    Kupfer‐katalysierte Monooxygenierung von Phenolen: Evidenz für einen mononuklearen Reaktionsmechanismus

    Die CuI-Salze [Cu(CH3CN)4]PF und [Cu(oDFB)2]PF mit dem sehr schwach koordinierenden Anion Al(OC(CF3)3)4− (PF), sowie [Cu(NEt3)2]PF mit dem einzigartigen, linearen Bis-Triethylamin-Komplex [Cu(NEt3)2]+ wurden synthetisiert und als Katalysatoren für die Umwandlung von Monophenolen zu o-Chinonen untersucht. Die Aktivitäten dieser CuI-Salze bei der Monooxygenierung von 2,4-Di-tert-butylphenol (DTBP-H) wurden mit denen der [Cu(CH3CN)4]X-Salze mit “klassischen” Anionen (BF4−, OTf−, PF6−) verglichen, wobei ein Anioneneffekt auf die Aktivität des Katalysators und ein Ligandeneffekt auf die Reaktionsgeschwindigkeit festgestellt wurden. Letztere wird durch den Einsatz von CuII-Semichinon-Komplexen als Katalysatoren drastisch erhöht, was darauf hinweist, dass die Bildung eines CuII-Komplexes dem eigentlichen katalytischen Zyklus vorausgeht. Diese und andere experimentelle Erkenntnisse zeigen, dass die Oxygenierung von Monophenolen mit den oben genannten Systemen nicht einem dinuklearen, sondern einem mononuklearen Weg folgt, analog zur Topachinon-Cofaktor-Biosynthese im Enzym Aminoxidase

    Constitutive and fracture behavior of ultra-strong supercrystalline nanocomposites

    Supercrystalline nanocomposites are a new class of hybrid and nanostructured materials that can reach exceptional mechanical strength and can be fabricated at low temperatures. Hierarchically arranged, they bridge the gap from the nano- to the macro-scale. Even though their mechanical properties are starting to be characterized, their constitutive behavior is still largely unexplored. Here, the mechanical behavior of supercrystalline nanocomposites of iron oxide nanoparticles, surface-functionalized with oleic acid and oleyl phosphate ligands, is investigated in both bending and compression, with loading-unloading tests. A new bar geometry is implemented to better detect deformation prior to unstable crack propagation, and notched bending bars are tested to evaluate fracture toughness. Micro-mechanical tests result in the values of strength and elastic modulus that are extremely high for supercrystals, reaching record-high numbers in the oleic acid-based nanocomposites, which also show a significant tension-compression asymmetry. The constitutive behavior of both materials is predominantly linear elastic, with some more marked nonlinearities arising in the oleyl phosphate-based nanocomposites. The fracture toughness of both types of nanocomposites, ∼0.3 MPa√m, suggests that extrinsic toughening, associated with both material composition and nanostructure, plays an important role. Fractographic observations reveal analogies with shear and cleavage in atomic crystals. The influence of material composition, nanostructure, and processing method on the mechanical behavior of the nanocomposites is analyzed.</p

    Land use is a key factor to social wellbeing and has become a major component in political negotiations. This paper describes the mathematical structure of the European Forest and Agricultural Sector Optimization Model. The model represents simultaneously observed resource and technological heterogeneity, global commodity markets, and multiple environmental qualities. Land scarcity and land competition between traditional agriculture, forests, nature reserves, pastures, and bioenergy plantations is explicitly captured. Environmental change, technological progress, and policies can be investigated in parallel.     Осуществлен синтез бората алюминия на основе природных сырьевых материалов, изучено влияние добавки CaO на структуру и свойства материала. Было выявлено, что введение CaO в состав исходной сырьевой смеси способствует формированию игольчатых кристаллов и увеличению выхода фазы бората алюминия, а также снижению плотности материала. Показано, что двухстадийный обжиг позволяет повысить прочность образцов с добавкой CaO с сохранением низкой плотности

    Neurofilament as a blood marker for diagnosis and monitoring of primary progressive aphasias

    Objective: To assess the utility of serum neurofilament for diagnosis and monitoring of primary progressive aphasia (PPA) variants. Methods: We investigated neurofilament light chain (NF-L) levels in blood of 99 patients with PPA (40 with nonfluent variant PPA [nfvPPA], 38 with semantic variant PPA [svPPA], 21 with logopenic variant PPA [lvPPA]) and compared diagnostic performance with that reached by CSF NF-L, phosphorylated neurofilament heavy chain (pNF-H), b-amyloid (Ab(1)-42), tau, and phosphorylated tau. The longitudinal change of blood NF-L levels was measured and analyzed for correlation with functional decline and brain atrophy. Results: Serum NF-L is increased in PPA compared to controls and discriminates between nfvPPA/svPPA and lvPPA with 81% sensitivity and 67% specificity (cutoff 31 pg/mL). CSF NF-L, pNF-H, tau, phosphorylated tau, and Ab1-42 achieved similar performance, and pNF-H was the only marker for discrimination of nfvPPA from svPPA/lvPPA. In most patients with nfvPPA and svPPA, but not lvPPA, serum NF-L increased within follow-up. The increase correlated with functional decline and progression of atrophy of the left frontal lobe of all patients with PPAs and the right middle frontal gyrus of patients with nfvPPA and svPPA. Conclusions: Blood level of NF-L can aid the differential diagnosis of PPA variants, especially in combination with CSF pNF-H. Because serum NF-L correlates with functional decline and atrophy in the disease course, it qualifies as an objective disease status marker. Extended follow-up studies with cases of known neuropathology are imperative

    Serum neurofilament light chain in behavioral variant frontotemporal dementia

    Objective To determine the association of serum neurofilament light chain (NfL) with functional deterioration and brain atrophy during follow-up of patients with behavioral variant frontotemporal dementia (bvFTD). Methods Blood NfL levels from 74 patients with bvFTD, 26 with Alzheimer disease (AD), 17 with mild cognitive impairment (MCI), and 15 healthy controls (Con) at baseline and follow-up were determined and analyzed for the diagnostic potential in relation to functional assessment (Clinical Dementia Rating Scale Sum of Boxes [CDR-SOB], frontotemporal lobar degeneration-related CDR-SOB, Mini-Mental State Examination [MMSE]) and brain volumetry. Results At baseline, serum NfL level correlated with CSFNfL (bvFTD r = 0.706, p < 0.0001;AD/MCI r = 0.666, p = 0.0003). Highest serum levels were observed in bvFTD (p < 0 0.0001 vs Con and MCI, p = 0.0078 vs AD, respectively). Discrimination of bvFTD from Con/MCI/AD was possible with 91%/74%/74% sensitivity and 79%/74%/58% specificity. At follow-up, serum NfL increased in bvFTD and AD (p = 0.0039 and p = 0.0006, respectively). At baseline and follow-up, NfL correlated with functional scores of patients with bvFTD (e.g., CDR-SOB [baseline] r = 0.4157, p = 0.0006;[follow-up] r = 0.5629, p < 0.0001) and with atrophy in the gray and white matter of many brain regions including frontal and subcortical areas (e.g., frontal lobe: r = -0.5857, p < 0.0001;95% confidence interval -0.7415 to -0.3701). For patients with AD/MCI, NfL correlated with the functional performance as well (e.g., CDR-SOB [baseline] r = 0.6624, p < 0.0001;[follow-up] r = 0.5659, p = 0.0003) but not with regional brain volumes. Conclusions As serum NfL correlates with functional impairment and brain atrophy in bvFTD at different disease stages, we propose it as marker of disease severity, paving the way for its future use as outcome measure for clinical trials. Classification of evidence This study provides Class III evidence that for patients with cognitive problems, serum NfL concentration discriminates bvFTD from other forms of dementia

    Effectiveness of neonatal pulse oximetry screening for detection of critical congenital heart disease in daily clinical routine—results from a prospective multicenter study

    Pulse oximetry screening (POS) has been proposed as an effective, noninvasive, inexpensive tool allowing earlier diagnosis of critical congenital heart disease (cCHD). Our aim was to test the hypothesis that POS can reduce the diagnostic gap in cCHD in daily clinical routine in the setting of tertiary, secondary and primary care centres. We conducted a prospective multicenter trial in Saxony, Germany. POS was performed in healthy term and post-term newborns at the age of 24–72 h. If an oxygen saturation (SpO2) of ≤95% was measured on lower extremities and confirmed after 1 h, complete clinical examination and echocardiography were performed. POS was defined as false-negative when a diagnosis of cCHD was made after POS in the participating hospitals/at our centre. From July 2006–June 2008, 42,240 newborns from 34 institutions have been included. Seventy-two children were excluded due to prenatal diagnosis (n = 54) or clinical signs of cCHD (n = 18) before POS. Seven hundred ninety-five newborns did not receive POS, mainly due to early discharge after birth (n = 727; 91%). In 41,445 newborns, POS was performed. POS was true positive in 14, false positive in 40, true negative in 41,384 and false negative in four children (three had been excluded for violation of study protocol). Sensitivity, specificity, positive and negative predictive value were 77.78%, 99.90%, 25.93% and 99.99%, respectively. With POS as an adjunct to prenatal diagnosis, physical examination and clinical observation, the percentage of newborns with late diagnosis of cCHD was 4.4%. POS can substantially reduce the postnatal diagnostic gap in cCHD, and false-positive results leading to unnecessary examinations of healthy newborns are rare.     Different inflammatory signatures based on CSF biomarkers relate to preserved or diminished brain structure and cognition

    Neuroinflammation is a hallmark of Alzheimer's disease (AD) and both positive and negative associations of individual inflammation-related markers with brain structure and cognitive function have been described. We aimed to identify inflammatory signatures of CSF immune-related markers that relate to changes of brain structure and cognition across the clinical spectrum ranging from normal aging to AD. A panel of 16 inflammatory markers, A beta 42/40 and p-tau181 were measured in CSF at baseline in the DZNE DELCODE cohort (n = 295);a longitudinal observational study focusing on at-risk stages of AD. Volumetric maps of gray and white matter (GM/WM;n = 261) and white matter hyperintensities (WMHs, n = 249) were derived from baseline MRIs. Cognitive decline (n = 204) and the rate of change in GM volume was measured in subjects with at least 3 visits (n = 175). A principal component analysis on the CSF markers revealed four inflammatory components (PCs). Of these, the first component PC1 (highly loading on sTyro3, sAXL, sTREM2, YKL-40, and C1q) was associated with older age and higher p-tau levels, but with less pathological A beta when controlling for p-tau. PC2 (highly loading on CRP, IL-18, complement factor F/H and C4) was related to male gender, higher body mass index and greater vascular risk. PC1 levels, adjusted for AD markers, were related to higher GM and WM volumes, less WMHs, better baseline memory, and to slower atrophy rates in AD-related areas and less cognitive decline. In contrast, PC2 related to less GM and WM volumes and worse memory at baseline. Similar inflammatory signatures and associations were identified in the independent F.ACE cohort. Our data suggest that there are beneficial and detrimental signatures of inflammatory CSF biomarkers. While higher levels of TAM receptors (sTyro/sAXL) or sTREM2 might reflect a protective glia response to degeneration related to phagocytic clearance, other markers might rather reflect proinflammatory states that have detrimental impact on brain integrity
