    Applicability of Different Bio-based Polymers for Wiring Boards

    The paper gives a review of experiments for the application of biodegradable, sustainable polymers as a wiring board material. In the paper two different biobased materials and the standard PCB materials FR4 and FR2 were compared. The investigations refer to mechanical, electrical performance, surface quality the flammability and applicability for Polymer Thick Film Technology (PTFT) of the biobased material. The biobased materials are polylactic acid (PLA) mixed with different contents of cellulose acetate (CA) or flame retardant zinc pyrophosphate (ZnPP) and polyurethane (PU) with CA. The results show that the addition of CA to the polymers leads to a change of different properties for PLA and to a significant change in mechanical properties for PU. The use of ZnPP in PLA shows major improvements regarding the flammability of the polymer. The properties of the samples are in the same order of magnitude as the properties of FR2

    Traditional Use of Herbal Remedies in Livestock by Farmers in 3 Swiss Cantons (Aargau, Zurich, Schaffhausen)

    Background: This study investigated the extent of traditional knowledge and use of homemade herbal remedies for livestock by farmers in 3 Swiss cantons (Aargau, Zurich, Schaffhausen). The study focused on organic farms. Methods: At 21 farms, 24 farmers aged 36–83 years were interviewed with a semi-structured, detailed questionnaire. For each homemade herbal remedy, the plant species, mode of preparation, source of knowledge, and application were gathered. Satisfaction of the farmers with the application was estimated with the aid of a visual analogue scale. Results: Information on a total of 165 homemade remedies was collected of which 123 contained a single plant species only (homemade mono-species herbal remedies, HMHR). The 123 HMHR were selected for this paper. They corresponded to 150 different applications and originated from 43 plant species from 30 families. Plants belonging to the families of Asteraceae, Lamiaceae, and Apiaceae were used most frequently. The single most applied species were Matricaria recutita L., Calendula officinalis L., Symphytum officinale L., and Coffea arabica L. For each formulation, 1–4 different applications were mentioned, most of them for cattle. The main applications were skin alterations and sores, gastrointestinal and metabolic diseases as well as infertility and diseases of the female genitalia. Approximately half of the applications were used during the last 12 months prior to the interview. Conclusion: This study shows that HMHR are used by Swiss farmers for the treatment of different livestock diseases. In general, the farmers were satisfied with the outcome of the applications

    Erioflorin stabilizes the tumor suppressor Pdcd4 by inhibiting its interaction with the E3-ligase β-TrCP1

    Loss of the tumor suppressor Pdcd4 was reported for various tumor entities and proposed as a prognostic marker in tumorigenesis. We previously characterized decreased Pdcd4 protein stability in response to mitogenic stimuli, which resulted from p70S6K1-dependent protein phosphorylation, β-TrCP1-mediated ubiquitination, and proteasomal destruction. Following high-throughput screening of natural product extract libraries using a luciferase-based reporter assay to monitor phosphorylation-dependent proteasomal degradation of the tumor suppressor Pdcd4, we succeeded in showing that a crude extract from Eriophyllum lanatum stabilized Pdcd4 from TPA-induced degradation. Erioflorin was identified as the active component and inhibited not only degradation of the Pdcd4-luciferase-based reporter but also of endogenous Pdcd4 at low micromolar concentrations. Mechanistically, erioflorin interfered with the interaction between the E3-ubiquitin ligase β-TrCP1 and Pdcd4 in cell culture and in in vitro binding assays, consequently decreasing ubiquitination and degradation of Pdcd4. Interestingly, while erioflorin stabilized additional β-TrCP-targets (such as IκBα and β-catenin), it did not prevent the degradation of targets of other E3-ubiquitin ligases such as p21 (a Skp2-target) and HIF-1α (a pVHL-target), implying selectivity for β-TrCP. Moreover, erioflorin inhibited the tumor-associated activity of known Pdcd4- and IκBα-regulated αtranscription factors, that is, AP-1 and NF-κB, altered cell cycle progression and suppressed proliferation of various cancer cell lines. Our studies succeeded in identifying erioflorin as a novel Pdcd4 stabilizer that inhibits the interaction of Pdcd4 with the E3-ubiquitin ligase β-TrCP1. Inhibition of E3-ligase/target-protein interactions may offer the possibility to target degradation of specific proteins only as compared to general proteasome inhibition

    Automictic Reproduction in Interspecific Hybrids of Poeciliid Fish

    SummaryAutomixis, the process whereby the fusion of meiotic products restores the diploid state of the egg, is a common mode of reproduction in plants but has also been described in invertebrate animals [1, 2]. In vertebrates, however, automixis has so far only been discussed as one of several explanations for isolated cases of facultative parthenogenesis [3, 4]. Analyzing oocyte formation in F1 hybrids derived from Poecilia mexicana limantouri and P. latipinna crosses (the cross that led to the formation of the gynogenetic Poecilia formosa[5, 6]), we found molecular evidence for automictic oocyte production [7]. The mechanism involves the random fusion of meiotic products after the second meiotic division. The fertilization of diploid oocytes gives rise to fully viable triploid offspring. Although the automictic production of diploid oocytes as seen in these F1 hybrids clearly represents a preadaptation to parthenogenetic reproduction [8], it is also a powerful intrinsic postzygotic isolation mechanism because the resulting next generation triploids were always sterile. The mechanism described here can explain facultative parthenogenesis [9], as well as varying ploidy levels reported in different animal groups [10]. Most importantly, at least some of the reported cases of triploidy in humans [11] can now be traced back to automixis

    The Reverse Transcription Signature of N-\u3csub\u3e1\u3c/sub\u3e-Methyladenosine in RNA-Seq is Sequence Dependent

    The combination of Reverse Transcription (RT) and high-throughput sequencing has emerged as a powerful combination to detect modified nucleotides in RNA via analysis of either abortive RT-products or of the incorporation of mismatched dNTPs into cDNA. Here we simultaneously analyze both parameters in detail with respect to the occurrence of N-1-methyladenosine (m1A) in the template RNA. This naturally occurring modification is associated with structural effects, but it is also known as a mediator of antibiotic resistance in ribosomal RNA. In structural probing experiments with dimethylsulfate, m1A is routinely detected by RT-arrest. A specifically developed RNA-Seq protocol was tailored to the simultaneous analysis of RT-arrest and misincorporation patterns. By application to a variety of native and synthetic RNA preparations, we found a characteristic signature of m1A, which, in addition to an arrest rate, features misincorporation as a significant component. Detailed analysis suggests that the signature depends on RNA structure and on the nature of the nucleotide 3’ of m1A in the template RNA, meaning it is sequence dependent.
