Fatigue Assessment of Moorings for Floating Offshore Wind Turbines by Advanced Spectral Analysis Methods

The fatigue assessment of mooring lines for floating offshore wind turbines represents a challenging issue not only for the reliable design of the stationkeeping system but also for the economic impact on the installation and maintenance costs over the entire lifetime of the offshore wind farm. After a brief review about the state-of-art, the nonlinear time-domain hydrodynamic model of floating offshore wind turbines moored by chain cables is discussed. Subsequently, the assessment of the fatigue damage in the mooring lines is outlined, focusing on the combined-spectrum approach. The relevant fatigue parameters, due to the low-and wave-frequency components of the stress process, are estimated by two different methods. The former is based on the time-domain analysis of the filtered stress process time history. The latter, instead, is based on the spectral analysis of the stress process by two advanced methods, namely the Welch and Thomson ones. Subsequently, a benchmark study is performed, assuming as reference floating offshore wind turbine the OC4-DeepCWind semisubmersible platform, equipped with the 5 MW NREL wind turbine. The cumulative fatigue damage is determined for eight load conditions, including both power production and parked wind turbine situations. A comparative analysis between time-domain and spectral analysis methods is also performed. Current results clearly show that the endorsement of advanced spectral analysis methods can be helpful to improve the reliability of the fatigue life assessment of mooring lines

Underwater photogrammetry for 3D modeling of floating objects: The case study of a 19-foot motor boat

3D modeling of floating or semi-submerged objects is a challenging and attractive task for the marine industry especially if the manufacturing of components that have to be replaced or repaired after a damage is necessary or the ship itself has to be converted. Up to now the 3D reverse engineering of ships has required docking operations to carry out a geodetic or photogrammetric survey with high costs for shipowners. In this paper an innovative 3D acquisition method for digital recording of floating objects is presented. The method is based on digital photogrammetry both underwater and terrestrial. Preliminary tests are presented for the case study of a 19-foot motor boat. Two surveys of the boat in floating conditions are carried out and then joined by means of special rigid orientation devices built ad hoc

Citrus allergy from pollen to clinical symptoms

Allergy to citrus fruits is often associated with pollinosis and sensitization to other plants due to a phenomenon of cross-reactivity. The aims of the present study were to highlight the cross-reactivity among citrus and the major allergenic pollens/fruits, throughout clinical and molecular investigations, and to evaluate the sensitization frequency to citrus fruits in a population of children and adults with pollinosis. We found a relevant percentage of sensitisation (39%) to citrus fruits in the patients recruited and in all of them the IgE-mediated mechanism has been confirmed by the positive response to the prick-to-prick test. RT-PCR experiments showed the expression of Cit s 1, Cit s 3 and a profilin isoform, already described in apple, also in Citrus clementine pollen. Data of multiple sequence alignments demonstrated that Citrus allergens shared high percentage identity values with other clinically relevant species (i.e. Triticum aestivum, Malus domestica), confirming the possible cross-allergenicity citrus/grasses and citrus/apple. Finally, a novelty of the present work has been the expression of two phospholipaseA2 isoforms (PLA2 \u3b1 and \u3b2) in Citrus as well as in Triticum pollens; being PLA2 able to generate pro-inflammatory factors, this enzyme could participate in the activation of the allergenic inflammatory cascade

CXCR3 identifies human naive CD8+ T cells with enhanced effector differentiation potential

In mice, the ability of naive T (TN) cells to mount an effector response correlates with TCR sensitivity for self-derived Ags, which can be quantified indirectly by measuring surface expression levels of CD5. Equivalent findings have not been reported previously in humans. We identified two discrete subsets of human CD8+ TN cells, defined by the absence or presence of the chemokine receptor CXCR3. The more abundant CXCR3+ TN cell subset displayed an effector-like transcriptional profile and expressed TCRs with physicochemical characteristics indicative of enhanced interactions with peptide-HLA class I Ags.Moreover, CXCR3+ TN cells frequently produced IL-2 and TNF in response to nonspecific activation directly ex vivo and differentiated readily into Ag-specific effector cells in vitro. Comparative analyses further revealed that human CXCR3+ TN cells were transcriptionally equivalent to murine CXCR3+ TN cells, which expressed high levels of CD5. These findings provide support for the notion that effector differentiation is shaped by heterogeneity in the preimmune repertoire of human CD8+ T cells
