    Cooperativity and flexibility of the protonmotive activity of mitochondrial respiratory chain

    AbstractFunctional and structural data are reviewed which provide evidence that proton pumping in cytochrome c oxidase is associated with extended allosteric cooperativity involving the four redox centers in the enzyme . Data are also summarized showing that the H+/e− stoichiometry for proton pumping in the cytochrome span of the mitochondrial respiratory chain is flexible. The ΔpH component of the bulk-phase membrane electrochemical proton gradient exerts a decoupling effect on the proton pump of both the bc1 complex and cytochrome c oxidase. A slip in the pumping efficiency of the latter is also caused by high electron pressure. The mechanistic and physiological implications of proton-pump slips are examined. The easiness with which bulk phase ΔpH causes, at least above a threshold level, decoupling of proton pumping indicates that for active oxidative phosphorylation efficient protonic coupling between redox complexes and ATP synthase takes place at the membrane surface, likely in cristae, without significant formation of delocalized ΔμH+. A role of slips in modulating oxygen free radical production by the respiratory chain and the mitochondrial pathway of apoptosis is discussed

    Karalis is a paralle MPI, Finite-Volume, multiblock CFD code which solves the fully compressible Euler and Navier-Stokes equations where all couplings between dynamics and thermodynamics are allowed. This the most general mathematical model for all fluid flows. The code solves the coupled system of continuity, momentum and full energy equation for the velocity components, pressure and temperature. Once, u, v, w, p are and T are updated, arbitrary thermodynamics is supplied. The second order Roe’s upwind TVD scheme is used to compute convective fluxes through the Finite-Volume cell interfaces. A V-cycle Coarse Grid Correction Multi-Grid algorithm is used, together with a 5-stage Runge-Kutta explicit time-marching method, to accelerate convergence to a steady state. This formulation, typical of aerodynamic flows, shows an eccellent efficiency even for incompressible flows as well as for flows of incompressible fluids (typically buoyancy flows), once equipped with a preconditioner. Merkel’s preconditioner has been chosen because it can be easily formulated for arbitrary equations of state given as a functional relation of two independent thermodynamic variables (typically the pressure p and the temperature T), or even in tabular form, read in as an input file and used with bilinear interpolation. Karalis implement two among the most popular turbulence models, namely the one-equation model by Spalart and Allmaras and the two-equations model by Wilcox, the k-ω model, which allow a good compromise between accuracy, robustness and stability of turbulent calculations. Code validation is presented for some typical benchmark test cases of incompressible fluid dynamics. Comparison with solutions obtained with a few popular commercial CFD codes is also presented

    Size and age at onset of sexual maturity of female Norway lobster Nephrops norvegicus L. (Crustacea: Nephropidae) in the Strait of Sicily (Central Mediterranean Sea)

    The reproductive cycle of the Norway lobster (Nephrops norvegicus) female was investigated to determine the size and age at onset of sexual maturity (SOM), as a baseline to establish the minimum catchable size. Samples were gathered in four seasonal (Spring 1986 - Winter 1987) trawl surveys on the Italian side of the Strait of Sicily (Central Mediterranean Sea). The relative frequency of females in different stages of maturation and in berried conditions was observed. According to the ovary stages, gonadal maturation is attained during Spring and Summer; berried animals occur almost exclusively in Summer and Autumn, in about 40% of the sample. Length structure changes slightly by location and by season; recruitment to the gear occurs more or less continuously. Gaussian components appear to be highly overlapped; the resolution of the length frequency distribution values leads to the identification of multiple cohorts, five of which were well represented. The population attains 50% of maturity at 30-32 mm CL, corresponding to an age of 5 years. The mean size of the first Gaussian component from the catch (about 22 mm CL) is well below the estimated size at 50% maturity, suggesting a precocious recruitment to the fishery.No disponibl

    Risk factors and outcome among a large patient cohort with community-acquired acute hepatitis C in Italy

    BACKGROUND: The epidemiology of acute hepatitis C has changed during the past decade in Western countries. Acute HCV infection has a high rate of chronicity, but it is unclear when patients with acute infection should be treated. METHODS: To evaluate current sources of hepatitis C virus (HCV) transmission in Italy and to assess the rate of and factors associated with chronic infection, we enrolled 214 consecutive patients with newly acquired hepatitis C during 1999-2004. The patients were from 12 health care centers throughout the country, and they were followed up for a mean (+/- SD) period of 14+/-15.8 months. Biochemical liver tests were performed, and HCV RNA levels were monitored. RESULTS: A total of 146 patients (68%) had symptomatic disease. The most common risk factors for acquiring hepatitis C that were reported were intravenous drug use and medical procedures. The proportion of subjects with spontaneous resolution of infection was 36%. The average timespan from disease onset to HCV RNA clearance was 71 days (range, 27-173 days). In fact, 58 (80%) of 73 patients with self-limiting hepatitis experienced HCV RNA clearance within 3 months of disease onset. Multiple logistic regression analyses showed that none of the variables considered (including asymptomatic disease) were associated with increased risk of developing chronic hepatitis C. CONCLUSIONS: These findings underscore the importance of medical procedures as risk factors in the current spread of HCV infection in Italy. Because nearly all patients with acute, self-limiting hepatitis C - both symptomatic and asymptomatic - have spontaneous viral clearance within 3 months of disease onset, it seems reasonable to start treatment after this time period ends to avoid costly and useless treatment

    A Study of background for IXPE

    Focal plane X-ray polarimetry is intended for relatively bright sources with a negligible impact of background. However this might not be always possible for IXPE (Imaging X-ray Polarimetry Explorer) when observing faint extended sources like supernova remnants. We present for the first time the expected background of IXPE by Monte Carlo simulation and its impact on real observations of point and extended X-ray sources. The simulation of background has been performed by Monte Carlo based on GEANT4 framework. The spacecraft and the detector units have been modeled, and the expected background components in IXPE orbital environment have been evaluated. We studied different background rejection techniques based on the analysis of the tracks collected by the Gas Pixel Detectors on board IXPE. The estimated background is about 2.9 times larger than the requirement, yet it is still negligible when observing point like sources. Albeit small, the impact on supernova remnants indicates the need for a background subtraction for the observation of the extended sources.Comment: 16 pages, 16 figure
