    Deep optical imaging of the field of PC1643+4631A&B, II: Estimating the colours and redshifts of faint galaxies

    In an investigation of the cause of the cosmic microwave background decrement in the field of the z = 3.8 quasar pair PC1643+4631, we have carried out a study to photometrically estimate the redshifts of galaxies in deep multi-colour optical images of the field taken with the WHT. To examine the possibility that a massive cluster of galaxies lies in the field, we have attempted to recover simulated galaxies with intrinsic colours matching those of the model galaxies used in the photometric redshift estimation. We find that when such model galaxies are added to our images, there is considerable scatter of the recovered galaxy redshifts away from the model value; this scatter is larger than that expected from photometric errors and is the result of confusion, simply due to ground-based seeing, between objects in the field. We have also compared the likely efficiency of the photometric redshift technique against the colour criteria used to select z>3 galaxies via the strong colour signature of the Lyman-limit break. We find that these techniques may significantly underestimate the true surface density of z>3, due to confusion between the high-redshift galaxies and other objects near the line of sight. We argue that the actual surface density of z=3 galaxies may be as much as 6 times greater than that estimated by previous ground-based studies, and note that this conclusion is consistent with the surface density of high-redshift objects found in the HDF.     Deep optical imaging of the field of PC1643+4631A&B, I: Spatial distributions and the counts of faint galaxies

    We present deep optical images of the PC1643+4631 field obtained at the WHT. This field contains two quasars at redshifts z=3.79 & 3.83 and a cosmic microwave background (CMB) decrement detected with the Ryle Telescope. The images are in U,G,V,R and I filters, and are complete to 25th magnitude in R and G and to 25.5 in U. The isophotal galaxy counts are consistent with the results of Metcalde et al. (1996), Hogg et al. (1997), and others. We find an excess of robust high-redshift Ly-break galaxy candidates with 25.0<R<25.5 compared with the mean number found in the fields studied by Steidel et al. -we expect 7 but find 16 - but we do not find that the galaxies are concentrated in the direction of the CMB decrement. However, we are still not sure of the distance to the system causing the CMB decrement. We have also used our images to compare the commonly used object-finding algorithms of FOCAS and SExtractor: we find FOCAS the more efficient at detecting faint objects and the better at dealing with composite objects, whereas SExtractor's morphological classification is more reliable, especially for faint objects near the resolution limit. More generally, we have also compared the flux lost using isophotal apertures on a real image with that on a noise-only image: recovery of artificial galaxies from the noise-only image significantly overestimates the flux lost from the galaxies, and we find that the corrections made using this technique suffer a systematic error of some 0.4 magnitudes.Comment: 17 pages, 40 figures, submitted to MNRAS, 1 large figure avaliable at ftp://ftp.mrao.cam.ac.uk:/pub/PC1643/paper1.figure18.p

Liberals, on the other hand, are less likely to endorse liberal conspiracy theories if they have both greater political knowledge and more trust in government

    Diagnosing horizontal and inter-channel observation error correlations for SEVIRI observations using observation-minus-background and observation-minus-analysis statistics

    It has been common practice in data assimilation to treat observation errors as uncorrelated; however, meteorological centres are beginning to use correlated inter-channel observation errors in their operational assimilation systems. In this work, we are the first to characterise inter-channel and spatial error correlations for Spinning Enhanced Visible and Infrared Imager (SEVIRI) observations that are assimilated into the Met Office high-resolution model. The errors are calculated using a diagnostic that calculates statistical averages of observation-minus-background and observation-minus-analysis residuals. This diagnostic is sensitive to the background and observation error statistics used in the assimilation, although, with careful interpretation of the results, it can still provide useful information. We find that the diagnosed SEVIRI error variances are as low as one-tenth of those currently used in the operational system. The water vapour channels have significantly correlated inter-channel errors, as do the surface channels. The surface channels have larger observation error variances and inter-channel correlations in coastal areas of the domain; this is the result of assimilating mixed pixel (land-sea) observations. The horizontal observation error correlations range between 30 km and 80 km, which is larger than the operational thinning distance of 24 km. We also find that estimates from the diagnostics are unaffected by biased observations, provided that the observation-minus-background and observation-minus-analysis residual means are subtracted

    The structure of a reduced form of OxyR from Neisseria meningitidis

In N. meningitidis, expression of catalase, which is responsible for detoxifying hydrogen peroxide, is controlled by OxyR, a redox responsive LysR-type regulator. OxyR responds directly to intracellular hydrogen peroxide through the reversible formation of a disulphide bond between C199 and C208 in the regulatory domain of the protein. Results: We report the first crystal structure of the regulatory domain of an OxyR protein (NMB0173 from N. meningitidis) in the reduced state i.e. with cysteines at positions 199 and 208. The protein was crystallized under reducing conditions and the structure determined to a resolution of 2.4 Å. The overall fold of the Neisseria OxyR shows a high degree of similarity to the structure of a C199S mutant OxyR from E. coli, which cannot form the redox sensitive disulphide. In the neisserial structure, C199 is located at the start of helix α3, separated by 18 Å from C208, which is positioned between helices α3 and α4. In common with other LysR-type regulators, full length OxyR proteins are known to assemble into tetramers. Modelling of the full length neisserial OxyR as a tetramer indicated that C199 and C208 are located close to the dimer-dimer interface in the assembled tetramer. The formation of the C199-C208 disulphide may thus affect the quaternary structure of the protein. Conclusion: Given the high level of structural similarity between OxyR from N. meningitidis and E. coli, we conclude that the redox response mechanism is likely to be similar in both species, involving the reversible formation of a disulphide between C199-C208. Modelling suggests that disulphide formation would directly affect the interface between regulatory domains in an OxyR tetramer which in turn may lead to an alteration in the spacing/orientation of the DNA-binding domains and hence the interaction of OxyR with its DNA binding sites.This work was supported by UK Medical Research Council, the Biotechnology Biological Research Council, and by a MRC Research Studentship

    Evaluating the effectiveness of mindfulness alone compared to exercise and mindfulness on fatigue in women with gynaecology cancer (GEMS): Protocol for a randomised feasibility trial

    BackgroundIn 2020 Globocan reported nearly 1.4 million new cases of gynaecology cancer worldwide. Cancer related fatigue has been identified as a symptom that can be present for gynaecology cancer patients many years after treatment. The current evidence around the management of this symptom suggests that exercise has the most positive outcome. However, some ambiguity remains around the evidence and whether it can address all areas of fatigue effectively. More recently, other interventions such as mindfulness have begun to show a favourable response to the management of symptoms for cancer patients. To date there has been little research that explores the feasibility of using both these interventions together in a gynaecology cancer population. This study aims to explore the feasibility of delivering an intervention that involves mindfulness and mindfulness and exercise and will explore the effect of this on fatigue, sleep, mood and quality of life.Methods/designThis randomised control trial will assess the interventions outcomes using a pre and post design and will also include a qualitative process evaluation. Participants will be randomised into one of 2 groups. One group will undertake mindfulness only and the other group will complete exercise and mindfulness. Both groups will use a mobile application to complete these interventions over 8 weeks. The mobile app will be tailored to reflect the group the participants have drawn during randomisation. Self-reported questionnaire data will be assessed at baseline prior to commencing intervention and at post intervention. Feasibility will be assessed through recruitment, adherence, retention and attrition. Acceptability and participant perspective of participation (process evaluation), will be explored using focus groups.DiscussionThis trial will hope to evidence and demonstrate that combination of two interventions such as mindfulness and exercise will further improve outcomes of fatigue and wellbeing in gynaecology cancer. The results of this study will be used to assess (i) the feasibility to deliver this type of intervention to this population of cancer patients using a digital platform; (ii) assist this group of women diagnosed with cancer to manage fatigue and other symptoms of sleep, mood and impact their quality of life.Trial registrationNCT05561413

    A Randomized, Crossover Study of the Acute Cognitive and Cerebral Blood Flow Effects of Phenolic, Nitrate and Botanical Beverages in Young, Healthy Humans

    Background: In whole foods, polyphenols exist alongside a wide array of other potentially bioactive phytochemicals. Yet, investigations of the effects of combinations of polyphenols with other phytochemicals are limited. Objective: The current study investigated the effects of combining extracts of beetroot, ginseng and sage with phenolic-rich apple, blueberry and coffee berry extracts. Design: This randomized, double-blind, placebo-controlled crossover design investigated three active beverages in 32 healthy adults aged 18&ndash;49 years. Each investigational beverage comprised extracts of beetroot, ginseng and sage. Each also contained a phenolic-rich extract derived from apple (containing 234 mg flavanols), blueberry (300 mg anthocyanins) or coffee berry (440 mg chlorogenic acid). Cognition, mood and CBF parameters were assessed at baseline and then again at 60, 180 and 360 min post-drink. Results: Robust effects on mood and CBF were seen for the apple and coffee berry beverages, with increased subjective energetic arousal and hemodynamic responses being observed. Fewer effects were seen with the blueberry extract beverage. Conclusions: Either the combination of beetroot, ginseng and sage was enhanced by the synergistic addition of the apple and coffee berry extract (and to a lesser extent the blueberry extract) or the former two phenolic-rich extracts were capable of evincing the robust mood and CBF effects alone

    An interested and engaged electorate is widely believed to be an indicator of democratic health. As such, the aggregate level of political interest of an electorate – macrointerest – is an essential commodity in a democracy, and understanding the forces that change macrointerest is important for diagnosing the health of a democracy. Because being interested in politics requires time and effort, the article theorizes that the electorate’s level of political interest will be highest when the electorate believes the government cannot be trusted or is performing poorly. To test hypotheses derived from a proposed theory against rival explanations, the study develops a measure of macrointerest using a quarterly time series of aggregated survey items (1973–2014) of political interest. The authors find support for the theory that the electorate responds as reasonable agents when determining how closely to monitor elected officials: interest is positively related to decreases in trust in government

    Use of Accelerometers in a Large Field-Based Study of Children: Protocols, Design Issues, and Effects on Precision

    Background: Objective methods can improve accuracy of physical activity measurement in field studies but uncertainties remain about their use. Methods: Children age 11 years from the Avon Longitudinal Study of Parents and Children (ALSPAC), were asked to wear a uni-axial accelerometer (MTI Actigraph) for 7 days. Results: Of 7159 children who attended for assessment, 5595 (78%) provided valid measures. The reliability coefficient for 3 days of recording was .7 and the power to detect a difference of 0.07 SDs (P ≤ .05) was \u3e 90%. Measures tended to be higher on the first day of recording (17 counts/min; 95% CI, 10-24) and if children wore the monitor for fewer days, but these differences were small. The children who provided valid measures of activity were different from those who did not, but the differences were modest. Conclusion: Objective measures of physical activity can be incorporated into large longitudinal studies of children
