    Sensitive Question Techniques and Careless Responding: Adjusting the Crosswise Model for Random Answers

    The crosswise model is a popular sensitive question technique often considered more accurate than direct questioning. When this technique is used, a sensitive question is paired with a nonsensitive question that has a known prevalence and respondents are asked to give a joint answer to the pair of questions. Recent research has shown that prevalence estimates based on the crosswise model are biased towards 50% when respondents answer randomly, and that random answers are frequent. I develop methods to adjust the crosswise model for self-reported random answers. Results from an exploratory online survey (n = 103) show that (i) fewer respondents report random answers than might be expected given unadjusted results, (ii) results differ considerably between questions, and (iii) one of three questions yields an estimate that is substantially and significantly above the true value even after adjusting for random answers

    Comparing Continuous and Dichotomous Scoring of Social Desirability Scales: Effects of Different Scoring Methods on the Reliability and Validity of the Winkler-Kroh-Spiess BIDR Short Scale

    Survey researchers often include measures of social desirability in questionnaires. The Balanced Inventory of Desirable Responding (BIDR; Paulhus, 1991) is a widely used instrument that measures two components of socially desirable responding: self-deceptive enhancement (SDE) and impression management (IM). An open question is whether these scales should be scored dichotomously (counting only extreme values) or continuously (taking the mean of the answers). This paper compares the two methods with respect to test-retest reliability (stability) and internal consistency using a short German version of the BIDR (Winkler, Kroh, & Spiess, 2006). Tests of criterion validity are also presented. Data are taken from a post-stratified national probability sample of German family doctors (n = 166). All retest reliabilities exceed .70. No significant differences in test-retest reliability are found for the SDE subscale and the combined scale; the IM subscale attains significantly higher test-retest reliability when scored continuously. Internal consistency is significantly higher for the continuously scored scales in one of two Waves. Tests of criterion validity yield expected results. Overall, these results suggest that the short German scale is a valid measure of socially desirable responding and support the case for continuous rather than dichotomous scoring of BIDR scales

    Macro-level Research on Immigration and Crime in the Contemporary USA: Problems and Simple Solutions

    Extant research uses regression analysis with macro-level data to study the effect of immigration on crime in the contemporary USA. These studies have found mostly null or negative associations between the two variables. I point out three problems with these analyses. (i) Some studies use negative binomial regression inappropriately, in effect measuring determinants of frequencies rather than rates; (ii) all existing estimates arguably suffer from severe undercontrol, overcontrol, or both and cannot establish the direction of causality (if any) between immigration and crime; (iii) all studies present estimates that give equal weight to areas of differing population size. Taken together, these limitations render the research inconclusive. I show how to avoid these problems in a regression analysis of the effect of recent immigration on homicide rates in a sample of 91 US cities in 2000. Estimates point to a negative effect of immigration on homicide, but are not statistically significant

    Phenocopy – A Strategy to Qualify Chemical Compounds during Hit-to-Lead and/or Lead Optimization

    A phenocopy is defined as an environmentally induced phenotype of one individual which is identical to the genotype-determined phenotype of another individual. The phenocopy phenomenon has been translated to the drug discovery process as phenotypes produced by the treatment of biological systems with new chemical entities (NCE) may resemble environmentally induced phenotypic modifications. Various new chemical entities exerting inhibition of the kinase activity of Transforming Growth Factor β Receptor I (TGF-βR1) were qualified by high-throughput RNA expression profiling. This chemical genomics approach resulted in a precise time-dependent insight to the TGF-β biology and allowed furthermore a comprehensive analysis of each NCE's off-target effects. The evaluation of off-target effects by the phenocopy approach allows a more accurate and integrated view on optimized compounds, supplementing classical biological evaluation parameters such as potency and selectivity. It has therefore the potential to become a novel method for ranking compounds during various drug discovery phases

    ccdc80-l1 Is Involved in Axon Pathfinding of Zebrafish Motoneurons

    Axon pathfinding is a subfield of neural development by which neurons send out axons to reach the correct targets. In particular, motoneurons extend their axons toward skeletal muscles, leading to spontaneous motor activity. In this study, we identified the zebrafish Ccdc80 and Ccdc80-like1 (Ccdc80-l1) proteins in silico on the basis of their high aminoacidic sequence identity with the human CCDC80 (Coiled-Coil Domain Containing 80). We focused on ccdc80-l1 gene that is expressed in nervous and non-nervous tissues, in particular in territories correlated with axonal migration, such as adaxial cells and muscle pioneers. Loss of ccdc80-l1 in zebrafish embryos induced motility issues, although somitogenesis and myogenesis were not impaired. Our results strongly suggest that ccdc80-l1 is involved in axon guidance of primary and secondary motoneurons populations, but not in their proper formation. ccdc80-l1 has a differential role as regards the development of ventral and dorsal motoneurons, and this is consistent with the asymmetric distribution of the transcript. The axonal migration defects observed in ccdc80-l1 loss-of-function embryos are similar to the phenotype of several mutants with altered Hedgehog activity. Indeed, we reported that ccdc80-l1 expression is positively regulated by the Hedgehog pathway in adaxial cells and muscle pioneers. These findings strongly indicate ccdc80-l1 as a down-stream effector of the Hedgehog pathway

    Le programme de travail qui aboutit à ce livre s’inscrit dans le cadre du réseau d’excellence européen Ramses2, initié par la Maison méditerranéenne des sciences de l’homme. Une demi-douzaine de tables rondes ont réuni entre 2006 et 2008, d’un bout à l’autre de la Méditerranée (à Empúries, Aix-en-Provence, Palerme, Naples, Athènes), quelque soixante-dix chercheurs essentiellement français, italiens et espagnols, mais aussi anglais, grecs, bulgares, roumains, canadiens et russes. Il s’agissait d’étudier les rapports d’acculturation entre colons grecs et populations indigènes, en tenant compte des différences géographiques et chronologiques mais aussi de l’historiographie et des habitudes de recherche des diverses institutions. Les nombreuses communications qui ont jalonné les six tables rondes sont ici la plupart du temps précédées de textes introductifs. Une première partie, consacrée aux approches régionales, permet d’illustrer l’état de la recherche dans quelques régions choisies (autour d’Empuries, d’Himère, de Marseille, de Vélia, en Thrace et en mer Noire). La seconde partie, thématique, aborde un certain nombre de thèmes de recherche dans les régions précédentes, mais aussi dans d’autres régions du monde de la colonisation grecque. Le point de vue adopté dans ce livre est d’abord celui de la culture matérielle ; l’approche en est essentiellement archéologique. On se demandera par exemple quels sont les indices archéologiques qui permettent de dire si un site est habité par des Grecs, par des indigènes ou par une population “mixte”, et comment ces indices ont été appréciés selon les périodes et selon les régions. Beaucoup de communications présentent des synthèses régionales ou thématiques, mais une large place est faite également à des sites inédits, pour lesquels on n’a pas hésité à livrer une abondante documentation (plans, matériel de fouille). C’est en effet par le renouvellement de la documentation archéologique que nous pouvons espérer avancer dans la compréhension des rapports d’acculturation entre les colons grecs et les populations locales

    In Vivo Targeting of Alveolar Macrophages Via RAFT-Based Glycopolymers

    Targeting cell populations via endogenous carbohydrate receptors is an appealing approach for drug delivery. However, to be effective, this strategy requires the production of high affinity carbohydrate ligands capable of engaging with specific cell-surface lectins. To develop materials that exhibit high affinity towards these receptors, we synthesized glycopolymers displaying pendent carbohydrate moieties from carbohydrate-functionalized monomer precursors via reversible addition-fragmentation chain transfer (RAFT) polymerization. These glycopolymers were fluorescently labeled and used to determine macrophage-specific targeting both in vitro and in vivo. Mannose- and N-acetylglucosamine-containing glycopolymers were shown to specifically target mouse bone marrow-derived macrophages (BMDMs) in vitro in a dose-dependent manner as compared to a galactose-containing glycopolymer (30- and 19-fold higher uptake, respectively). In addition, upon macrophage differentiation, the mannose glycopolymer exhibited enhanced uptake in M2-polarized macrophages, an anti-inflammatory macrophage phenotype prevalent in injured tissue. This carbohydrate-specific uptake was retained in vivo, as alveolar macrophages demonstrated 6-fold higher internalization of mannose glycopolymer, as compared to galactose, following intratracheal administration in mice. We have shown the successful synthesis of a class of functional RAFT glycopolymers capable of macrophage-type specific uptake both in vitro and in vivo , with significant implications for the design of future targeted drug delivery systems
