    Consistent Point Data Assimilation in Firedrake and Icepack

    We present methods and tools that significantly improve the ability to estimate quantities and fields which are difficult to directly measure, such as the fluidity of ice, using point data sources, such as satellite altimetry. These work with both sparse and dense point data with estimated quantities and fields becoming more accurate as the number of measurements are increased. Such quantities and fields are often used as inputs to mathematical models that are used to make predictions so improving their accuracy is of vital importance. We demonstrate how our methods and tools can increase the accuracy of results, ensure posterior consistency, and aid discourse between modellers and experimenters. To do this, we bring point data into the finite element method ecosystem as discontinuous fields on meshes of disconnected vertices. Point evaluation can then be formulated as a finite element interpolation operation (dual-evaluation). Our new abstractions are well-suited to automation. We demonstrate this by implementing them in Firedrake, which generates highly optimised code for solving PDEs with the finite element method. Our solution integrates with dolfin-adjoint/pyadjoint which allows PDE-constrained optimisation problems, such as data assimilation, to be solved through forward and adjoint mode automatic differentiation. We demonstrate our new functionality through examples in the fields of groundwater hydrology and glaciology

    Improved detection of global copy number variation using high density, non-polymorphic oligonucleotide probes

    <p>Abstract</p> <p>Background</p> <p>DNA sequence diversity within the human genome may be more greatly affected by copy number variations (CNVs) than single nucleotide polymorphisms (SNPs). Although the importance of CNVs in genome wide association studies (GWAS) is becoming widely accepted, the optimal methods for identifying these variants are still under evaluation. We have previously reported a comprehensive view of CNVs in the HapMap DNA collection using high density 500 K EA (Early Access) SNP genotyping arrays which revealed greater than 1,000 CNVs ranging in size from 1 kb to over 3 Mb. Although the arrays used most commonly for GWAS predominantly interrogate SNPs, CNV identification and detection does not necessarily require the use of DNA probes centered on polymorphic nucleotides and may even be hindered by the dependence on a successful SNP genotyping assay.</p> <p>Results</p> <p>In this study, we have designed and evaluated a high density array predicated on the use of non-polymorphic oligonucleotide probes for CNV detection. This approach effectively uncouples copy number detection from SNP genotyping and thus has the potential to significantly improve probe coverage for genome-wide CNV identification. This array, in conjunction with PCR-based, complexity-reduced DNA target, queries over 1.3 M independent NspI restriction enzyme fragments in the 200 bp to 1100 bp size range, which is a several fold increase in marker density as compared to the 500 K EA array. In addition, a novel algorithm was developed and validated to extract CNV regions and boundaries.</p> <p>Conclusion</p> <p>Using a well-characterized pair of DNA samples, close to 200 CNVs were identified, of which nearly 50% appear novel yet were independently validated using quantitative PCR. The results indicate that non-polymorphic probes provide a robust approach for CNV identification, and the increasing precision of CNV boundary delineation should allow a more complete analysis of their genomic organization.</p

    Entropic Fluctuations in Statistical Mechanics I. Classical Dynamical Systems

    Within the abstract framework of dynamical system theory we describe a general approach to the Transient (or Evans-Searles) and Steady State (or Gallavotti-Cohen) Fluctuation Theorems of non-equilibrium statistical mechanics. Our main objective is to display the minimal, model independent mathematical structure at work behind fluctuation theorems. Besides its conceptual simplicity, another advantage of our approach is its natural extension to quantum statistical mechanics which will be presented in a companion paper.     The effect of curricular and extracurricular activities on university students? entrepreneurial intention and competences

    This study examines how the alliance-building process affects the intention to enter into international alliances in the case of small and medium-sized enterprises (SMEs). From a psychological perspective (Perceived Behavioural Control), the authors analyse the alliance-building process as an inhibitor of the international collaboration intention, considering to what extent the experience affects the intention of the partners involved. The study explores these hypotheses based on a sample of 220 Spanish SMEs. The results provide empirical evidence showing that the intention to develop international alliances is negatively affected by the search and the selection process as well as by the negotiation of the agreement, which reduces the intention to establish an international agreement. In addition, the intention is moderated by the experience of the SME manager. Moreover, there is a negative relationship between the extent of the SME manager's international experience and the intention to develop an international alliance

    Relational practices and reflexivity: Exploring the responses of women entrepreneurs to changing household dynamics

    This qualitative study explores how and why women, positioned as mothers, wives, or carers, navigate changing household dynamics, related to care and reproductive resources, and become entrepreneurial. Drawing on relational reflexivity, we show how women’s embodied, intimate relations with important others in the household form the focal point for entrepreneurial activities and offer evidence of their entrepreneurial agency. Our analysis reveals the emergence of three relational practices that result in a new venture as the entrepreneurial response of women.     Defining and assessing enterprise capability in schools

    This paper describes the development of an instrument for assessing enterprise capability in schools. The approach to assessing enterprise capability builds on previous work by including three dimensions: self-efficacy, aspirations and knowledge and awareness. We find significant but weak associations between these three constructs suggesting that that, whilst they can be considered as providing a coherent description of enterprise capability they can also be regarded as distinct dimensions. The instrument also distinguishes between aspiration towards not-for-profit and for-profit enterprise and also between self-efficacy towards two broad enterprise capabilities: (i) project planning and (ii) working with people and information and two specific, market related capabilities: (iii) market risk and (iv) price and profit. We found only modest associations between students’ sense of enterpriser self-efficacy and their enterprise knowledge and awareness

    The effect of tourism education on students? entrepreneurial vocation

    This paper aims to examine how higher education affects entrepreneurial vocation in a very specific segment, namely university education in tourism. We used a theoretical approach based on the psychological foundations of intentional theory to analyse a sample of 122 graduate and undergraduate university students in tourism from the perspective of higher education. The results show the differential effect of curricular and extracurricular activities on intentions, attitudes and behavioural control, while there is very little effect on the development of entrepreneurial competencies
