In the course of German reunification the East German states, including the state of Saxony passed through a breathtaking transition process in all spheres of life. The major problem was the transformation of a comparatively inefficient industry-based and centrally governed economy into a modern market-based economy. The industrial state of Saxony recorded a tremendous number of factory closures and capacity reductions, leading to a rapid growth of unemployment in the first half of 1990s. This was a major reason for emigration especially of young and well educated persons. Hence Saxonyâs present and Âmid-term age structure appears quite unfavourable. The artificially-induced process of suburbanisation exacerbates the problems in the form of migration of wealthy families (valuable tax payers) from the core cities of agglomeration into the rural hinterland. However, admirable efforts to deal with the structural crises by using the traditional endogeneous potentials of the Saxonian regions can be recognized. Through governmental schemes new investors could be attracted, in particular in the prosperous fields of car production, engineering andâIT. The development of « industrial cores » also promotes the sector of medium and small-scale industry, though some lack of governmental assistance for these is apparent. Another sector gaining from the overall positive industrial development since the mid 1990âs is the service sector. Itâprovides more jobs than in the period before 1990. However, current efforts are not reflected in a significant change in the labour market. The inadequate support of largely mechanized industrial units is obviously not enough to tackle the basic problem of unemployment. In order to enhance Saxonyâs competitiveness at European and Global levels the state planning authorities and scientists follow the strategy of regional cooperation. Based on the city region « Sachsendreieck » the comparative regional advantages will be used in order to offer a wide range of services for the demands of national, continental and global markets. Despite serious problems Saxony has the potential to ensure, in the long run, a sustainable socio-economic development as the outcome of a painful transitional process during the past decade. To achieve this goal the joint actions of Saxonian citizens, the political class and the Saxonian entrepreneurs are required.
German reunification brought a complete alteration in the retail trade structures of the New LĂ€nder. This restructuring, forced by West German trading enterprises, led to a competitive situation between city centres and out-off town retailing. The aim of this contribution is to describe the urban development of Leipzig. Out-of-town retailing increased their sales areas until 1996, while city centres and traditional city districts were characterised by deficits. To counteract this imbalance a so-called « Stadtentwicklungsplan Zentren » is currently being brought into action as part of the urban planning guidelines. The aim of this programme is the embedding of centres within the urban boundary. Regeneration of the city centre as a main location of shopping is of prime importance. In order to attract retail capital to the city centre of Leipzig a project was initiated to bring the out-of-townâformat into the centre. One of the projects carried out by private investors is the implementation of a shopping mall in the central railway station of Leipzig which is still in use. This increase in attractiveness triggered department stores (a main and traditional attraction of German city centres) to locate their facilities in the city centre. As in Leipzig, the town centre development of Dresden and Chemnitz is based on inner city shopping malls and department stores. Recollection of traditions as well as new concepts for retail trade structures, to counteract the loss of city functions, are the new strategies of urban development in Leipzig, Dresden and Chemnitz. Since only a combination of these functions will transform city centres into places of urban life, the new strategies are not only concerned with retail trade but also with other functions of a city centre such as culture, education and leisure facilities.

Colon cancer cell-derived 12(S)-HETE induces the retraction of cancer-associated fibroblast via MLC2, RHO/ROCK and Ca2+ signalling
Retraction of mesenchymal stromal cells supports the invasion of colorectal cancer cells (CRC) into the adjacent compartment. CRC-secreted 12(S)-HETE enhances the retraction of cancer-associated fibroblasts (CAFs) and therefore, 12(S)-HETE may enforce invasivity of CRC. Understanding the mechanisms of metastatic CRC is crucial for successful intervention. Therefore, we studied pro-invasive contributions of stromal cells in physiologically relevant three-dimensional in vitro assays consisting of CRC spheroids, CAFs, extracellular matrix and endothelial cells, as well as in reductionist models. In order to elucidate how CAFs support CRC invasion, tumour spheroid-induced CAF retraction and free intracellular Ca2+ levels were measured and pharmacological-or siRNA-based inhibition of selected signalling cascades was performed. CRC spheroids caused the retraction of CAFs, generating entry gates in the adjacent surrogate stroma. The responsible trigger factor 12(S)-HETE provoked a signal, which was transduced by PLC, IP3, free intracellular Ca2+, Ca(2+)calmodulin-kinase-II, RHO/ROCK and MYLK which led to the activation of myosin light chain 2, and subsequent CAF mobility. RHO activity was observed downstream as well as upstream of Ca2+ release. Thus, Ca2+ signalling served as central signal amplifier. Treatment with the FDA-approved drugs carbamazepine, cinnarizine, nifedipine and bepridil HCl, which reportedly interfere with cellular calcium availability, inhibited CAF-retraction. The elucidation of signalling pathways and identification of approved inhibitory drugs warrant development of intervention strategies targeting tumour-stroma interaction
