    Epigenetic modification does not determine the time of POU5F1 transcription activation in cloned bovine embryos.

    Purpose To investigate the effect of epigenetic modification on pattern, time and capacity of transcription activation of POU5F1, the key marker of pluripotency, in cloned bovine embryos. Methods Bovine fibroblasts were stably transfected with POU5F1 promoter-driven enhanced green fluorescent protein (EGFP). This provided a visible marker to investigate the effect of post-activation treatment of cloned bovine embryos with trichostatin A (TSA) on time and capacity of POU5F1 expression and its subsequent effect on in vitro development of cloned bovine embryos. Results Irrespective of TSA treatment, POU5F1 expression was not detected until 8–16 cell stage, but was detected in both inner cell mass and trophectoderm at the blastocyst stage. TSA treatment significantly increased POU5F1 expression, and the yield and quality of cloned embryo development compared to control. Conclusion The POU5F1 expression of cloned embryos is strictly controlled by the stage of embryo development and may not be altered by TSA-mediated changes occur in DNA-methylation and histone-acetylation of the genome

    AbstractBackground and aim:Nowadays, infertility and attitudes toward it, cause issues like depression, helplessness and anxiety. The present studyaimed to investigate the relationship between general health with happiness, inferiority feeling, and marital conflict ininfertile women.Methods and Materials:In this cross-sectional analytical study, 100 infertile women selected by convenience sampling. General HealthQuestionnaire (GHQ), the oxford happiness scale, inferiority feeling and Kansas Marital Conflict Scale (KMCS) wereused as data gathering tools. The Validity of the questionnaires were determined by the content and face validity. Thereliability of the questionnaires was determined using the internal consistency method. The calculated Cronbach’s alphafor general health questionnaire, happiness, inferiority feeling and the marital conflicts scale were 88%, 93%, 76%, and95% respectively. Data was analyzed using SPSS-16 software by a Pearson correlation coefficient and linear regressionResults:100 infertile women 18 - 43 years old with an average age of 29.3 were studied. 55% of women had moderate levels ofeconomic conditions and only 6% of them were referred to a psychologist or psychiatrist. The general health of thewomen with a minimum of 32 and maximum of 87 and a mean (56/14) was at moderate level. The results showed thatthere were significant relationship between general health, happiness and inferiority feeling (p <0.01), while there wasno significant relationship between marital conflict and general health. Regression analysis showed that happiness andinferiority feeling with respect to the beta coefficient (-0.56, 0.21) can vary significantly predict their general health.Conclusion:General health of infertile women could be affected by the happiness and inferiority feeling. According to the results, itseems that professional assistance from psychologists and clinical psychiatrists is useful in helping infertile patients tocope better with problems caused by infertility.Keywords:Health; Happiness; Inferiority Feeling; Marital Conflict; Infertilit

    Finding target units in FDH model by least-distance measure model

    summary:Recently, some authors used the Least-Distance Measure model in order to obtain the shortest distance between the evaluated Decision Making Unit (DMU) and the strongly efficient production frontier. But, their model is not applicable for situation in which the production possibility set satisfies free disposability property. In this paper, we propose a new approach to this end in FDH model which improves the application potential of the Least-Distance Measure and overcomes the mentioned shortcoming. The applicability of the proposed method is illustrated with two numerical examples and proves to be persuasive and acceptable to real-world problem

    Comparative survey of environmental health status of schools have health trainer and without health trainer in Bushehr province

    Background: School, as a reliable place in student's education, has an important role in training. If health principles do not comply with in schools, students may be suffering from infectious diseases. Health trainer can be effective in improving school health. This study aimed to determine the effectiveness of health trainer on environmental health level of schools in Bushehr province. Material and Methods This study was a descriptive - analytic study. A total of 20 samples were selected from each of the elementary schools have and not have health trainer using the method of stratified random sampling in the Bushehr province in the year 91-92. Data collected by researcher direct observation and using an assessment form of school environmental health contains 30 questions in 9 different sections. The statistical method used to analyze and test questions in this study were the chi square and Fisher exact test. Results: Based on findings of this research, between the environment health situation in schools with and without health trainer in 19 cases of health variables the significant difference was observed. The statistical analysis showed significant differences between health status of toilets (p<0.005), drinking water taps and buffet (p<0.001), yard and corridors (p<0.01), and solid waste disposal (p<0.025) in schools with and without health trainer in the year 91-92. Conclusion: Overall, the results indicate that the presence of health trainers in schools can lead to improved health status of the school. The schools without health trainer had very poor health status this issue required further try of concerned authorities to employee health trainers in schools

    Extracellular Production of Silver Nanoparticles by Using Three Common Species of Dermatophytes: Trichophyton rubrum, Trichophyton mentagrophytes and Microsporum canis

    ABSTRACT Background: To develop a new green approach for biosynthesis of silver nanoparticles, myconanotechnology has been represented as a novel field of study in nanotechnology. In this study, we have reported the extracellular synthesis of highly stable silver nanoparticles using three species of dermatophytes: Trichophyton rubrum, Trichophyton mentagrophytes and Microsporum canis. Methods: Clinical strains of these species were grown in a liquid medium containing mineral salt and incubated at 25°C for 5-7 days. The cell-free filtrate of each culture was obtained and subjected to synthesize silver nanoparticles in the presence of 1 mM AgNO 3 . Results: The reduction of Ag+ ions in metal nanoparticles was investigated virtually by tracing the solution color which was switched into reddish-light brown after 72 h. For T. mentagrophytes, a UV-visible spectra demonstrating a strong, quite narrow peak located between 422 and 425 nm was obtained. For M. canis, a fairly wide peak centering at 441 nm and for T. rubrum, a weak spectrum to decipher were observed. According to transmission electron microscopy (TEM) results, fairly uniform, spherical, and small in size with almost less than 50 nm particles were forms in case of T. mentagrophytes. For the other two species, TEM images showed existence of small spherical nanosilvers but not as small as nanoparticles synthesized by T. mentagrophytes. Conclusion: We observed that species belong to a single genus of the fungi have variable ability to synthesize silver nanoparticles extracellulary with different efficiency. Furthermore, the extracellular synthesis may make the process simpler and easier for following processes. Iran

    Case Report A Rare Case of Vasculitis Patched Necrosis of Cecum due to Behçet&apos;s Disease

    Isolated cecal necrosis is a rare form of acute ischemic colitis and a rare cause of surgical abdomen. Behçet&apos;s disease is a multisystemic autoimmune condition which can induce vasculitis. This can result in cecal necrosis while disease involves colon vessels. We describe a patient with complicated Behçet&apos;s disease and ischemic colitis admitted to our hospital. Patient was a 62-year-old female with more accompanying diseases. Histopathologic findings confirmed the diagnosis of ischemic colitis and regarding patient&apos;s vision problem and skin lesions, rheumatologic tests were performed which were positive for HLAB5 and HLAB51 suggestive of Behçet&apos;s disease; the patient was improved with surgery. Abdominal pain can indicate a disease with vascular involvement like Behçet&apos;s disease, especially in the presence of other clinical findings suggestive of the disease including blurred vision and skin lesions. An abdominal computerized tomography (CT) scan is very diagnostic in the same patients

    Symptomatic intestinal malrotation first presenting in the adults is rare. Midgut volvulus is the most common complication of malrotation in the adults. Because of more differential diagnosis, Computed Tomography (CT) scan can play an important role in the evaluation of patients with this abnormality. The whirl pattern around the superior mesenteric artery found on CT scan in patients with midgut volvulus is pathognomonic and diagnostic.     Cryopreservation of Semen in Domestic Animals: A Review of Current Challenges, Applications, and Prospective Strategies

    Cryopreservation is a way to preserve germplasm with applications in agriculture, biotechnology, and conservation of endangered animals. Cryopreservation has been available for over a century, yet, using current methods, only around 50% of spermatozoa retain their viability after cryopreservation. This loss is associated with damage to different sperm components including the plasma membrane, nucleus, mitochondria, proteins, mRNAs, and microRNAs. To mitigate this damage, conventional strategies use chemical additives that include classical cryoprotectants such as glycerol, as well as antioxidants, fatty acids, sugars, amino acids, and membrane stabilizers. However, clearly current protocols do not prevent all damage. This may be due to the imperfect function of antioxidants and the probable conversion of media components to more toxic forms during cryopreservation

    Biosynthesis of selenium nanoparticles using Klebsiella pneumoniae and their recovery by a simple sterilization process

    The use of biologically derived metal nanoparticles for various proposes is going to be an issue of considerable importance; thus, appropriate methods should be developed and tested for the biological synthesis and recovery of these nanoparticles from bacterial cells. In this research study, a strain of Klebsiella pneumoniae was tested for its ability to synthesize elemental selenium nanoparticles from selenium chloride. A broth of Klebsiella pneumoniae culture containing selenium nanoparticles was subjected to sterilization at 121ºC and 17 psi for 20 minutes. Released selenium nanoparticles ranged in size from 100 to 550 nm, with an average size of 245 nm. Our study also showed that no chemical changes occurred in selenium nanoparticles during the wet heat sterilization process. Therefore, the wet heat sterilization process can be used successfully to recover elemental selenium from bacterial cells
