostic markers for AD

    Reticular crack is generally found on the surface of ceramic material that has been subjected to a thermal-shock condition. In the present study, a quantitative effect of thermal shock and quench temperature has been studied and investigated. Experimental tests were carried out to characterize the reticular crack that has been found in the Ge Kiln, which is a famous art of the ancient Chinese culture. After comparative analysis between thermal-shock cracks and the glaze crack patterns of the Ge Kiln porcelain, it is found that this study is expected to provide a powerful tool for recurrence of the long-lost firing and cooling process of the Ge Kiln porcelain
