    Pyramiding of blast and bacterial leaf blight resistance genes into rice cultivar RD6 using marker assisted selection

    Blast caused by the fungus Magnaporthe oryzae (Hebert) Barr. and bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) are two major diseases of rice (Oryza sativa). The use of varietal resistance is the most appropriate strategy for controlling the diseases, and molecular assisted selection can potentially accelerate breeding programs. The objective of this study was to pyramid genes conferring resistance to blast and bacterial leaf blight diseases to rice cultivar RD6, using molecular assisted selection. Near-isogenic lines (NIL) derived from two blast resistant crosses (RD6 × P0489 and RD6 × Jao Hom Nin) were pyramided with IR62266 (xa5), to transfer bacterial leaf blight resistance to RD6 introgression lines. Five flanking sets of simple sequence repeat (SSR) markers (RM319/RM212, RM48/RM207, RM224/RM144, RM313/RM277 and RM122/RM159: four for blast and one for BLB resistance) were used for screening of introgression lines carrying five quantitative trait loci (QTLs) from the BC1F2 generation through to BC2F2:3 generation, and 12 pyramiding lines were identified. Gene validation for blast and bacterial leaf blight diseases was accomplished using artificial inoculation under greenhouse conditions. BC2F2:3 2-8-2-24 and BC2F2:3 2-8-2-25 showed greater levels of blast broad spectrum resistance (BSR) whereas BC2F2:3 2-8-2-36 expressed the highest of bacterial leaf blight resistance with a high blast BSR.Keywords: Gene pyramiding, introgression lines, molecular marker, Near-isogenic lines, SSR.African Journal of Biotechnology Vol. 12(28), pp. 4432-443

    Genetics Analysis and Heritability of Fruit Characters in Muskmelon (Cucumis Melo L.) Using Extreme Parental Differences

    Taeng-Thai is the Thai name for oriental pickling melon or muskmelon (Cucumis melo L.) which is a Cucumis species. Based on consumption as Thai traditional dessert, fruit characters at ripening stage are very important to improve the muskmelon cultivar. Understanding the genetic control of fruit traits is the key to a successful for crop improvement. Research aimed to determine the genetic effects, number of genes and heritability of fruit characters in muskmelon. The results have shown that both additive and dominant effects governed for fruit weight, fruit length, fruit width and fruit thickness with the number of gene as 40.26, 1.55, 10.70 and 11.10 genes, respectively. Simple phenotypic correlation coefficients shown, had a significant positive correlation among the four fruit traits. Due to the quantitative inheritance of fruit characters, quantitative traits loci (QTL) mapping of those traits were necessary to identify all controlling genes of fruit traits in further muskmelon improvement

    Stability of Four New Sources of Bacterial Leaf Blight Resistance in Thailand Obtained From Indigenous Rice Varieties

    Bacterial leaf blight (BLB) disease caused by Xanthomonas oryzae pv. oryzae (Xoo) is one of the most serious diseases in rice production. Breeding varieties specifically for their resistance to BLB disease is therefore an efficient and cost-effective strategy. However, the resistance gene for BLB can be race and non-race specific, meaning it is often overcome by the pathogen. The identification of new sources of resistance genes for Xoo is crucial in rice breeding programmes. In this study, six rice varieties were assessed using six Xoo isolates in multiple screening conditions. The GGE biplot analysis considers both genotype (G) and genotype environment (GE) interaction effects and demonstrates GE interaction. The first two principal components (PCs) accounted for 95.46% of the total GE variation in the data. Based on lesion length and stability performance, Phaladum was the most ideal genotype against all Xoo isolates in the four screening conditions. The results relayed that Phaladum indigenous rice varieties could be considered as new sources of bacterial leaf blight resistance in Thailand. In the future, the BLB resistance gene in this variety will be identified in regard to mode of inheritance and used as parental line in rice breeding programmes for resistance to BLB

    Genotypic variation in grain yield and flowering pattern in terminal and intermittent drought screening methods in rainfed lowland rice

    Rainfed lowland rice in the Mekong region is mostly grown in drought-prone areas, and drought resistant varieties could be selected using either terminal drought (severe water stress after flowering) or intermittent drought (frequent mild stress) screening. Delay in flowering is considered as a putative trait for selection of drought resistant genotypes, and its effectiveness as an indirect selection trait and its association with genotype's flowering duration, i.e. the time taken from the first to last flower in a plot, was evaluated under the two drought screening methods. These screening methods together with flood control were used at two locations, Chum Phae and Ubon in north-east Thailand using a total of 70 genotypes with different growing duration. Delay in flowering was estimated for the two drought screening methods and flowering of individual panicles within a plot was recorded to determine flowering duration of each genotype in the flood and terminal drought conditions.Mean reduction in grain yield was 52-55% under the terminal drought screening while it was 10-19% for the whole population and 23-33% for early flowering genotypes in the intermittent drought. Delay in flowering under intermittent drought condition at both locations was associated with yield reduction; genotypes that exhibited shorter delay in flowering had smaller yield reduction percentage and larger number of panicles. At Chum Phae, where grain yield reduction was associated with flowering time, genotypes with longer flowering duration under terminal drought had smaller yield reduction, and this was found even among genotypes that flowered about the same time under flood condition. However, there was no association between delay in flowering and flowering duration. It is concluded that delay in flowering appears promising as a selection criterion under intermittent drought conditions, provided a higher yield reduction can be achieved for example by using an increased irrigation interval

    On-farm multi-location evaluation of occurrence of drought types and rice genotypes selected from controlled- water on-station experiments in Northeast Thailand

    Rainfed lowland rice is the most common crop ecosystem in Northeast Thailand and varieties well adapted to drought-prone environments are required. On-farm multi-location experiments were conducted for 3 years to determine the drought type commonly developed, to identify genotype by environment interaction for rainfed lowland rice and to examine whether genotypic variation in these experiments was related to that in on-station controlled water- environment screening experiments conducted previously (Monkham et al., 2015). Among 32 on-farm experiments, 4 experienced continuous drought with no standing water during the whole growing season, and 3 of them all in high toposequence positions died before maturity. For the remaining experiments, both intermittent and terminal droughts were common, while some experiments developed early season drought and delayed transplanting but resulted in no apparent yield penalty. The first set of multi-location experiments consisted of 14 experiments in 6 provinces in Northeast Thailand across 2 years with 24 common genotypes. Mean grain yield of 13 environments that were harvested was 2300–3900 kg ha and no severe drought developed at any environment. While main effect of genotype was not significant for grain yield, genotype by environment interaction was significant, and the results of clustering analysis identified 3 environmental groups and 3 genotype groups that explained the main source of the genotype by environment interaction. The genotypic variation obtained in the on-farm multi-location experiments in one environmental group of low yielding environments was correlated with that obtained under earlier intermittent drought selection experiments on-station. The results also indicate the usefulness of screening genotypes based on short delays in the intermittent drought conditions on-station. A second set of multi-location experiments conducted throughout Northeast Thailand to test the performance of four genotypes selected from the first set of multi-location experiments confirmed that these genotypes outperformed the popular variety KDML105. The present study confirmed the usefulness of initial screening under intermittent drought conditions conducted on research stations followed by further on-farm selection. It is suggested that on-station selection experiments should target yield levels representative of on-farm yield levels

    Chemical and molecular characterization of fragrance in Black Glutinous Rice from Lao PDR

    Fifty three accessions of lowland Black Glutinous Rice (BGR) and three white rice varieties from Lao PDR were evaluated for variations in 2-acetyl-1-pyrroline (2-AP), a main aroma compound present in fragrant rice and a 257 bp genomic fragment associated with a gene encoding 2-AP. The objective of this study was to characterize the variation in aromatic characters of Lao BGR by using chemical and molecular techniques. Thirty six out of the 56 accessions had aromatic properties, containing 2-acetyl-1-pyrroline (2-AP). The concentrations of 2-AP in unpolished samples were about 50% higher than those of the polished samples. The concentrations of 2-AP in BGR accessions were lower than those of white aromatic rice varieties in which 2-AP was highest in Kai Noi Leuang (0.688 ppm). Seventeen accessions and two of white rice (Kai Noi Leuang and TDK5) had a common 355 bp allele corresponding to that found in Calrose, a non-fragrant variety. A 257 bp allele was found in the 36 varieties of BGR and a local aromatic white rice variety, Hom Nang Nouan, sharing a common allele as in the fragrant variety KDML 105. Surprisingly, Kai Noi Leuang, a popular Lao local aromatic rice variety with the highest 2-AP concentration did not possess a 257 bp fragment which has been identified previously as a fragrant allele. This is the first study of the chemical and molecular aromatic properties of lowland BGR varieties of the Lao PDR and the data obtained will be valuable in screening other germplasm and in planning a breeding program in the country geared towards fragrant rices with superior agronomic properties

    Use of new generation single nucleotide polymorphism genotyping for rapid development of near-isogenic lines in rice

    Studies aimed at determining gene function or identity are greatly facilitated by introgression lines and phenotyping tools. However, developing introgression lines can take six to eight generations to reach the desired level of background purity and homozygosity. The objective of this study was to use new generation genome-wide single nucleotide polymorphism (SNP) genotyping, along with a molecular marker for the allele of interest, and a relevant phenotyping tool, to develop research populations with just four generations of backcrossing. Two populations were created for future research on aroma in rice (Oryza sativa L.): one derived from two indica parents and the other from one indica and one tropical japonica parent. The same recurrent parent was used for both populations.
