Focusing a deterministic single-ion beam
We focus down an ion beam consisting of single 40Ca+ ions to a spot size of a
few mum using an einzel-lens. Starting from a segmented linear Paul trap, we
have implemented a procedure which allows us to deterministically load a
predetermined number of ions by using the potential shaping capabilities of our
segmented ion trap. For single-ion loading, an efficiency of 96.7(7)% has been
achieved. These ions are then deterministically extracted out of the trap and
focused down to a 1sigma-spot radius of (4.6 \pm 1.3)mum at a distance of 257mm
from the trap center. Compared to former measurements without ion optics, the
einzel-lens is focusing down the single-ion beam by a factor of 12. Due to the
small beam divergence and narrow velocity distribution of our ion source,
chromatic and spherical aberration at the einzel-lens is vastly reduced,
presenting a promising starting point for focusing single ions on their way to

The Rashba Hamiltonian and electron transport
The Rashba Hamiltonian describes the splitting of the conduction band as a
result of spin-orbit coupling in the presence of an external field and is
commonly used to model the electronic structure of confined narrow-gap
semiconductors. Due to the mixing of spin states some care has to be exercised
in the calculation of transport properties. We derive the velocity operator for
the Rashba-split conduction band and demonstrate that the transmission of an
interface between a ferromagnet and a Rashba-split semiconductor does not
depend on the magnetization direction, in contrast with previous assertions in

Cytopathicity of Chlamydia is largely reproduced by expression of a single chlamydial protease
Chlamydiae replicate in a vacuole within epithelial cells and commonly induce cell damage and a deleterious inflammatory response of unknown molecular pathogenesis. The chlamydial protease-like activity factor (CPAF) translocates from the vacuole to the cytosol, where it cleaves several cellular proteins. CPAF is synthesized as an inactive precursor that is processed and activated during infection. Here, we show that CPAF can be activated in uninfected cells by experimentally induced oligomerization, reminiscent of the activation mode of initiator caspases. CPAF activity induces proteolysis of cellular substrates including two novel targets, cyclin B1 and PARP, and indirectly results in the processing of pro-apoptotic BH3-only proteins. CPAF activation induces striking morphological changes in the cell and, later, cell death. Biochemical and ultrastructural analysis of the cell death pathway identify the mechanism of cell death as nonapoptotic. Active CPAF in uninfected human cells thus mimics many features of chlamydial infection, implicating CPAF as a major factor of chlamydial pathogenicity, Chlamydia-associated cell damage, and inflammation
Increased plasma zonulin in patients with sepsis
Introduction: Zonulin is a eukaryotic protein structurally similar to Vibrio choleraeâs zonula occludens toxin. It plays an important role in the opening of small intestine tight junctions. The loss of gut wall integrity during sepsis might be pivotal and has been described in various experimental as well as human studies. Increased levels of zonulin could be demonstrated in diseases associated with increased intestinal inflammation, such as celiac disease and type 1 diabetes. We therefore investigated the role of plasma levels of zonulin in patients with sepsis as a non-invasive marker of gut wall integrity.
Materials and methods: Plasma level of zonulin was measured in 25 patients with sepsis, severe sepsis or septic shock according to ACCP/SCCM criteria at the first day of diagnosed sepsis. 18 non-septic post-surgical ICU-patients and 20 healthy volunteers served as control. Plasma levels were determined by using commercially available ELISA kit. Data are given as median and interquartile range (IQR).
Results: Significantly higher plasma concentration of zonulin were found in the sepsis group: 6.61 ng/mL (IQR 3.51-9.46), as compared to the to the post-surgical control group: 3.40 ng/mL (IQR 2.14-5.70) (P = 0.025), as well as to the healthy group: 3.55 ng/mL (IQR 3.14-4.14) (P = 0.008).
Conclusion: We were able demonstrate elevated levels of plasma zonulin, a potential marker of intestinal permeability in septic patients. Increased zonulin may serve as an additional mechanism for the observed increased intestinal permeability during sepsis and SIRS
Structure and Function of the Transmembrane Domain of NsaS, an Antibiotic Sensing Histidine Kinase in Staphylococcus aureus
NsaS is one of four intramembrane histidine kinases (HKs) in Staphylococcus aureus that mediate the pathogen's response to membrane active antimicrobials and human innate immunity. We describe the first integrative structural study of NsaS using a combination of solution state NMR spectroscopy, chemical-cross-linking, molecular modeling and dynamics. Three key structural features emerge: First, NsaS has a short N-terminal amphiphilic helix that anchors its transmembrane (TM) bundle into the inner leaflet of the membrane such that it might sense neighboring proteins or membrane deformations. Second, the transmembrane domain of NsaS is a 4-helix bundle with significant dynamics and structural deformations at the membrane interface. Third, the intracellular linker connecting the TM domain to the cytoplasmic catalytic domains of NsaS is a marginally stable helical dimer, with one state likely to be a coiled-coil. Data from chemical shifts, heteronuclear NOE, H/D exchange measurements and molecular modeling suggest that this linker might adopt different conformations during antibiotic induced signaling
Saturated Ferromagnetism and Magnetization Deficit in Optimally Annealed (Ga,Mn)As Epilayers
We examine the Mn concentration dependence of the electronic and magnetic
properties of optimally annealed Ga1-xMnxAs epilayers for 1.35% < x < 8.3%. The
Curie temperature (Tc), conductivity, and exchange energy increase with Mn
concentration up to x ~ 0.05, but are almost constant for larger x, with Tc ~
110 K. The ferromagnetic moment per Mn ion decreases monotonically with
increasing x, implying that an increasing fraction of the Mn spins do not
participate in the ferromagnetism. By contrast, the derived domain wall
thickness, an important parameter for device design, remains surprisingly

Levothyroxine Treatment of Euthyroid Children with Autoimmune Hashimoto Thyroiditis: Results of a Multicenter, Randomized, Controlled Trial
Background:
Levothyroxine (L-T4) treatment of euthyroid children with Hashimoto thyroiditis (HT) is a controversial issue.
Patients and Methods:
We conducted a prospective, randomized, controlled clinical trial. Out of 79 identified euthyroid patients, 59 started the study; 25 patients (21 female, 4 male; age: 11.8 ± 2.3 years) received L-T4 at a mean dose of 1.6 ”g/kg (SD, 0.8) daily, and 34 (27 female, 7 male; age: 12.6 ± 1.2 years) were not treated. Patients developing subclinical hypothyroidism during follow-up (n = 13) were treated with L-T4 and removed from the observation group. As the main outcome measures, thyroid gland volume (determined by ultrasound) as well as serum levels of TSH, free T4, and antibodies against thyroid peroxidase and thyroglobulin were assessed every 6 months for 36 months.
Results:
At the start, the mean thyroid volume (standard deviation score, SDS) was 2.5 in the treatment group and 1.6 in the observation group. There was a constant decline in mean thyroid volume (SDS) from 2.13 (month 12) to 1.12 (month 30) in the treated group, with a delta thyroid volume of -1.01 SDS. In the observation group, the mean delta thyroid volume increased to +0.27 SDS. The change of the delta thyroid volume was statistically significantly different between both groups during the 12- and 30-month time points (p < 0.05). L-T4 had no effect on thyroid function and serum thyroid antibodies.
Conclusions:
L-T4 treatment can decrease the thyroid volume in euthyroid children with HT, but the effect is limited to a definite time period
