    Biodegradation of some agricultural residues by fungi in agitated submerged cultures

    Digestibility of agricultural residues in animal feeding is deeply dependent on the amounts and types of their fibers. Biological treatment of agricultural residues is a new method for improvement of digestibility. Therefore, the capacity of a few fungi in biodegradation of some agricultural residues wasstudied. Losses of crude fiber (CF), neutral detergent fiber (NDF) and acid detergent fiber (ADF) of wheat, barley, rice, wood, and pea straw were investigated in agitated submerged culture during biodegradation by fungi. Biodegradation of the plant residues is dependent on the plant and fungusspecies. The biodegradation order of plant residues was  pea>barley>wheat>rice>wood. A. terreus and T. reesei were more able to degrade the easy degradable plant residues. Rice and wood were degradedmore by Armillaria sp., Polyporus sp. and P. chrysosporium. Crude fiber, NDF and ADF of agricultural residues were reduced more by P. chrysosporium. Generally, the reduction of agricultural residues NDF by fungi was more than their ADF. However, Polyporus sp. decreased ADF of wheat straw more. Thus, for improvement of digestibility of agricultural residues, the treatment by white-rot fungi may be recommended

    Phytoplankton of Aras dam reservoir (Iran): an attempt to assess water quality

    The Aras reservoir, located in the north-west of Iran, plays an important role in fisheries, drinking and agricultural water supplies and recreational activities in the region. This study was performed to characterize the seasonal fluctuations of phytoplankton communities and their relationship with environmental factors in the Aras reservoir from August 2013 to May 2014. Sampling was carried out seasonally from 5 sampling locations. In each location three samples were taken for phytoplankton identification and enumeration, chemical analysis and chlorophyll a determination. In total, 72 species belonging to 5 divisions were determined. Cyanobacteria contained the highest density (74%) during the study period with Pseudanabaena limnetica as the most abundant species. This group retained its dominance the whole year round which indicated the poor quality and high nutrient load of the Aras reservoir, mainly due to human activities. On average, Trophic State Index (TSI) showed that water in the reservoir was eu-hypereutrophic. The results indicated that phytoplankton density negatively correlated with Secchi disc depth (R^2 = -0.479), total alkalinity (R^2 = -0.564), total hardness (R^2 = -0.727) and HCO_3 concentration (R^2 = -0.589). On the other hand, there was a positive correlation between the phytoplankton density and TP (R^2 = 0.734). A comparison between the present and a previous study indicated that the cyanobacterial bloom pattern in the Aras reservoir has shifted from warm season toward an all year round cycle which in addition to basin pollution due to anthropogenic activities, can be related to global warming and climate change

    The Effect of Wetting and Drying Cycles on Selected Physical Indicators of Biochar- and Rockwool-Based Growth Media

    IntroductionMonitoring the changes in physical and hydraulic properties and stability of growth media due to root growth effects and wetting and drying cycles is important. Wetting and drying cycles can probably change physical characteristics, availability of water, air and nutrients for the plant and, as a result, might affect the growth and yield of the greenhouse plants. The growth period greatly affects the physical characteristics of the growth substrates; therefore, the watering of growth substrates should be managed according to these changes to avoid improper irrigation.Materials and MethodsIn this study, 14 growth media were prepared from individual substrates with different volumetric ratios. In order to evaluate the changes of growth media over the time (i.e., during consecutive irrigation events) in the greenhouse, 10 wetting and drying cycles were applied on the growth media in the lab. Several physical indicators including easily available water (EAW), air after irrigation (AIR), water buffering capacity (WBC) and water holding capacity (WHC) of the growth media were determined before and after the wetting and drying cycles. Besides, the subsidence, decrease of mass and decomposition of the growth media were determined over the time. Total porosity (TP), bulk density (BD), particle density (PD), pH and electrical conductivity of the mixtures were measured as well.Results and DiscussionThe pH values in the growth media varied from 5.72 to 6.94. The maximum pH value was related to sawdust- sugarcane bagasse biochar produced at 300◦C vermiculite-zeolite, and wheat straw-vermiculite substrates, and the minimum value was related to the cocopeat-perlite substrate. The values of EC in the growth media varied from 0.21 to 1.43 dS m-1. The highest and lowest EC values among the growth substrates were related to date palm bunches-vermiculite-rockwool and rockwool (0.2)-perlite substrates, respectively. The bulk density (BD) values of the growth media varied in the range of 0.163-0.401 Mg m-3. The values ​​of total porosity (TP) of the growth media varied in the range of 64.8-82.8%v/v. The highest TP was related to the cocopeat-perlite substrate. The TP values ​​of most of the substrates were greater than 70%v/v. The average values of EAW in the growth substrates ranged from 0.123 to 0.272 cm3 cm-3. The highest EAW was related to the sawdust-sawdust biochar produced at 500 ◦C vermiculite-zeolite substrate. The application of wetting and drying cycles increased EAW in most of the growth media. Therefore, it can be stated that the time had a positive effect on the EAW in most of the growth media. The average values of AIR before and after the application of wetting and drying cycles for the growth media varied in the range of 0.063-0.240 cm-3 cm3. The highest value of this indicator was observed in the sawdust-date palm bunches biochar produced at 300◦C vermiculite substrate. In all substrates (with the exception of the sawdust-sawdust biochar produced at 500◦C vermiculite-zeolite), the AIR increased after wetting and drying cycles. The range of WHC values before and after applying wetting and drying cycles was 0.453-0.699 cm3 cm-3. The highest WHC belonged to the wheat straw-vermiculite substrate. The WHC values of five growth media, including cocopeat-perlite, decreased due to the application of wetting and drying cycles, and the WHC values of nine growth media decreased. The most stable substrate after the wetting and drying cycles was rockwool-sawdust-vermiculite. The effect of time on the quantity of WBC was positive, so that with the application of wetting and drying cycles, the WBC values of most of the substrates increased. In all substrates, subsidence and dry weight reduction were observed after the wetting and drying cycles. These changes were low for the substrates with a high volumetric ratio of inorganic materials. The least change among the growth substrates in terms of decomposition (dry weight reduction) was related to the completely inorganic substrate rockwool (0.1)-perlite (%0.17). The most stable substrate in terms of subsidence after wetting and drying cycles was the rockwool-sawdust-vermiculite, which has a large volumetric ratio of individual inorganic substrates. The highest subsidence was observed in the substrates containing wheat straw (wheat straw-vermiculite and date palm bunches biochar produced at 300◦C wheat straw-vermiculite). The organic matter content in all the growth substrates decreased over time (after wetting and drying cycles). The decrease of organic matter in the substrates can be related to the decomposition of organic materials as a result of wetting and drying cycles.ConclusionThe BD, TP, EAW and WHC of the majority of growth media were in the optimal ranges and for some mixtures even better than cocopeat-perlite. Wetting and drying cycles could affect the growth media through several processes such as decomposition of organic compounds, displacement and rearrangement of particles, fragmentation of particles, shrinkage, hardening and subsidence. The growth media with a high percent of organic substrates were unstable as compared with those containing a high proportion of inorganic substrates. In general, the wetting and drying cycles increased the frequency of micropores in the growth media. The wetting and drying cycles positively affected EAW, WHC, AIR and WBC of most growth media. These findings imply that wetting and drying cycles may improve the growth media according to the studied extensive variables. However, it is necessary to study the intensive variables such as hydraulic conductivity, oxygen diffusion and pore tortuosity in the growth media for better evaluation of the impact of wetting and drying cycles as well

    A Novel Cre Recombinase Imaging System for Tracking Lymphotropic Virus Infection In Vivo

    BACKGROUND:Detection, isolation, and identification of individual virus infected cells during long term infection are critical to advance our understanding of mechanisms of pathogenesis for latent/persistent viruses. However, current approaches to study these viruses in vivo have been hampered by low sensitivity and effects of cell-type on expression of viral encoded reporter genes. We have designed a novel Cre recombinase (Cre)-based murine system to overcome these problems, and thereby enable tracking and isolation of individual in vivo infected cells. METHODOLOGY/PRINCIPAL FINDINGS:Murine gammaherpesvirus 68 (MHV-68) was used as a prototypic persistent model virus. A Cre expressing recombinant virus was constructed and characterised. The virus is attenuated both in lytic virus replication, producing ten-fold lower lung virus titres than wild type virus, and in the establishment of latency. However, despite this limitation, when the sEGFP7 mouse line containing a Cre-activated enhanced green fluorescent protein (EGFP) was infected with the Cre expressing virus, sites of latent and persistent virus infection could be identified within B cells and macrophages of the lymphoid system on the basis of EGFP expression. Importantly, the use of the sEGFP7 mouse line which expresses high levels of EGFP allowed individual virus positive cells to be purified by FACSorting. Virus gene expression could be detected in these cells. Low numbers of EGFP positive cells could also be detected in the bone marrow. CONCLUSIONS/SIGNIFICANCE:The use of this novel Cre-based virus/mouse system allowed identification of individual latently infected cells in vivo and may be useful for the study and long-term monitoring of other latent/persistent virus infections

    Estradiol inhibits the effects of extracellular ATP in human sperm by a non genomic mechanism of action

    Steroid hormones, beside their classical genomic mechanism of action, exert rapid, non genomic effects in different cell types. These effects are mediated by still poorly characterized plasma membrane receptors that appear to be distinct from the classic intracellular receptors. In the present study we evaluated the non genomic effects of estradiol (17βE2) in human sperm and its effects on sperm stimulation by extracellular ATP, a potent activator of sperm acrosome reaction. In human sperm 17βE2 induced a rapid increase of intracellular calcium (Ca2+) concentrations dependent on an influx of Ca2+ from the extracellular medium. The monitoring of the plasma membrane potential variations induced by 17βE2 showed that this steroid induces a rapid plasma membrane hyperpolarization that was dependent on the presence of Ca2+ in the extracellular medium since it was absent in Ca2+ free-medium. When sperm were pre-incubated in the presence of the K+ channel inhibitor tetra-ethylammonium, the 17βE2 induced plasma membrane hyperpolarization was blunted suggesting the involvement of K+ channels in the hyperpolarizing effects of 17βE2. Extracellular ATP induced a rapid plasma membrane depolarization followed by acrosome reaction. Sperm pre-incubation with 17βE2 inhibited the effects of extracellular ATP on sperm plasma membrane potential variations and acrosome reaction. The effects of 17βE2 were specific since its inactive steroisomer 17αE2 was inactive. Furthermore the effects of 17βE2 were not inhibited by tamoxifen, an antagonist of the classic 17βE2 intracellular receptor

    Constraints on Energy Intake in Fish: The Link between Diet Composition, Energy Metabolism, and Energy Intake in Rainbow Trout

    The hypothesis was tested that fish fed to satiation with iso-energetic diets differing in macronutrient composition will have different digestible energy intakes (DEI) but similar total heat production. Four iso-energetic diets (2×2 factorial design) were formulated having a contrast in i) the ratio of protein to energy (P/E): high (HP/E) vs. low (LP/E) and ii) the type of non-protein energy (NPE) source: fat vs. carbohydrate which were iso-energetically exchanged. Triplicate groups (35 fish/tank) of rainbow trout were hand-fed each diet twice daily to satiation for 6 weeks under non-limiting water oxygen conditions. Feed intake (FI), DEI (kJ kg−0.8 d−1) and growth (g kg−0.8 d−1) of trout were affected by the interaction between P/E ratio and NPE source of the diet (P<0.05). Regardless of dietary P/E ratio, the inclusion of carbohydrate compared to fat as main NPE source reduced DEI and growth of trout by ∼20%. The diet-induced differences in FI and DEI show that trout did not compensate for the dietary differences in digestible energy or digestible protein contents. Further, changes in body fat store and plasma glucose did not seem to exert a homeostatic feedback control on DEI. Independent of the diet composition, heat production of trout did not differ (P>0.05). Our data suggest that the control of DEI in trout might be a function of heat production, which in turn might reflect a physiological limit related with oxidative metabolism

    Wnt5a Regulates Ventral Midbrain Morphogenesis and the Development of A9–A10 Dopaminergic Cells In Vivo

    Wnt5a is a morphogen that activates the Wnt/planar cell polarity (PCP) pathway and serves multiple functions during development. PCP signaling controls the orientation of cells within an epithelial plane as well as convergent extension (CE) movements. Wnt5a was previously reported to promote differentiation of A9–10 dopaminergic (DA) precursors in vitro. However, the signaling mechanism in DA cells and the function of Wnt5a during midbrain development in vivo remains unclear. We hereby report that Wnt5a activated the GTPase Rac1 in DA cells and that Rac1 inhibitors blocked the Wnt5a-induced DA neuron differentiation of ventral midbrain (VM) precursor cultures, linking Wnt5a-induced differentiation with a known effector of Wnt/PCP signaling. In vivo, Wnt5a was expressed throughout the VM at embryonic day (E)9.5, and was restricted to the VM floor and basal plate by E11.5–E13.5. Analysis of Wnt5a−/− mice revealed a transient increase in progenitor proliferation at E11.5, and a precociously induced NR4A2+ (Nurr1) precursor pool at E12.5. The excess NR4A2+ precursors remained undifferentiated until E14.5, when a transient 25% increase in DA neurons was detected. Wnt5a−/− mice also displayed a defect in (mid)brain morphogenesis, including an impairment in midbrain elongation and a rounded ventricular cavity. Interestingly, these alterations affected mostly cells in the DA lineage. The ventral Sonic hedgehog-expressing domain was broadened and flattened, a typical CE phenotype, and the domains occupied by Ngn2+ DA progenitors, NR4A2+ DA precursors and TH+ DA neurons were rostrocaudally reduced and laterally expanded. In summary, we hereby describe a Wnt5a regulation of Wnt/PCP signaling in the DA lineage and provide evidence for multiple functions of Wnt5a in the VM in vivo, including the regulation of VM morphogenesis, DA progenitor cell division, and differentiation of NR4A2+ DA precursors
