    Restoration of Crown Hall

    This essay documents the research of restoration and modifications to Mies van der Rohe’s masterwork, Crown Hall, the heart of the Chicago campus of the Illinois Institute of Technology. Restoration was necessitated by 5 decades of use during which the building had fallen into major disrepair. During the restoration, practical and philosophical issues arose from the building’s landmark designation by regional and national authorities. The essay describes the forensic research that preceded design, investigation and selection of alternate materials modifications to the building envelope. This includes a detailed description of modifications balancing original materials and systems with current codes. The essay concludes by placing the restoration of Crown Hall in the larger context of preservation of modern buildings and the threat to these works which, unlike Crown Hall, are rarely protected by landmark designation

    Using a motorcycle rider behaviour questionnaire (MRBQ) to investigate the predictors of motorcyclists' crash risk

    In 2005, there were over 23,000 motorcyclists (including moped riders) involved in injury road traffic crashes in Great Britain and 6,142 of these riders were killed or seriously injured (Department for Transport, 2006). In order to reduce the casualty rates it is necessary to understand why motorcycle crashes happen. The present study approached this issue by investigating the role of aberrant rider behaviour, using a Motorcycle Rider Behaviour Questionnaire (MRBQ) as a framework. The aims of the study were to test the reliability and discriminant validity of the MRBQ and to examine which MRBQ behaviours relate to crash risk. Following the Driver Behaviour Questionnaire (e.g., Reason et al., 1990), which classifies driver behaviour into a system of errors ('the failure of planned actions to achieve their intended consequences') and violations ('deliberate deviations from those practices necessary to maintain the safe operation of a potentially hazardous system'), the MRBQ measured errors and violations, but with regard to motorcycling rather than car driving (see Elliott, Baughan & Sexton, 2007). The questionnaire consisted of 43 items to measure the self-reported frequency of specific riding behaviours. The questionnaire was administered to a sample of motorcyclists via a postal survey (N = 8,666). Principal components analysis revealed a 5-factor solution (TRAFFIC ERRORS, CONTROL ERRORS, SPEED VIOLATIONS, performance of STUNTS, and use of SAFETY EQUIPMENT). Generalised linear modelling showed that, while controlling for the effects of age, experience and annual mileage, TRAFFIC ERRORS were the main predictors of crash risk. For crashes in which respondents accepted some degree of blame, CONTROL ERRORS and SPEED VIOLATIONS were also significant predictors of crash risk. Implications of the findings will be discussed in relation to deciding on which countermeasures may be most effective at reducing motorcycle casualty rates

    Influence of the TARP γ8-selective negative allosteric modulator JNJ-55511118 on AMPA receptor gating and channel conductance

    AMPARs mediate excitatory signaling in the brain and are therapeutic targets for the treatment of diverse neurological disorders. The receptors interact with a variety of auxiliary subunits, including the transmembrane AMPAR regulatory proteins (TARPs). The TARPs influence AMPAR biosynthesis and trafficking and enhance receptor responses by slowing desensitization and deactivation and increasing single-channel conductance. TARP γ8 has an expression pattern that is distinct from other TARPs, being enriched in the hippocampus. Recently, several compounds have been identified that selectivity inhibit γ8-containing AMPARs. One such inhibitor, JNJ-55511118, has shown considerable promise for the treatment of epilepsy. However, key details of its mechanism of action are still lacking. Here, using patch-clamp electrophysiological recording from heterologously expressed AMPARs, we show that JNJ-55511118 inhibits peak currents of γ8-containing AMPARs by decreasing their single-channel conductance. The drug also modifies hallmark features of AMPAR pharmacology, including the TARP-dependent actions of intracellular polyamines and the partial agonist kainate. Moreover, we find that JNJ-5551118 reduces the influence of γ8 on all biophysical measures, aside from its effect on the recovery from desensitization. The drug is also effective when applied intracellularly, suggesting it may access its binding site from within the membrane. Additionally, we find that AMPARs incorporating TARP γ2 mutated to contain the JNJ-55511118 binding site, exhibit greater block than seen with AMPARs containing γ8, potentially reflecting differences in TARP stoichiometry. Taken together, our data provide new insight into the mechanism by which γ8-selective drugs inhibit AMPARs

    Software Citation in HEP: Current State and Recommendations for the Future

    In November 2022, the HEP Software Foundation (HSF) and the Institute for Research and Innovation for Software in High-Energy Physics (IRIS-HEP) organized a workshop on the topic of Software Citation and Recognition in HEP. The goal of the workshop was to bring together different types of stakeholders whose roles relate to software citation and the associated credit it provides in order to engage the community in a discussion on: the ways HEP experiments handle citation of software, recognition for software efforts that enable physics results disseminated to the public, and how the scholarly publishing ecosystem supports these activities. Reports were given from the publication board leadership of the ATLAS, CMS, and LHCb experiments and HEP open source software community organizations (ROOT, Scikit-HEP, MCnet), and perspectives were given from publishers (Elsevier, JOSS) and related tool providers (INSPIRE, Zenodo). This paper summarizes key findings and recommendations from the workshop as presented at the 26th International Conference on Computing In High Energy and Nuclear Physics (CHEP 2023).Comment: 7 pages, 2 listings.
