    Rapid Liquid Chromatography-Tandem Mass Spectrometry Analysis of Two Urinary Oxidative Stress Biomarkers: 8-oxodG and 8-isoprostane.

    Human biomonitoring of oxidative stress relies on urinary effect biomarkers such as 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG), and 8-iso-prostaglandin F2α (8-isoprostane); however, their levels reported for similar populations are inconsistent in the scientific literature. One of the reasons is the multitude of analytical methods with varying degrees of selectivity used to quantify these biomarkers. Single-analyte methods are often used, requiring multiple injections that increase both time and cost. We developed a rapid ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method to quantify both urinary biomarkers simultaneously. A reversed-phase column using a gradient consisting of 0.1% acetic acid in water and 0.1% acetic acid in methanol/acetonitrile (70:30) was used for separation. The MS detection was by positive (8-oxodG) and negative (8-isoprostane) ion-mode by multiple reaction monitoring. Very low limit of detection (<20 pg/mL), excellent linearity (R2 > 0.999), accuracy (near 100%), and precision (CV < 10%) both for intra-day and inter-day experiments were achieved, as well as high recovery rates (>91%). Matrix effects were observed but were compensated by using internal standards. Our newly developed method is applicable for biomonitoring studies as well as large epidemiological studies investigating the effect of oxidative damage, as it requires only minimal clean up using solid phase extraction

    Spatial Distance Between Parents and Adult Children in the United States

    ObjectiveThis brief report presents contemporary national estimates of the spatial distance between residences of parents and adult children in the United States, including distance to one’s nearest parent or adult child and whether one lives near all of their parents and adult children.BackgroundThe most recent national estimates of parent–child spatial proximity come from data for the early 1990s. Moreover, research has rarely assessed the spatial clustering of all parents and adult children.MethodData are from the 2013 Panel Study of Income Dynamics on residential locations of adults aged 25 years and older and each of their parents and adult children. The following two measures of spatial proximity were estimated: the share of adults who have their nearest parent or adult child at a given distance and the share of adults who have all parents and/or all adult children at a given distance. Sociodemographic and geographic differences were examined for both measures.ResultsAmong the adults with at least one living parent or adult child, a significant majority (74.8%) had their nearest parent or adult child within 30 miles, and about one third (35.5%) had all parents and adult children living that close. Spatial proximity differed substantially among sociodemographic groups, with those who were disadvantaged more likely to have their parents or adult children nearby. In most cases, sociodemographic disparities were much higher when spatial proximity was measured by proximity to all parents and all adult children instead of to the nearest parent or nearest adult child.ConclusionDisparities in having all parents and/or adult children nearby may be a result of family solidarity and also may affect family solidarity.     Disparities in Vulnerability to Severe Complications from COVID-19 in the United States

    Preexisting health conditions increase vulnerability to severe complications from COVID-19. Among middle-aged and older Americans, vulnerability to severe COVID-19 complications based on preexisting conditions is 2-3 times greater for those with low versus high income. Vulnerability is about 40% higher for middle-aged and older adults with a high school degree or less than adults with a four-year college degree. In every age group, Blacks are more vulnerable than Whites, but Hispanics are at lower risk based on fewer preexisting health conditions

    The Cost-Effectiveness of New York City’s Safe Routes to School Program

    Objective. We evaluated the cost-effectiveness of a package of roadway modifications in New York City funded under the Safe Routes to School (SRTS) program. Methods. We used a Markov model to estimate long-term impacts of SRTS on injury reduction and the associated savings in medical costs, lifelong disability, and death. Model inputs included societal costs (in 2013 US dollars) and observed spatiotemporal changes in injury rates associated with New York City’s implementation of SRTS relative to control intersections. Structural changes to roadways were assumed to last 50 years before further investment is required. Therefore, costs were discounted over 50 consecutive cohorts of modified roadway users under SRTS. Results. SRTS was associated with an overall net societal benefit of $230 million and 2055 quality-adjusted life years gained in New York City. Conclusions. SRTS reduces injuries and saves money over the long run

    The effect of a diet with fructan-rich chicory roots on intestinal helminths and microbiota with special focus on Bifidobacteria and Campylobacter in piglets around weaning

    The restrictions on the use of antibiotic and anthelmintic treatments in organic pig farming necessitate alternative non-medical control strategies. Therefore, the antibiotic and parasite-reducing effect of a fructan-rich (prebiotic) diet of dried chicory was investigated in free-ranging piglets. Approximately half of 67 piglets from 9 litters were experimentally infected with Ascaris suum and Trichuris suis in the suckling period (1 to 7 weeks of age) and 58 of the piglets were challenged daily with E. coli O138:F8 for 9 days after weaning to induce weaning diarrhoea. The litters were fed either chicory (30% DM) or a control diet. The effect of chicory on intestinal helminths, intestinal microbiota, especially Bifidobacteria and Campylobacter spp., and E. coli post-weaning diarrhoea was assessed. The weight gain of the piglets was not impaired significantly by chicory. The intestinal A. suum worm burden was reduced by 64% (P=0.034) in the chicory-fed piglets, whereas these same piglets had 63% more T. suis worms (P=0.016). Feeding with chicory elicited no changes among the main bacterial groups in ileum according to terminal restriction fragment length polymorphism (T-RFLP) analysis. However, the terminal-restriction fragment (T-RF) 208 bp, which may belong to Lachnospiraceae, was stimulated by the chicory feed (P=0.03), and T-RF 370 bp that matches Enterobacter belonging to the Enterobacteria was reduced (P=0.004). Additionally, chicory increased the level of Bifidobacteria (P=0.001) and the faecal Campylobacter excretion level was transitorily reduced in chicory-fed piglets at 7 weeks of age (P=0.029). Unfortunately, it was not possible to assess the effect of chicory on post-weaning diarrhoea as it did not develop. In conclusion, feeding piglets chicory around the time of weaning caused complex changes of the microbiota and parasite communities within the intestinal tract, and feeding piglets chicory may therefore serve as an animal-friendly strategy to control pathogens
