    Comparative Cytogenetics Analysis of Chlamys farreri, Patinopecten yessoensis, and Argopecten irradians with C0t-1 DNA by Fluorescence In Situ Hybridization

    The chromosomes of Chlamys farreri, Patinopecten yessoensis, and Argopecten irradians were studied by FISH using C. farreri C0t-1 DNA probes. The results showed that C0t-1 DNA signals spread on all chromosomes in the three scallops, whereas signal density and intensity were different strikingly. Clustering brighter signals presented in the centromeric and telomeric regions of most C. farreri chromosomes, and in the centromeric or pericentromeric regions of several P. yessoensis chromosomes. Comparative analysis of the mapping indicated a relatively higher homology in the repetitive DNA sequences of the genome between C. farreri and P. yessoensis than that between C. farreri and A. irradians. In addition, FISH showed that the distribution of C0t-1 DNA clustering signals in C. farreri displayed completely similar signal bands between homologous chromosomes. Based on the C0t-1 DNA fluorescent bands, a more exact karyotype of C. farreri has been obtained. In this study, the comparative analysis based on C0t-1 DNA provides a new insight into the chromosomal reconstructions during the evolution process, and it is helpful for understanding an important source of genomic diversity, species relationships, and genome evolution

    Three-dimensional DEM investigation of the fracture behaviour of thermally degraded rocks with consideration of material anisotropy

    A complete understanding of the fracture behaviour of anisotropic rocks under elevated temperatures is fundamentally important for rock and reservoir engineering applications. This paper shows a three-dimensional numerical investigation of the fracture behaviour of anisotropic sandstone, with consideration of the effects of temperature and material anisotropy. In the study, a 3D semi-circular bend (SCB) model was established by using the Discrete Element Method (DEM). The thermal responses of different minerals and the strength anisotropy of incipient bedding planes were considered in the model. The DEM model was calibrated against a series of laboratory experiments on Midgley Grit sandstone (MGS) that exhibits intrinsic anisotropy. The pure mode I, mode II, and mixed-mode (I+II) fracture characteristics of the MGS were investigated under elevated temperatures (up to 600 °C) using the established DEM model. The thermal degradation (i.e., fracturing) of the rock, the fracture load, the evolution of micro-cracks, and the stress-strain relationship around notch tips were analysed, with emphasis on enlightening the micro-mechanisms underlying the fracture behaviour. The results of the study were discussed and then compared with experimental observations and theoretical predictions

    Cognitive decline and white matter changes in mesial temporal lobe epilepsy

    Noninvasive imaging plays a pivotal role in assessing the brain structural and functional changes in presurgical mesial temporal lobe epilepsy (MTLE) patients. Our goal was to study the relationship between the changes of cerebral white matter (WM) and cognitive functions in MTLE patients.Voxel-based morphometry (VBM) and tract-based spatial statistics (TBSS) MRI were performed on 24 right-handed MTLE patients (12 with left MTLE and 12 with right MTLE) and 12 matching healthy controls. Gray matter (GM), WM, and whole brain (WB) volumes were measured with VBM while fractional anisotropy (FA), mean diffusivity (MD), axial diffusivity (AD), and radial diffusivity (RD) were measured with TBSS. All patients and controls also underwent Montreal Cognitive Assessment (MoCA) before MRI.WM volume and the ratio of WM volume versus WB volume were significantly lower in MTLE patients compared with controls. WM volume in MTLE patients had a positive correlation with MoCA score (r = 0.71, P < .001) and a negative correlation with the duration of epilepsy (r = -0.693, P < .001). Volumetric differences were mainly located in the corpus callosum, uncinate fasciculus, inferior longitudinal fasciculus, and superior longitudinal fasciculus. FA of both left MTLE and right MTLE groups was significantly decreased, while MD, AD, and RD were significantly increased. Most left MTLE patients showed bilateral WM fiber tract changes versus ipsilateral changes for right MTLE patients.Changes in DTI parameters and WM volume were found in MTLE patients and more ipsilateral changes were seen with right-sided MTLE. Cognitive changes of MTLE patients were found to be correlated with the changes in WM structure. These findings not only provide useful information for lateralization of the seizure focus but can also be used to explain functional connectivity disorders which may be an important physiological basis for cognitive changes in patients with MTLE
