    Seasonal behavior of mercury species in waters and sediments from the Negro River Basin, Amazon, Brazil

    Total (HgT), reactive (HgR) and organic (Hg ORG) mercury species have been quantified in non-filtered waters collected from the Negro River Basin, Amazon (from January/2002 through January/2004), in both black and white water bodies. Black waters presented the highest HgT, HgORG and HgR concentration (29.1 ng L-1, 1.63 ng L-1 and 3.9 ng L-1, respectively), while, white waters presented the lowest HgT, Hg ORG and HgR concentration (8.8 ng L-1, 0.49 ng L-1 and 1.2 ng L-1, respectively). An inverse correlation between HgT and the water level over the basin was obtained, although the HgORG concentration has increased in the first rainy months and then decreased as the water level rose. Total mercury in surface sediments (0-10 cm) ranged from 70 to 271 mg kg-1 being the methylmercury 0.47-1.79 % of this stock. The results indicate that HgR and the labile dissolved organic matter were introduced into the aquatic environment during the flooding season, especially in the earlier stages, thus contributing to mercury methylation. ©2007 Sociedade Brasileira de Química

    As substâncias de alarme, liberadas após danos às células epidérmicas de peixes, é um dos sinais mais eficientes no grupo Osteriophysan, isto é, 72% das espécies de água doce. A hipoxantina-3-N-óxido é indicada como um dos possíveis componentes ativos dessas substâncias. Ensaios foram feitos com as espécies Matrinxã e Tambaqui através de aquários com capacidade de 100 L, instrumentalizados com sistemas de introdução de amostra e aquisição de imagens de forma remota. Os resultados indicaram na concentração de 6,0 µg L-1 alterações comportamentais incomuns como letargia, forrageio para o fundo e ataques, que apoiam a premissa de que a hipoxantina-3-N-óxido é eficaz na indução de sinais de alarme. A hipoxantina pode ser futuramente um bom método de manejo para proteção da ictiofauna, sobretudo no setor hidrelétrico, por ter eficácia comprovada, ser uma substância sintética, comercial e não tóxica à biota e ao ambiente aquático

    The influence of seasonalness on the structural characteristics of aquatic humic substances extracted from Negro River (Amazon state) waters: Interactions with Hg(II)

    In this work, humic substances were extracted from water samples collected monthly from the Negro River basin in the Amazon state (Brazil) to study their properties in the Amazonian environment and interactions with the mercury ion considering the influence of seasonalness in this formation. The C/H, C/N and C/O atomic ratio parameters, functional groups, concentration of semiquinone-type free radicals, pH, pluviometric and fluviometric indices, and mercury concentrations were interpreted using hierarchical cluster analysis (HCA) and principal component analysis (PCA). The statistical analyses showed that when the pluviometric index was greater and the fluviometric index was smaller, the degree of humification of aquatic substances was greater. The following decreasing order of the degree of humification of the AHS collected monthly was established: Nov/02 to Feb/03 > Mar/02 to May/02 > Jun/02 to Oct/02. The greatest concentrations of mercury were detected in more humidified samples. These results suggest that due to inter and/or intra-molecular rearrangements, the degree of humification of aquatic humic substances is related to its affinity for Hg(II) ions. ©2007 Sociedade Brasileira de Química

    Nanoscaled hydrated antimony (V) oxide as a new approach to first-line antileishmanial drugs

    Background: Coordination compounds of pentavalent antimony have been, and remain, the first-line drugs in leishmaniasis treatment for >70 years. Molecular forms of Sb (V) complexes are commercialized as sodium stibogluconate (Pentostam®) and meglumine antimoniate (MA) (Glucantime®). Ever-increasing drug resistance in the parasites limits the use of antimonials, due to the low drug concentrations being administered against high parasitic counts. Sb5+ toxicity provokes severe side effects during treatment. To enhance therapeutic potency and to increase Sb (V) concentration within the target cells, we decided to try a new active substance form, a hydrosol of Sb2O5·nH2O nanoparticles (NPs), instead of molecular drugs. Methodology/principal findings: Sb2O5·nH2O NPs were synthesized by controlled SbCl5 hydrolysis in a great excess of water. Sb2O5·nH2O phase formation was confirmed by X-ray diffraction. The surface of Sb (V) NPs was treated with ligands with a high affinity for target cell membrane receptors. The mean particle size determined by dynamic light scattering and transmission electron microscopy was ~35–45 nm. In vitro tests demonstrated a 2.5–3 times higher antiparasitic activity of Sb (V) nanohybrid hydrosols, when compared to MA solution. A similar comparison for in vivo treatment of experimental cutaneous leishmaniasis with Sb5+ nanohybrids showed a 1.75–1.85 times more effective decrease in the lesions. Microimages of tissue fragments confirmed the presence of NPs inside the cytoplasm of infected macrophages. Conclusion/significance: Sb2O5·nH2O hydrosols are proposed as a new form of treatment for cutaneous leishmaniasis caused by Leishmania amazonensis. The NPs penetrate directly into the affected cells, creating a high local concentration of the drug, a precondition to overcoming the parasite resistance to molecular forms of pentavalent antimonials. The nanohybrids are more effective at a lower dose, when compared to MA, the molecular drug. Our data suggest that the new form of treatment has the potential to reduce and simplify the course of cutaneous leishmaniasis treatment. At the same time, Sb2O5·nH2O hydrosols provide an opportunity to avoid toxic antimony (V) spreading throughout the body. © 2016 Franco et al

    Potential of Bacterial Isolates from a Stream in Manaus-Amazon to Bioremediate Chromium-Contaminated Environments

    Igarapé do Quarenta (IgQ), a stream located in the Manaus-AM, BR, has directly experienced the impacts of urban expansion over the last five decades, which contributed for its contamination. As an affluent of Rio Negro, IgQ also affects the water quality of this important river that bathes Manaus. However, the stress caused by the prolonged exposition to chemical agents may have selected microorganisms that exhibit great bioremediation potential. In the present study, bacteria isolated from four distinct sites of the IgQ were identified, and their potential to degrade hexavalent chromium (Cr(VI)) was investigated using the s-diphenylcarbazide method. Among the investigated isolates, 14 exhibited resistance against Cr(VI) at a concentration of 300 mg/L and eight isolates reduced over 50% (53.5–97.4%) chromium ratios after 72 h of incubation. Those isolates were identified by gene sequencing and classified in 10 genera (Acidovorax sp., Acinetobacter sp., Alicycliphilus sp., Bacillus sp., Comamonas sp., Enterobacter sp., Micrococcus sp., Proteus sp., Serratia sp., and Vagococcus sp.). Under control conditions, the isolate of Vagococcus sp. genus, in only 24 h of incubation, reduced 96.8% of the rate of Cr(VI) added to the culture medium at the concentration of 10 mg/L. Obtained results indicate that the Vagococcus sp. exhibits a great potential to be used in the bioremediation of areas contaminated with chromium. The mechanisms of action of microorganisms should be investigated for more specific applications in the decontamination of effluents and direct use of its by-products to bioremediate polluted environments. © 2018, The Author(s)
