    Tolerability of inhaled N-chlorotaurine in the pig model

    <p>Abstract</p> <p>Background</p> <p>N-chlorotaurine, a long-lived oxidant produced by human leukocytes, can be applied in human medicine as an endogenous antiseptic. Its antimicrobial activity can be enhanced by ammonium chloride. This study was designed to evaluate the tolerability of inhaled N-chlorotaurine (NCT) in the pig model.</p> <p>Methods</p> <p>Anesthetized pigs inhaled test solutions of 1% (55 mM) NCT (n = 7), 5% NCT (n = 6), or 1% NCT plus 1% ammonium chloride (NH<sub>4</sub>Cl) (n = 6), and 0.9% saline solution as a control (n = 7), respectively. Applications with 5 ml each were performed hourly within four hours. Lung function, haemodynamics, and pharmacokinetics were monitored. Bronchial lavage samples for captive bubble surfactometry and lung samples for histology and electron microscopy were removed.</p> <p>Results</p> <p>Arterial pressure of oxygen (PaO<sub>2</sub>) decreased significantly over the observation period of 4 hours in all animals. Compared to saline, 1% NCT + 1% NH<sub>4</sub>Cl led to significantly lower PaO<sub>2 </sub>values at the endpoint after 4 hours (62 ± 9.6 mmHg vs. 76 ± 9.2 mmHg, p = 0.014) with a corresponding increase in alveolo-arterial difference of oxygen partial pressure (AaDO<sub>2</sub>) (p = 0.004). Interestingly, AaDO<sub>2 </sub>was lowest with 1% NCT, even lower than with saline (p = 0.016). The increase of pulmonary artery pressure (PAP) over the observation period was smallest with 1% NCT without difference to controls (p = 0.91), and higher with 5% NCT (p = 0.02), and NCT + NH<sub>4</sub>Cl (p = 0.05).</p> <p>Histological and ultrastructural investigations revealed no differences between the test and control groups. The surfactant function remained intact. There was no systemic resorption of NCT detectable, and its local inactivation took place within 30 min. The concentration of NCT tolerated by A549 lung epithelial cells <it>in vitro </it>was similar to that known from other body cells (0.25–0.5 mM).</p> <p>Conclusion</p> <p>The endogenous antiseptic NCT was well tolerated at a concentration of 1% upon inhalation in the pig model. Addition of ammonium chloride in high concentration provokes a statistically significant impact on blood oxygenation.</p

    Analysis of primary T cell responses to intact and fractionated microbial pathogens

    Freshly isolated human T lymphocytes were tested for their response to mycobacteria, mycobacteriallysates, 2 dimensional (2D) PAGE separated mycobacteriallysates, leishmania and defined leishmanial antigen preparations. While,o T cells proliferated vigourously in the presence of mycobacteria and mycobacteria derived lysates, a significant stimulation from 2 D gel separated lysates was not detected. In addition '10 T cells failed to respond towards leishmania or leishmanial components. In the ab T cell compartment some donors, presumably according to their state of immunity against mycobacteria, responded to mycobacteria, mycobacterial lysates and 2 D gel separated mycobacterial lysates. Neither freshly isolated '10 T cells nor ab T cells from naive donors did mount a significant immune response against leishmania

    Agentic narcissism, communal narcissism, and prosociality

    Grandiose narcissism and prosociality are important topics in personality and social psychology, but research on their interplay is lacking. We present a first large-scale, systematic, and multimethod investigation linking the two. In 2 studies (N1 = 688, N2 = 336), we assessed grandiose narcissism comprehensively (i.e., agentic and communal narcissism) and examined its relations with instantiations of prosociality, namely, objective prosociality (actual behavior in Study 1; round-robin informant-reports in a real-life setting in Study 2) and subjective prosociality (self-perceptions in Studies 1 and 2). We obtained a consistent set of results. Agentic narcissism was related to lower objective prosociality and lower subjective prosociality. Communal narcissism, by contrast, was unrelated to objective prosociality, but was related to higher subjective prosociality. Additionally, we tested for prosociality self-enhancement among agentic and communal narcissists. Agentic narcissists evinced the same (and modest) level of prosociality self-enhancement as their non-narcissistic counterparts. Communal narcissists, by contrast, evinced substantial levels of prosociality self-enhancement, whereas their non-narcissistic counterparts did not enhance their prosociality at all.     Which leadership style do more narcissistic subordinates prefer in supervisors?

    Background and Objective: Subordinates in Western cultures generally prefer supervisors with a democratic rather than autocratic leadership style. It is unclear, however, whether more narcissistic subordinates share or challenge this prodemocratic default attitude. On the one hand, more narcissistic individuals strive for power and thus may favor a democratic supervisor, who grants them power through participation. On the other hand, similarity attracts and, thus, more narcissistic subordinates may favor an autocratic supervisor, who exhibits the same leadership style that they would adopt in a leadership position. Method: Four studies (N total = 1284) tested these competing hypotheses with two narcissism dimensions: admiration and rivalry. Participants indicated the leadership style they generally prefer in a supervisor (Study 1), rated their own supervisor's leadership style (Study 2a: individual ratings; Study 2b: team ratings), and evaluated profiles of democratic and autocratic supervisors (Study 3). Results: We found a significantly weaker prodemocratic default attitude among more narcissistic subordinates: Subordinates' narcissism was negatively related to endorsement of democratic supervisors and positively related to endorsement of autocratic supervisors. Those relations were mostly driven by narcissistic rivalry rather than narcissistic admiration. Conclusion: The results help clarify the narcissistic personality and, in particular, how more narcissistic subordinates prefer to be led.</p
