    HYPERGLYCAEMIA impairs recovery from transient cerebral ischaemia: the importance of tissue acidification for this phenomenon has not been clarified in detail. We investigated this issue in a less complex in vitro preparation of isolated rat dorsal spinal roots exposed for 30 min to hyperglycaemic hypoxia. Peak height of compound action potentials recovered minimally in 5 mM bicarbonate. However, recovery was greatly improved by addition of the weak base trimethylamine during re-oxygenation. Addition of the weak acid propionate had no such effect. Cytoplasmic alkalinization improved recovery in a brief time window only: application of trimethylamine after 15 min of re-oxygenation was without beneficial effect. These data emphasize the importance of cytoplasmic acidification for neurophysiological recovery from hyper-glycaemic hypoxia during the initial period of re-oxygenation

    The effect of short-term vs. long-term soil moisture stress on the physiological response of three cocoa (Theobroma cacao L.) cultivars

    Understanding water stress signaling mechanisms and screening for tolerant cocoa cultivars are major challenges when facing prolonged dry and rainy seasons in cocoa-producing areas. While abscisic acid (ABA) and proline are supposed to enhance drought tolerance in cocoa, the role of polyamines remains unclear. The aim of this study was to investigate the biochemical response and phenological adaptation of cocoa (Theobroma cacao) on different soil moisture conditions, with a focus on short-term (20 days) and long-term (89 days) stress conditions, and to compare the performance of three cocoa cultivars. In a split plot design with four blocks, cocoa seedlings of an international high-yielding cultivar (TSH-565) and two locally selected cultivars (IIa-22 and III-06) from the drought-exposed Alto Beni region, Bolivia, were arranged in pots under a roof shelter (cultivar: three levels). The seedlings were exposed to strong (VERY DRY) and moderate (DRY) soil moisture deficits, water logging (WET) and regular irrigation (MOIST) that served as a control (moisture: four levels). We examined the growth performance and the levels of ABA, proline, and polyamines in the leaves. Growth was reduced already at a moderate drought, while severe drought enhanced seedling mortality. Severe drought increased the levels of ABA by 453% and of proline by 935%, inducing a short-term stress response; both compounds were degraded over the long-term period. The polyamine concentration was unrelated to soil moisture. The cocoa cultivars did not differ in their biochemical response to soil moisture stress (proline: p-value = 0.5, ABA: p-value = 0.3), but the local cultivar III-06 showed a stronger height growth increment than the international cultivar TSH-565 (237%, p-value = 0.002) under drought conditions

    Sequential anti-cytomegalovirus response monitoring may allow prediction of cytomegalovirus reactivation after allogeneic stem cell transplantation

    Background: Reconstitution of cytomegalovirus-specific CD3+CD8+ T cells (CMV-CTLs) after allogeneic hematopoietic stem cell transplantation (HSCT) is necessary to bring cytomegalovirus (CMV) reactivation under control. However, the parameters determining protective CMV-CTL reconstitution remain unclear to date. Design and Methods: In a prospective tri-center study, CMV-CTL reconstitution was analyzed in the peripheral blood from 278 patients during the year following HSCT using 7 commercially available tetrameric HLA-CMV epitope complexes. All patients included could be monitored with at least CMV-specific tetramer. Results: CMV-CTL reconstitution was detected in 198 patients (71%) after allogeneic HSCT. Most importantly, reconstitution with 1 CMV-CTL per µl blood between day +50 and day +75 post-HSCT discriminated between patients with and without CMV reactivation in the R+/D+ patient group, independent of the CMV-epitope recognized. In addition, CMV-CTLs expanded more daramtaically in patients experiencing only one CMV-reactivation than those without or those with multiple CMV reactivations. Monitoring using at least 2 tetramers was possible in 63% (n = 176) of the patients. The combinations of particular HLA molecules influenced the numbers of CMV-CTLs detected. The highest CMV-CTL count obtained for an individual tetramer also changed over time in 11% of these patients (n = 19) resulting in higher levels of HLA-B*0801 (IE-1) recognizing CMV-CTLs in 14 patients. Conclusions: Our results indicate that 1 CMV-CTL per µl blood between day +50 to +75 marks the beginning of an immune response against CMV in the R+/D+ group. Detection of CMV-CTL expansion thereafter indicates successful resolution of the CMV reactivation. Thus, sequential monitoring of CMV-CTL reconstitution can be used to predict patients at risk for recurrent CMV reactivation
