    Measurement of Position Acuity in Strabismus and Amblyopia: Specificity of the Vernier VEP Paradigm

    PURPOSE. An objective measure of positional acuity is desirable in the nonverbal clinical population. This study was conducted to investigate the specificity of the vernier VEP as a measure of positional acuity, evaluating the potential confound of asymmetric motion responses that may be present in some groups of patients. These motion responses could masquerade as position-specific responses, since they occur at the same response frequency as the vernier-related response. METHODS. Twelve observers with early-onset esotropia (EOE), 30 children with untreated amblyopia, and 15 control children underwent swept vernier VEP acuity testing accompanied by a swept motion control stimulus. The control condition was used to detect the presence of artifactual responses not related to position sensitivity. The patients with EOE were selected for high levels of motion asymmetry as documented with oscillating gratings presented monocularly. As a measure of motion confound (penetration), the proportion of first-harmonic responses recorded in the control condition was determined. RESULTS. The penetration rate in the vernier condition in each study group (EOE: 0.93%; amblyopes: 4.26%; normal subjects: 2.40%) and the entire group (2.85%) was acceptably low. The level of penetration was not significantly influenced by the presence of amblyopia. CONCLUSIONS. The vernier VEP paradigm, when applied in the manner described, can be interpreted as a measure of position sensitivity. The presence of motion asymmetry or untreated amblyopia does not affect the validity of vernier measurements made. Visual evoked potentials (VEPs) provide another means of assessing visual function in nonverbal subjects. VEPs specific to vernier offsets have been measured in several studies. The steady state vernier VEP 26 -29 involves the periodic introduction and withdrawal of vernier offsets in a high-contrast square-wave grating target 26 A difficulty in applying the steady state vernier VEP in clinical populations is the possible presence of motion responses that are not symmetric. A clear example of asymmetric motion VEP responses occurs in patients with early-onset esotropia (EOE) in which the monocular response to rapidly oscillating gratings is dominated by odd-harmonic components that reflect the asymmetry of motion processing. 30 -35 A similar asymmetry is found in the monocular response of normal infants.     Shear-strain-induced two-dimensional slip avalanches in rhombohedral MoS2

    Slip avalanches are ubiquitous phenomena occurring in 3D materials under shear strain and their study contributes immensely to our understanding of plastic deformation, fragmentation, and earthquakes. So far, little is known about the role of shear strain in 2D materials. Here we show some evidence of two-dimensional slip avalanches in exfoliated rhombohedral MoS2, triggered by shear strain near the threshold level. Utilizing interfacial polarization in 3R-MoS2, we directly probe the stacking order in multilayer flakes and discover a wide variety of polarization domains with sizes following a power-law distribution. These findings suggest slip avalanches can occur during the exfoliation of 2D materials, and the stacking orders can be changed via shear strain. Our observation has far-reaching implications for developing new materials and technologies, where precise control over the atomic structure of these materials is essential for optimizing their properties as well as for our understanding of fundamental physical phenomena.Comment: To be published in Nano Letter

    HE4 (WFDC2) Promotes Tumor Growth in Endometrial Cancer Cell Lines

    HE4, also known as WFDC2, is a useful biomarker for ovarian cancer when either used alone or in combination with CA125. HE4 is also overexpressed in endometrial cancer (EC), but its function in cancer cells is not clear. In this study, we investigate the role of HE4 in EC progression. An HE4-overexpression system was established by cloning the HE4 prototypic mRNA variant (HE4-V0) into a eukaryotic expression vector. Following transfection, stable clones in two EC cell lines were selected. The effects of HE4 overexpression on cell growth and function were measured with the use of cell proliferation assay, matrigel invasion, and soft agar gel colony formation assays. HE4-induced cancer cell proliferation in vivo was examined in a mouse xenograft model. HE4 overexpression significantly enhanced EC cell proliferation, matrigel invasion, and colony formation in soft agar. Moreover, HE4 overexpression promoted tumor growth in the mouse xenograft model. HE4 overexpression enhanced several malignant phenotypes in cell culture and in a mouse model. These results are consistent with our previous observation that high levels of serum HE4 closely correlate with the stage, myometrial invasion and tumor size in patients with EC.     Crowded Letter and Crowded Picture LogMAR acuity in Children with Amblyopia: a quantitative comparison.

    Aims Clinically, picture acuity tests are thought to overestimate visual acuity (VA) compared with letter tests, but this has not been systematically investigated in children with amblyopia. This study compared VA measurements with the LogMAR Crowded Kay Picture test to the LogMAR Crowded Keeler Letter acuity test in a group of young children with amblyopia. Methods 58 children (34 male) with amblyopia (22 anisometropic, 18 strabismic and 18 with both strabismic/anisometropic amblyopia) aged 4-6 years (mean=68.7, range=48-83 months) underwent VA measurements. VA chart testing order was randomised, but the amblyopic eye was tested before the fellow eye. All participants wore up-to-date refractive correction. Results The Kay Picture test significantly overestimated VA by 0.098 logMAR (95% limits of agreement (LOA), 0.13) in the amblyopic eye and 0.088 logMAR (95% LOA, 0.13) in the fellow eye, respectively (p< 0.001). No interactions were found from occlusion therapy, refractive correction or type of amblyopia on VA results (p> 0.23). For both the amblyopic and fellow eyes, Bland-Altman plots demonstrated a systematic and predictable difference between Kay Picture and Keeler Letter charts across the range of acuities tested (Keeler acuity: amblyopic eye 0.75 to -0.05 logMAR; fellow eye 0.45 to -0.15 logMAR). Linear regression analysis (p< 0.00001) and also slope values close to one (amblyopic 0.98, fellow 0.86) demonstrate that there is no proportional bias. Conclusions The Kay Picture test consistently overestimated VA by approximately 0.10 logMAR when compared with the Keeler Letter test in young children with amblyopia. Due to the predictable difference found between both crowded logMAR acuity tests, it is reasonable to adjust Kay Picture acuity thresholds by +0.10 logMAR to compute expected Keeler Letter acuity scores

    The first comprehensive study of a giant nebula around a radio-quiet quasar in the z<1z < 1 Universe

    We present the first comprehensive study of a giant,  ⁣ ⁣70\approx \! \! 70 kpc-scale nebula around a radio-quiet quasar at z<1z<1. The analysis is based on deep integral field spectroscopy with MUSE of the field of HE\,0238-1904, a luminous quasar at z=0.6282z=0.6282. The nebula emits strongly in [OII]\mathrm{[O \, II]}, Hβ\rm H \beta, and [OIII]\mathrm{[O \, III]}, and the quasar resides in an unusually overdense environment for a radio-quiet system. The environment likely consists of two groups which may be merging, and in total have an estimated dynamical mass of Mdyn4×1013M_{\rm dyn}\approx 4\times 10^{13} to $10^{14}\ {\rm M_\odot}.Thenebulaexhibitslargelyquiescentkinematicsandirregularmorphology.Thenebulamayariseprimarilythroughinteractionrelatedstrippingofcircumgalacticandinterstellarmedium(CGM/ISM)ofgroupmembers,withsomepotentialcontributionsfromquasaroutflows.Thesimultaneouspresenceofthegiantnebulaandaradioquietquasarinarichenvironmentsuggestsacorrelationbetweensuchcircumquasarnebulaeandenvironmentaleffects.Thispossibilitycanbetestedwithlargersamples.Theupperlimitsontheelectronnumberdensityimpliedbythe. The nebula exhibits largely quiescent kinematics and irregular morphology. The nebula may arise primarily through interaction-related stripping of circumgalactic and interstellar medium (CGM/ISM) of group members, with some potential contributions from quasar outflows. The simultaneous presence of the giant nebula and a radio-quiet quasar in a rich environment suggests a correlation between such circum-quasar nebulae and environmental effects. This possibility can be tested with larger samples. The upper limits on the electron number density implied by the \mathrm{[O \, II]}doubletratiorangefrom doublet ratio range from \log(n_{\rm e, \, [O \, II]} / \mathrm{cm^{-3}}) < 1.2to to 2.8.However,assumingaconstantquasarluminosityandnegligibleprojectioneffects,thedensitiesimpliedfromthemeasuredlineratiosbetweendifferentions(e.g.,. However, assuming a constant quasar luminosity and negligible projection effects, the densities implied from the measured line ratios between different ions (e.g., \mathrm{[O\,II]},, \mathrm{[O\,III]},and, and \mathrm{[Ne\,V]})andphotoionizationsimulationsareoften) and photoionization simulations are often 10{-}400timeslarger.Thislargediscrepancycanbeexplainedbyquasarvariabilityonatimescaleof times larger.     A comparative investigation of emerging adults’ moral thinking and communication competencies in Taiwan, the USA, and the UK

    Emerging adulthood is a unique and distinct period demographically, subjectively, and in terms of identity exploration in developed and developing countries. This study aimed to investigate emerging adults’ moral thinking and communication competencies, and their differences by socio-demographic factors (i.e., gender, religious affiliation, college major, educational stages), in Taiwan, the USA, and the UK, as well as compare similarities/common trends and diversities between the three groups. We modified and utilized the MTC-II scale, including two dilemma stories relating to an individual and societal moral dilemma, to assess 743 valid sample participants. We found that females in the Taiwan group scored highest on the MTC-II scale across the three samples, whereas religious affiliation differentiated the USA and the UK groups. We also noted differences in scores relating to the two stories and associated moral levels. These findings are interpreted as a foundation for future research and educational practice

    DHODH modulates transcriptional elongation in the neural crest and melanoma

    Melanoma is a tumour of transformed melanocytes, which are originally derived from the embryonic neural crest. It is unknown to what extent the programs that regulate neural crest development interact with mutations in the BRAF oncogene, which is the most commonly mutated gene in human melanoma1. We have used zebrafish embryos to identify the initiating transcriptional events that occur on activation of human BRAF(V600E) (which encodes an amino acid substitution mutant of BRAF) in the neural crest lineage. Zebrafish embryos that are transgenic for mitfa:BRAF(V600E) and lack p53 (also known as tp53) have a gene signature that is enriched for markers of multipotent neural crest cells, and neural crest progenitors from these embryos fail to terminally differentiate. To determine whether these early transcriptional events are important for melanoma pathogenesis, we performed a chemical genetic screen to identify small-molecule suppressors of the neural crest lineage, which were then tested for their effects on melanoma. One class of compound, inhibitors of dihydroorotate dehydrogenase (DHODH), for example leflunomide, led to an almost complete abrogation of neural crest development in zebrafish and to a reduction in the self-renewal of mammalian neural crest stem cells. Leflunomide exerts these effects by inhibiting the transcriptional elongation of genes that are required for neural crest development and melanoma growth. When used alone or in combination with a specific inhibitor of the BRAF(V600E) oncogene, DHODH inhibition led to a marked decrease in melanoma growth both in vitro and in mouse xenograft studies. Taken together, these studies highlight developmental pathways in neural crest cells that have a direct bearing on melanoma formation

    Overexpression of IL-7 enhances cisplatin resistance in glioma

    Cisplatin is one of the most commonly used chemotherapeutic agents for glioma patients. In this study, array comparative genomic hybridization (aCGH) was used to identify genes associated with cisplatin resistance in a human glioma cell line. The cisplatin-resistant U251/CP2 cell line was derived by stepwise selection using cisplatin. The genetic aberrations of the U251 parental cell line and the U251/CP2 cells were analyzed using aCGH. RT-PCR was used to detect the expression of the altered genes revealed by aCGH. The sensitivity of glioma cells to cisplatin was determined by using the MTT assay. Apoptosis was detected using flow cytometry and western blot analysis. The IC50 value of cisplatin in U251/CP2 cells was five times higher than its IC50 in U251 cells. The U251 cells lost at least one copy each of the CFHR1 and CFHR3 genes, and both CFHR1 and CFHR3 were homozygously deleted in U251/CP2 cells. The U251/CP2 cells gained two to three copies of C8orf70 and IL-7 genes. IL-7 mRNA expression was studied in 12 glioma cell lines, and expression was positively correlated with the IC50 of cisplatin. Furthermore, IL-7 mRNA expression was also positively correlated with the IC50 of cisplatin in 91 clinical glioma specimens. Additionally, treatment with recombinant human IL-7 (rhIL-7) enhanced cisplatin resistance and increased the relative growth rate of the glioma cells. Moreover, the apoptosis induced by cisplatin could be inhibited by IL-7. In conclusion, our results suggest that IL-7 may play an important role in cisplatin resistance in glioma

    Numerical evaluation of inflationary 3-point functions on curved field space

    We extend the public CppTransport code to calculate the statistical properties of fluctuations in multiple-field inflationary models with curved field space. Our implementation accounts for all physical effects at tree-level in the 'in-in' diagrammatic expansion. This includes particle production due to time-varying masses, but excludes scenarios where the curvature perturbation is generated by averaging over the decay of more than one particle. We test our implementation by comparing results in Cartesian and polar field-space coordinates, showing excellent numerical agreement and only minor degradation in compute time. We compare our results with the PyTransport 2.0 code, which uses the same computational approach but a different numerical implementation, finding good agreement. Finally, we use our tools to study a class of gelaton-like models which could produce an enhanced non-Gaussian signal on equilateral configurations of the Fourier bispectrum. We show this is difficult to achieve using hyperbolic field-space manifolds and simple inflationary potentials
