    Birth weight, current body mass index, and insulin sensitivity and secretion in young adults in two Latin American populations

    Background and aims: Although studies have shown association of birth weight (BW) and adult body mass index (BMI) with insulin sensitivity in adults, there is limited evidence that BW is associated with insulin secretion. We assessed the associations between BW and current BMI with insulin sensitivity and secretion in young Latin American adults. Methods and results: Two birth cohorts, one from Ribeirao Preto, Brazil, based on 1984 participants aged 23-25 years, and another from Limache, Chile, based on 965 participants aged 22-28 years were studied. Weight and height at birth, and current fasting plasma glucose and insulin levels were measured. Insulin sensitivity (HOMA%S) and secretion (HOMA%beta) were estimated using the Homeostatic Model Assessment (HOMA2). Multiple linear regression analyses were carried out to test the associations between BW and adult BMI z-scores on log HOMA%S and log HOMA%beta. BW z-score was associated with HOMA%S in the two populations and HOMA%beta in Ribeirao Preto when adult BMI z-score was included in the model. BW z-score was associated with decreasing insulin secretion even without adjusting for adult BMI, but only in Ribeirao Preto. BMI z-score was associated with low HOMA%S and high HOMA%beta. No interactions between BW and BMI z-scores on insulin sensitivity were shown. Conclusions: This study supports the finding that BW may affect insulin sensitivity and secretion in young adults. The effect size of BW on insulin status is small in comparison to current BMI. (C) 2010 Elsevier B.V.     Concentrações sangüíneas de metais pesados e praguicidas organoclorados em crianças de 1 a 10 anos

    Foi realizado estudo em 6 bairros do Município de Cubatão, SP, Brasil, situados às margens dos principais rios do município, com uma amostragem de 251 crianças de 1 a 10 anos de idade. Foram verificadas as concentrações sangüíneas de praguicidas organoclorados em 242 crianças, sendo que 73 crianças (30%) apresentaram concentrações de p-p'DDE (média = 0,85 ± 2,13 µg/l.), e em 47 crianças (19%) foi encontrado HCH total (média = 0,28 ± 0,79 µg/l.). Verificou-se também a concentração sangüínea de chumbo e mercúrio, sendo que se obteve a plumbemia média de 17,8 ± 5,8 µg/dl. e concentração média de mercúrio de 9,1 ± 6,4 µg/l. Crianças consumidoras de organismos aquáticos de origem exclusiva dos rios de Cubatão apresentaram teores médios de mercúrio (média = 14,7 ±7,1 µg/l.) significativamente maiores (p < 0,04) em comparação às crianças não consumidoras de organismos aquáticos de qualquer origem (média = 10,0 ± 6,5 µg/l.).A study of a sample of 251 children aged between 1 and 10 was carried out in 6 districts of Cubatão county, S. Paulo State, Brazil. This study showed the presence of organochlorine pesticides in the blood serum of 242 children, p-p' DDE was detected in 73 of the samples (30%) and HCH in 47 them (19%). The mean value of p-p' DDE was 0.85 ± 2.13 µg/l. and that of HCH was 0.28 ± 0.79 µg/l. Lead and mercury were also detected in the blood serum. The mean value for lead was 17.8 ± 5.8 µg/l. and for mercury was 9.1 ± 0.79 µg/l. The levels found in consumers of aquatic organisms taken from the rivers of Cubatão showed a mean mercury level (mean = 14.7 ± 7.1 µg/l.) higher than (p < 0.04) that of those who did not consume any aquatic organisms at all (mean = 10.0 ± 6.5 µg/l.)

    Levels of exposure to organochlorine pesticides in open-air dump dwellers

    OBJECTIVE: To estimate the degree of internal exposure to organochlorine pesticides (persistent organic pollutants, POP) among inhabitants of an open-air dump. METHODS: This was an observational study with simultaneous measurement and control, in which the criterion for subjects was that they dwelled in an area contaminated by POP and the effect of such contamination was the concentration of such substances into the blood. The study population consisted of 238 people living on an open-air dump in Pilões, in the municipality of Cubatão, SP, Brazil, and the control population was formed by 258 people, also resident in that municipality, in the Cota 200 district. The following pesticides were analysed: hexachlorobenzene (HCB), p-p'DDT, p-p'DDE, p-p'DDD, o-p'DDT, a HCH (hexachlorocyclohexane); b HCH; g HCH; aldrin; dieldrin; endrin; heptachlor; heptachlor-epoxy and mirex. RESULTS: The average blood level of HCB in Pilões was 4.66 µg/l, which was 155 times higher than the average for Cota 200 (0.03 µg/l). In Pilões the average blood concentration of total DDT was 3.71 µg/l, which was twice the level found in Cota 200 (1.85 µg/l). Total HCH presented blood concentrations that were six times greater in Pilões (0.84 µg/l versus 0.13 µg/l). CONCLUSIONS: A positive association was shown between dwelling in Pilões and the presence of POP in the blood, with much higher risks than in localities without the presence of these contaminants.OBJETIVO: Estimar o grau de exposição interna aos praguicidas organoclorados (POP) dos moradores de um atêrro a céu aberto. MÉTODOS: Estudo observacional, de aferição simultânea e controlado, cujo fator de estudo foi residir em área contaminada por POP e o efeito foi a concentração sanguínea dessas substâncias. A população de estudo foi de 238 pessoas residentes em um aterro a céu aberto (Pilões), no município de Cubatão, SP; e a população de controle ficou dimensionada em 258 pessoas, também residentes no mesmo município (Cota 200). Foram analisados os praguicidas HCB, p-p'DDT, p-p'DDE, p-p'DDD, o-p'DDT, a HCH; b HCH; g HCH; Aldrin; Dieldrin; Endrin; Heptaclor; Heptaclor-epoxi e Mirex. RESULTADOS: Os teores médios sangüíneos de HCB em Pilões foram de 4,66 µg/L, 155 vezes maior que a média na Cota 200 (0,03 µg/L). Em Pilões, os teores médios de DDT total foram de 3,71 µg/L, duas vezes maior do que na Cota 200 (1,85 µg/L) e o HCH total apresentou concentrações sangüíneas seis vezes maiores em Pilões, 0,84 µg/L, contra 0,13 µg/L. CONCLUSÕES: Evidenciou-se associação positiva entre residir em Pilões e apresentar teores sangüíneos de POP, com risco muitas vezes maior quando comparado a localidades sem a presença desses contaminantes

    The polycystic kidney disease 1 gene encodes a 14 kb transcript and lies within a duplicated region on chromosome 16

    Autosomal dominant polycystic kidney disease (ADPKD) is a common genetic disorder that frequently results in renal fallure due to progressive cyst development. The major locus, PKD1, maps to 16p13.3. We identified a chromosome translocation associated with ADPKD that disrupts a gene (PBP) encoding a 14 kb transcript in the PKD1 candidate region. Further mutations of the PBP gene were found in PKD1 patients, two deletions (one a de novo event) and a splicing defect, confirming that PBP is the PKD1 gene. This gene is located adjacent to the TSC2 locus in a genomic region that is reiterated more proximally on 16p. The duplicate area encodes three transcripts substantially homologous to the PKD1 transcript. Partial sequence analysis of the PKD1 transcript shows that it encodes a novel protein whose function is at present unknown
