    Second-order nonlinearities and crystal structure of 2-methoxy-4'-nitro-(E)-stilbene

    Second-order nonlinear optical (NLO) properties of crystalline materials depend both on the magnitude of the molecular hyperpolarizability (β) and on the orientation of the chromophores in the crystal lattice. If the molecular hyperpolarizability is to result in a nonzero macroscopic nonlinearity, the molecule must crystallize in a noncentrosymmetric space group. The design of new second-order NLO materials requires one both to address the structure-property relationships that govern β and to develop methodologies that favor noncentrosymmetric crystallization. Methods used to encourage noncentrosymmetric crystallization include the use of chiral molecules, the incorporation of functional groups that encourage asymmetric intermolecular hydrogen bonding, the synthesis of molecules with very small ground-state dipole moments but larger excited-state dipole moments, and the variation of counterion with ionic chromophore

    Mid-infrared Period-Luminosity Relations of RR Lyrae Stars Derived from the WISE Preliminary Data Release

    Interstellar dust presents a significant challenge to extending parallax-determined distances of optically observed pulsational variables to larger volumes. Distance ladder work at mid-infrared wavebands, where dust effects are negligible and metallicity correlations are minimized, have been largely focused on few-epoch Cepheid studies. Here we present the first determination of mid-infrared period-luminosity (PL) relations of RR Lyrae stars from phase-resolved imaging using the preliminary data release of the Wide-Field Infrared Survey Explorer (WISE). We present a novel statistical framework to predict posterior distances of 76 well-observed RR Lyrae that uses the optically constructed prior distance moduli while simultaneously imposing a power-law PL relation to WISE-determined mean magnitudes. We find that the absolute magnitude in the bluest WISE filter is M_W1 = (-0.421+-0.014) - (1.681+-0.147)*log(P/0.50118 day), with no evidence for a correlation with metallicity. Combining the results from the three bluest WISE filters, we find that a typical star in our sample has a distance measurement uncertainty of 0.97% (statistical) plus 1.17% (systematic). We do not fundamentalize the periods of RRc stars to improve their fit to the relations. Taking the Hipparcos-derived mean V-band magnitudes, we use the distance posteriors to determine a new optical metallicity-luminosity relation which we present in Section 5. The results of this analysis will soon be tested by HST parallax measurements and, eventually, with the Gaia astrometric mission.Comment: 33 pages, 12 figures, 2 tables.     An interpretative phenomenological analysis of posttraumatic growth in adults bereaved by suicide

    This study explored experiences of posttraumatic growth in adults bereaved by suicide. Six participants were interviewed using a semi-structured interview schedule. Transcribed interviews were analyzed from an interpretative phenomenological framework. Two superordinate themes, with three ordinate themes in each, were identified: (a) positive growth (“life view,” “knowledge of self,” and “relation to others”) and (b) social context (“gaze of others,” “public guise,” and “solace of other survivors”). Suicide survivors gain extra insights due to their experiences, but are reluctant to acknowledge that they do. This requires consideration in theoretical and clinical setting

    Constraints on the Progenitor System of the Type Ia Supernova SN 2011fe/PTF11kly

    Type Ia supernovae (SNe) serve as a fundamental pillar of modern cosmology, owing to their large luminosity and a well-defined relationship between light-curve shape and peak brightness. The precision distance measurements enabled by SNe Ia first revealed the accelerating expansion of the universe, now widely believed (though hardly understood) to require the presence of a mysterious "dark" energy. General consensus holds that Type Ia SNe result from thermonuclear explosions of a white dwarf (WD) in a binary system; however, little is known of the precise nature of the companion star and the physical properties of the progenitor system. Here we make use of extensive historical imaging obtained at the location of SN 2011fe/PTF11kly, the closest SN Ia discovered in the digital imaging era, to constrain the visible-light luminosity of the progenitor to be 10-100 times fainter than previous limits on other SN Ia progenitors. This directly rules out luminous red giants and the vast majority of helium stars as the mass-donating companion to the exploding white dwarf. Any evolved red companion must have been born with mass less than 3.5 times the mass of the Sun. These observations favour a scenario where the exploding WD of SN 2011fe/PTF11kly, accreted matter either from another WD, or by Roche-lobe overflow from a subgiant or main-sequence companion star.Comment: 22 pages, 6 figures, submitte

    Pharmacokinetics and pharmacodynamics of clofazimine for treatment of cryptosporidiosis

    Infection with Cryptosporidium spp. can cause severe diarrhea leading to long-term adverse impacts and even death in malnourished children and immunocompromised patients. The only FDA-approved drug for treating cryptosporidiosis, nitazoxanide, has limited efficacy in the populations impacted the most by the diarrheal disease, and safe, effective treatment options are urgently needed. Initially identified by a large-scale phenotypic screening campaign, the antimycobacterial therapeutic clofazimine demonstrated great promise in both in vitro and in vivo preclinical models of Cryptosporidium infection. Unfortunately, a Phase 2a clinical trial in HIV infected adults with cryptosporidiosis did not identify any clofazimine treatment effect on Cryptosporidium infection burden or clinical outcomes. To explore whether clofazimine's lack of efficacy in the Phase 2a trial may have been due to subtherapeutic clofazimine concentrations, a pharmacokinetic/pharmacodynamic modeling approach was undertaken to determine the relationship between clofazimine in vivo concentrations and treatment effects in multiple preclinical infection models. Exposure-response relationships were characterized using Emax and logistic models which allowed predictions of efficacious clofazimine concentrations for the control and reduction of disease burden. After establishing exposure-response relationships for clofazimine treatment of Cryptosporidium infection in our preclinical model studies, it was unmistakable that the clofazimine levels observed in the Phase 2a study participants were well below concentrations associated with anti-Cryptosporidium efficacy. Thus, despite a dosing regimen above the highest doses recommended for mycobacterial therapy, it is very likely the lack of treatment effect in the Phase 2a trial was at least partially due to clofazimine concentrations below those required for efficacy against cryptosporidiosis. It is unlikely that clofazimine will provide a remedy for the large number of cryptosporidiosis patients currently without a viable treatment option unless alternative, safe clofazimine formulations with improved oral absorption are developed

    Oncogenic ERBB3 Mutations in Human Cancers

    SummaryThe human epidermal growth factor receptor (HER) family of tyrosine kinases is deregulated in multiple cancers either through amplification, overexpression, or mutation. ERBB3/HER3, the only member with an impaired kinase domain, although amplified or overexpressed in some cancers, has not been reported to carry oncogenic mutations. Here, we report the identification of ERBB3 somatic mutations in ∼11% of colon and gastric cancers. We found that the ERBB3 mutants transformed colonic and breast epithelial cells in a ligand-independent manner. However, the mutant ERBB3 oncogenic activity was dependent on kinase-active ERBB2. Furthermore, we found that anti-ERBB antibodies and small molecule inhibitors effectively blocked mutant ERBB3-mediated oncogenic signaling and disease progression in vivo
