    Practice What You Preach: California\u27s Obligation to Give Full Faith and Credit to the Vermont Civil Union

    This Note evaluates the inter-jurisdictional effect of the Vermont Civil Union statute. A same-sex couple who enters into a civil union in Vermont currently enjoys all of the Vermont state benefits typically conferred only on married couples. The Federal Defense of Marriage Act ( DOMA ) removes the burden on states of having to afford full faith and credit to same-sex marriages. As such, this Note discusses whether the DOMA is applicable to the Vermont Civil Union in addition to same-sex marriages. Furthermore, this Note discusses whether California, if not obliged federally under the DOMA to recognize the civil union, is nonetheless obliged by its own state policy to recognize the rights conferred to parties who have entered into a civil union in Vermont. California might be considered fertile ground for this kind of inquiry because it also has a legislated domestic partnership statute that affords same-sex couples in California many of the benefits that the Vermont civil union provides to Vermont\u27s same-sex couples. This Note takes the position that in a case of first impression, a California court would have to recognize the rights conferred on a couple who went to Vermont to enter into a civil union

    Robustness to misalignment of low-cost, compact quantitative phase imaging architectures.

    Non-interferometric approaches to quantitative phase imaging could enable its application in low-cost, miniaturised settings such as capsule endoscopy. We present two possible architectures and both analyse and mitigate the effect of sensor misalignment on phase imaging performance. This is a crucial step towards determining the feasibility of implementing phase imaging in a capsule device. First, we investigate a design based on a folded 4f correlator, both in simulation and experimentally. We demonstrate a novel technique for identifying and compensating for axial misalignment and explore the limits of the approach. Next, we explore the implications of axial and transverse misalignment, and of manufacturing variations on the performance of a phase plate-based architecture, identifying a clear trade-off between phase plate resolution and algorithm convergence time. We conclude that while the phase plate architecture is more robust to misalignment, both architectures merit further development with the goal of realising a low-cost, compact system for applying phase imaging in capsule endoscopy

    Photochemical Methods for Peptide Macrocylization

    Photochemical reactions have been the subject of renewed interest over the last two decades, leading to the development of many new diverse and powerful chemical transformations. More recently, these developments have been expanded to enable the photochemical macrocylization of peptides and small proteins. These constructs benefit from increased stability, structural rigidity, and biological potency over their linear counterparts, providing opportunities for improved therapeutic agents. In this review, we provide an overview of both the established and emerging methods for photochemical peptide macrocyclization, highlighting both the limitations and opportunities for further innovation in the field

    A Public–Industry Partnership for Enhancing Corn Nitrogen Research and Datasets: Project Description, Methodology, and Outcomes

    Due to economic and environmental consequences of N lost from fertilizer applications in corn (Zea mays L.), considerable public and industry attention has been devoted to the development of N decision tools. Needed are research and databases and associated metadata, at numerous locations and years to represent a wide geographic range of soil and weather scenarios, for evaluating tool performance. The goals of this research were to conduct standardized corn N rate response field studies to evaluate the performance of multiple public-domain N decision tools across diverse soils and environmental conditions, develop and publish new agronomic science for improved crop N management, and train new scientists. The geographic scope, scale, and unique collaborative arrangement warrant documenting details of this research. The objectives of this paper are to describe how the research was undertaken, reasons for the methods, and the project’s anticipated value. The project was initiated in a partnership between eight U.S. Midwest land-grant universities, USDA-ARS, and DuPont Pioneer. Research using a standardized protocol was conducted over the 2014 through 2016 growing seasons, yielding a total of 49 sites. Preliminary observations of soil and crop variables measured from each site revealed a magnitude of differences in soil properties (e.g., texture and organic matter) as well as differences in agronomic and economic responses to applied N. The project has generated a valuable dataset across a wide array of weather and soils that allows investigators to perform robust evaluation of N use in corn and N decision tools

    Effects of microbiota-directed foods in gnotobiotic animals and undernourished children

    To examine the contributions of impaired gut microbial community development to childhood undernutrition, we combined metabolomic and proteomic analyses of plasma samples with metagenomic analyses of fecal samples to characterize the biological state of Bangladeshi children with severe acute malnutrition (SAM) as they transitioned, after standard treatment, to moderate acute malnutrition (MAM) with persistent microbiota immaturity. Host and microbial effects of microbiota-directed complementary food (MDCF) prototypes targeting weaning-phase bacterial taxa underrepresented in SAM and MAM microbiota were characterized in gnotobiotic mice and gnotobiotic piglets colonized with age- and growth-discriminatory bacteria. A randomized, double-blind controlled feeding study identified a lead MDCF that changes the abundances of targeted bacteria and increases plasma biomarkers and mediators of growth, bone formation, neurodevelopment, and immune function in children with MAM

    Rhesus TRIM5α disrupts the HIV-1 capsid at the inter-hexamer interfaces

    TRIM proteins play important roles in the innate immune defense against retroviral infection, including human immunodeficiency virus type-1 (HIV-1). Rhesus macaque TRIM5α (TRIM5αrh) targets the HIV-1 capsid and blocks infection at an early post-entry stage, prior to reverse transcription. Studies have shown that binding of TRIM5α to the assembled capsid is essential for restriction and requires the coiled-coil and B30.2/SPRY domains, but the molecular mechanism of restriction is not fully understood. In this study, we investigated, by cryoEM combined with mutagenesis and chemical cross-linking, the direct interactions between HIV-1 capsid protein (CA) assemblies and purified TRIM5αrh containing coiled-coil and SPRY domains (CC-SPRYrh). Concentration-dependent binding of CC-SPRYrh to CA assemblies was observed, while under equivalent conditions the human protein did not bind. Importantly, CC-SPRYrh, but not its human counterpart, disrupted CA tubes in a non-random fashion, releasing fragments of protofilaments consisting of CA hexamers without dissociation into monomers. Furthermore, such structural destruction was prevented by inter-hexamer crosslinking using P207C/T216C mutant CA with disulfide bonds at the CTD-CTD trimer interface of capsid assemblies, but not by intra-hexamer crosslinking via A14C/E45C at the NTD-NTD interface. The same disruption effect by TRIM5αrh on the inter-hexamer interfaces also occurred with purified intact HIV-1 cores. These results provide insights concerning how TRIM5α disrupts the virion core and demonstrate that structural damage of the viral capsid by TRIM5α is likely one of the important components of the mechanism of TRIM5α-mediated HIV-1 restriction. © 2011 Zhao et al

    The experience of providing young people attending general practice with an online risk assessment tool to assess their own sexual health risk

    <p>Abstract</p> <p>Background</p> <p>Targeted chlamydia screening has been advocated to reduce chlamydia associated reproductive sequelae. General practitioners are well positioned to play a major role in chlamydia control. The primary aim of this pilot study was to measure the effect of offering an online sexual health assessment tool, <it>Youth Check Your Risk</it>, on chlamydia testing rates among young people attending general practices. The secondary aim was to test the acceptability of the tool among general practitioners and young people.</p> <p>Methods</p> <p>General practitioners at three practices in Melbourne, Australia, referred patients aged 16 to 24 years to <it>Youth Check Your Risk </it><url>http://www.checkyourrisk.org.au</url> for use post-consultation between March to October 2007. The proportion of young people tested for chlamydia before and during the implementation of the tool was compared. Acceptability was assessed through a structured interviewer-administered questionnaire with general practitioners, and anonymous online data provided by <it>Youth Check Your Risk </it>users.</p> <p>Results</p> <p>The intervention did not result in any significant increases in the proportion of 16 to 24 year old males (2.7% to 3.0%) or females (6.3% to 6.4%) tested for chlamydia. A small increase in the proportion of 16 to 19 year old females tested was seen (4.1% to 7.2%). Of the 2997 patients seen during the intervention phase, 871 (29.1%) were referred to <it>Youth Check Your Risk </it>and 120 used it (13.8%). Major reasons for low referral rates reported by practitioners included lack of time, discomfort with raising the issue of testing, and difficulty in remembering to refer patients.</p> <p>Conclusion</p> <p>Offering an online sexual risk assessment tool in general practice did not significantly increase the proportion of young people tested for chlamydia, with GPs identifying a number of barriers to referring young people to <it>Youth Check Your Risk</it>. Future interventions aimed at increasing chlamydia screening in general practice with the aid of an online risk assessment tool need to identify and overcome barriers to testing.</p

    The Explanatory Value of Abstracting Away from Idiosyncratic and Messy Detail

    Some explanations are relatively abstract: they abstract away from the idiosyncratic or messy details of the case in hand. The received wisdom in philosophy is that this is a virtue for any explanation to possess. I argue that the apparent consensus on this point is illusory. When philosophers make this claim, they differ on which of four alternative varieties of abstractness they have in mind. What's more, for each variety of abstractness there are several alternative reasons to think that the variety of abstractness in question is a virtue. I identify the most promising reasons, and dismiss some others.
