Characterization of Navassa National Wildlife Refuge: A preliminary report for NF-06-05 (NOAA ship "Nancy Foster", April 18-30, 2006)
Navassa is a small, undeveloped island in the Windward Passage between Jamaica and Haiti. It was designated a National Wildlife Refuge under the jurisdiction of the U.S. Fish and Wildlife Service in 1999, but the remote location makes management and enforcement challenging, and the area is regularly fished by artisanal fishermen from Haiti. In April 2006, the NOAA Center for Coastal Fisheries and Habitat Research conducted a research cruise to Navassa. The cruise produced the first high-resolution multibeam bathymetry for the area, which will facilitate habitat mapping and assist in refuge management. A major emphasis of the cruise was to study the impact of Haitian fishing gear on benthic habitats and fish communities; however, in 10 days on station only one small boat was observed with five fishermen and seven traps. Fifteen monitoring stations were established to characterize fish and benthic communities along the deep (28-34 m) shelf, as these areas have been largely unstudied by previous cruises. The fish communities included numerous squirrelfishes, triggerfishes, and parrotfishes. Snappers and grouper were also present but no small individuals were observed. Similarly, conch surveys indicated the population was in low abundance and was heavily skewed towards adults. Analysis of the benthic photoquadrats is currently underway. Other cruise activities included installation of a temperature logger network, sample collection for stable isotope analyses to examine trophic structure, and drop camera surveys to ground-truth habitat maps and overhead imagery. Use of Data Comm by Flight Crew to Conduct Interval Management Operations to Parallel Dependent Runways
The Interval Management (IM) concept is being developed as a method to maintain or increase high traffic density airport arrival throughput while allowing aircraft to conduct near idle thrust descents. The Interval Management with Spacing to Parallel Dependent Runways (IMSPiDR1) experiment at NASA Langley Research Center used 24 commercial pilots to examine IM procedures to conduct parallel dependent runway arrival operations while maintaining safe but efficient intervals behind the preceding aircraft. The use of IM procedures during these operations requires a lengthy and complex clearance from Air Traffic Control (ATC) to the participating aircraft, thereby making the use of Controller Pilot Data Link Communications (CPDLC) highly desirable as the communication method. The use of CPDLC reduces the need for voice transmissions between controllers and flight crew, and enables automated transfer of IM clearance elements into flight management systems or other aircraft avionics. The result is reduced crew workload and an increase in the efficiency of crew procedures. This paper focuses on the subset of data collected related to the use of CPDLC for IM operations into a busy airport. Overall, the experiment and results were very successful, with the mean time under 43 seconds for the flight crew to load the clearance into the IM spacing tool, review the calculated speed, and respond to ATC. An overall mean rating of Moderately Agree was given when the crews were asked if the use of CPDLC was operationally acceptable as simulated in this experiment. Approximately half of the flight crew reported the use of CPDLC below 10,000 for IM operations was unacceptable, with 83% reporting below 5000 was unacceptable. Also described are proposed modifications to the IM operations that may reduce CPDLC Respond time to less than 30 seconds and should significantly reduce the complexity of crew procedures, as well as follow-on research issues for operational use of CPDLC during IM operations
Planar Double Bubbles on Flat Walls
In this paper, we investigate some properties of planar soap bubbles on a straight wall with a single corner. We show that when a wall has a single corner and a bubble consists of two connected regions, the perimeter minimizing bubble must be one of the types, two concentric circular arcs or a truncated standard double bubble, depending on the angle of the corner and areas of the regions
Opposing roles of PlexinA and PlexinB in axonal branch and varicosity formation
Establishing precise synaptic connectivity during development is crucial for neural circuit function. However, very few molecules have been identified that are involved in determining where and how many synapses form. The Plexin cell-surface molecules are a conserved family of axon guidance receptors that mediate axon fasciculation and repulsion during neural development, and later in development PlexinA receptors are involved in eliminating axonal branches and synapse numbers. Here we investigate the roles of PlexinA and PlexinB receptors in axonal branch and varicosity formation in Drosophila. We knocked down PlexinA or PlexinB expression using RNAi in identified mechanosensory neurons and analyzed axonal branching patterns and varicosity formations. Reducing PlexinA expression increased the axonal arbor complexity by increasing the number of branches and varicosities along the axon. In contrast, knocking down PlexinB expression decreased morphological complexity by decreasing the number of branches and the overall size of the axonal arbor, but did not reduce the number of varicosities. Our results demonstrate opposing roles for PlexinA and PlexinB in local wiring within a target region, where PlexinA functions to suppress excessive axonal branches and synapses and PlexinB facilitates axonal growth
The Development of Cockpit Display and Alerting Concepts for Interval Management (IM) in a Near-Term Environment
The National Aeronautics and Space Administration (NASA) Langley Research Center (LaRC) Interval Management (IM) research team has conducted a wide spectrum of work in the recent past, ranging from development and testing of the concept, procedures, and algorithm. This document focuses on the research and evaluation of the IM pilot interfaces, cockpit displays, indications, and alerting concepts for conducting IM spacing operations. The research team incorporated knowledge of human factors research, industry standards for cockpit design, and cockpit design philosophies to develop innovative displays for conducting these spacing operations. The research team also conducted a series of human-in-the-loop (HITL) experiments with commercial pilots and air traffic controllers, in as realistic a high-density arrival operation environment as could be simulated, to evaluate the spacing guidance display features and interface requirements needed to conduct spacing operations
Digital fragment analysis of short tandem repeats by high‐throughput amplicon sequencing
High‐throughput sequencing has been proposed as a method to genotype microsatellites and overcome the four main technical drawbacks of capillary electrophoresis: amplification artifacts, imprecise sizing, length homoplasy, and limited multiplex capability. The objective of this project was to test a high‐throughput amplicon sequencing approach to fragment analysis of short tandem repeats and characterize its advantages and disadvantages against traditional capillary electrophoresis. We amplified and sequenced 12 muskrat microsatellite loci from 180 muskrat specimens and analyzed the sequencing data for precision of allele calling, propensity for amplification or sequencing artifacts, and for evidence of length homoplasy. Of the 294 total alleles, we detected by sequencing, only 164 alleles would have been detected by capillary electrophoresis as the remaining 130 alleles (44%) would have been hidden by length homoplasy. The ability to detect a greater number of unique alleles resulted in the ability to resolve greater population genetic structure. The primary advantages of fragment analysis by sequencing are the ability to precisely size fragments, resolve length homoplasy, multiplex many individuals and many loci into a single high‐throughput run, and compare data across projects and across laboratories (present and future) with minimal technical calibration. A significant disadvantage of fragment analysis by sequencing is that the method is only practical and cost‐effective when performed on batches of several hundred samples with multiple loci. Future work is needed to optimize throughput while minimizing costs and to update existing microsatellite allele calling and analysis programs to accommodate sequence‐aware microsatellite data

Recommended Changes to Interval Management to Achieve Operational Implementation
A 19-day flight test of an Interval Management (IM) avionics prototype was conducted in Washington State using three aircraft to precisely achieve and maintain a spacing interval behind the preceding aircraft. NASA contracted with Boeing, Honeywell, and United Airlines to build this prototype, and then worked closely with them, the FAA, and other industry partners to test this prototype in flight. Four different IM operation types were investigated during this test in the en route, arrival, and final approach phases of flight. Many of the IM operations met or exceeded the design goals established prior to the test. However, there were issues discovered throughout the flight test, including the rate and magnitude of IM commanded speed changes and the difference between expected and actual aircraft deceleration rates
Wind Speed Dependence of Single-Site Wave-Height Retrievals from High-Frequency Radars
Wave-height observations derived from single-site high-frequency (HF) radar backscattered Doppler spectra are generally recognized to be less accurate than overlapping radar techniques but can provide significantly larger sampling regions. The larger available wave-sampling region may have important implications for observing system design. Comparison of HF radar–derived wave heights with acoustic Doppler profiler and buoy data revealed that the scale separation between the Bragg scattering waves and the peak energy-containing waves may contribute to errors in the single-site estimates in light-to-moderate winds. A wave-height correction factor was developed that explicitly considers this scale separation and eliminates the trend of increasing errors with increasing wind speed
