    Effect of Pyrodextrinization, Crosslinking and Heat- Moisture Treatment on In vitro Formation and Digestibility of Resistant Starch from African Locust Bean (Parkia biglobosa)

    Purpose: This investigation was carried out to determine the impact of Parkia biglobosa starch modification on the fractions, namely rapidly digestible starch (RDS), slowly digestible starch (SDS), and resistant starch (RS).Methods: Aqueous solution of sodium hypochlorite and potassium hydroxide was used to extract starch prior to modification by pyrodextrinization, cross-linking and heat-moisture treatment. Solubility,swelling power, x-ray diffraction, scanning electron microscopy (SEM) and thermal properties of the native and modified starches were also studied.Results: Pyrodextrinization (PD), cross-linking (CL), and heat-moisture treatment (HMT) reduced the swelling power to 6.73, 4.17 and 5.57 g/g, respectively but increased solubility by 59.0, 41, 41.5 and 39.5 %, respectively, and tended to decrease gelatinization enthalpy (Ă„H). Starch yield was 25.7 % on a whole seed basis. RS content significantly (p < 0.05) increased to 46.3, 49.2 and 45.3 %, respectively following PD, CL and HMT. X-ray diffraction resulrs indicate the presence of V-type crystallinity in the modified parkia starch while SEM showed PD and CL starch structures were more compact and dense than HMT starch which was irregularly-shaped formed.Conclusion: Native parkia starch modified by pyrodextrinization, cross-linking and heat-moisture treatment showed appreciably higher thermal stability which makes it suitable for incorporation in foods that are subject to high temperature processing and high shear.Keywords: Pyrodextrinization, Crosslinking, Heat-moisture treatment, Gelatinization, Resistant starch, Parkia biglobos

    First detection of bla TEM, SHV and CTX-M among Gram negative bacilli exhibiting extended spectrum β- lactamase phenotype isolated at University Hospital Center, Yalgado Ouedraogo, Ouagadougou, Burkina Faso

    Resistance to a wide variety of common antimicrobials is observed among clinical strains designed as extended spectrum β-lactmase (ESBL)  producers. They produce enzymatic protein which inactivates efficiently oxyimino cephalosporin and constitutes a serious global health concern that has complicated treatment strategies. Many studies report high prevalence of ESBL producers among Gram negative bacilli. The aim of this work was to identify the presence of TEM, SHV and CTX-M families in thesestrains which were initially screened by phenotypic method. Gram negative bacilli resisting third or four generation cephalosporin were isolated during anti-biogram study. The presence of ESBL positivity was detected using the double disk synergy test. Minimal inhibitory concentrations (MICs) of ceftriazon for any strain were determined using E-test manufacturing protocol. Polymerase chain reaction (PCR) analysis for β-lactamase (bla) genes of TEM, SHV and CTX-M family was carried out using designed primers in 171 ESBL isolates producers. Among 259 Gram negative bacilli collected, 171 (66, 02%) exhibited ESBL producers’ profile. Urine samples constitute major source of ESBL producers. The highest prevalence of ESBL was observed in Escherichia coli (75, 50%). Among ESBL isolates producers, gene prevalence of bla-CTX-M (65, 49%) was highest, followed by bla-TEM (25, 73%) and bla-SHV (18, 71%) in the present study. The frequency of ESBL producing strains among clinical isolates has been steadily increased. Continual drug resistance surveillance and molecular characteristics of ESBL isolates are necessary to guide the appropriate and judicious antibiotic use.Key words: Extended spectrum β-lactamase (ESBL), double disk synergy test, blaTEM, blaSHV, blaCTX-M, PCR

    Urinary tract infection among pregnant women at Bobo-Dioulasso: epidemiological and bacteriological aspects

    Urinary tract infections are the first infections in pregnant women and can cause serious complications during pregnancy. In order to improve its management in low income country like Burkina Faso, we conducted a prospect cross-sectional study, to describe its epidemiological and biological aspects in pregnant women at Souro Sanou hospital at Bobo-Dioulasso (Burkina Faso).Epidemiologically, the frequency of urinary tract infection was 27.5% and affected mostly 21 to 30 years pregnant women. Bacteriologically, the main isolates found were: E. coli, Staphylococcus aureus, Staphylococcus epidermis, Serratia, Acinetobacter, Enterobacter and Candida. The 81 isolates showed a good sensitivity to cephalosporin (C3G), aminoglycosides, oxacillin and erythromycin.     Microbiological and kinetic detection of gram negative bacilli producing extended-spectrum- β-lactamases (esbl) in emergencies and reanimation units of university hospital center, Yalgadoouedraogo, Burkina Faso

    Background: Epidemiology of extended-Spectrum- β-lactamases has become worldwide, and our aim was to establish the prevalence of isolates producer in university hospital center Yalgado OUEDRAOGO particularly in reanimation and emergencies units.Material and methods: Prospective study was drive during July 2009 to march 2012 in order to collect strains resisting to third generation of cephalosporin during diagnosis analysis of biological specimens. Susceptibility of bacteria to antimicrobial agents was evaluated by disc diffusion method. Production of extended-spectrum β-lactamases has been investigated by double disc diffusion and kinetic methods.Results: 259 isolates which resisted at least to one of third generation of cephalosporins were collected. Among them 188 (72, 58 %) were positive to synergy test by a double disc diffusion method. The MICs of ceftriaxone determined by E-test were under than 50kg/ml, 100kg/ml et 256kg/ml for respect 81,57°/° ; 55,26°/° et 39,74°/° of isolates. Hydrolyze of β-lactam ring by bacterial extract followed at spectrophotometer showed speeds running at 0 to 0,090UAb.mn-1 for both isolates. Extract of 171 bacterial strains positives to synergy test had hydrolyzed at least one of oxy-iminocephalosporins and were identified as producing extended- spectrum β-lactamases. Spices reported by this study were 99 Escherichia coli (57,89%) ; 28 Klebsisella pneumonia (16,37%) ; 15 Enterobactersp (8,77%) ; 19 Pseudomonas aeruginosa (11,11%) ; 4 Citrobactersp (2,33%) 2 Acinetobactersp (1,16%), 3 Proteus mirabilis (1,75%) and 1 Salmonella typhi (0,05%).Conclusion: This study showed that bacterial resistances by extended- spectrum β-lactamases are a reality in University Hospital center YalgadoOuedraogo.     Contribution of previous legumes to soil fertility and millet yields in West African Sahel

    Studies on combined effects of 4 legume crops residues and rock phosphate application on pearl millet yield were undertaken on sandy acid soil field from 2012 to 2015 at ICRISAT Sahelian center (ISC)-Sadore, Niger. The objective of the experiment was to assess the best combination of legume species x rate of crop residue x rock phosphate doses that can sustainably improve pearl millet yield in cereal monoculture system with a low input cost and minimum soil tillage. Over 3 years, the residual effect of previous legume crop residue significantly improved not only the grain yield (P<0.001) and dry residue yields (P<0.001) but also the growth parameters of pearl millet than millet mono-cropping. Treatments with or without natural rock phosphate did not show any statistical differences on millet yield while adding a micro dose of urea improved significantly the yield (P<0.001). The interaction effects of preceding legume crops in rotation with millet and restitution of dry residue on the earlier mentioned parameters across 3 years mono-cropping were studied in this experiment

    Effects of amodiaquine and artesunate on sulphadoxine-pyrimethamine pharmacokinetic parameters in children under five in Mali

    <p>Abstract</p> <p>Background</p> <p>Sulphadoxine-pyrimethamine, in combination with artesunate or amodiaquine, is recommended for the treatment of uncomplicated malaria and is being evaluated for intermittent preventive treatment. Yet, limited data is available on pharmacokinetic interactions between these drugs.</p> <p>Methods</p> <p>In a randomized controlled trial, children aged 6-59 months with uncomplicated <it>falciparum </it>malaria, received either one dose of sulphadoxine-pyrimethamine alone (SP), one dose of SP plus three daily doses of amodiaquine (SP+AQ) or one dose of SP plus 3 daily doses of artesunate (SP+AS). Exactly 100 ÎĽl of capillary blood was collected onto filter paper before drug administration at day 0 and at days 1, 3, 7, 14, 21 and 28 after drug administration for analysis of sulphadoxine and pyrimethamine pharmacokinetic parameters.</p> <p>Results</p> <p>Fourty, 38 and 31 patients in the SP, SP+AQ and SP+AS arms, respectively were included in this study. The concentrations on day 7 (that are associated with therapeutic efficacy) were similar between the SP, SP+AQ and SP+AS treatment arms for sulphadoxine (median [IQR] 35.25 [27.38-41.70], 34.95 [28.60-40.85] and 33.40 [24.63-44.05] ÎĽg/mL) and for pyrimethamine (56.75 [46.40-92.95], 58.75 [43.60-98.60] and 59.60 [42.45-86.63] ng/mL). There were statistically significant differences between the pyrimethamine volumes of distribution (4.65 [3.93-6.40], 4.00 [3.03-5.43] and 5.60 [4.40-7.20] L/kg; <it>p = 0.001</it>) and thus elimination half-life (3.26 [2.74 -3.82], 2.78 [2.24-3.65] and 4.02 [3.05-4.85] days; <it>p < 0.001</it>). This study confirmed the lower SP concentrations previously reported for young children when compared with adult malaria patients.</p> <p>Conclusion</p> <p>Despite slight differences in pyrimethamine volumes of distribution and elimination half-life, these data show similar exposure to SP over the critical initial seven days of treatment and support the current use of SP in combination with either AQ or AS for uncomplicated <it>falciparum </it>malaria treatment in young Malian children.</p

    Dynamics of antigenemia and transmission intensity of Wuchereria bancrofti following cessation of mass drug administration in a formerly highly endemic region of Mali

    Background After seven annual rounds of mass drug administration (MDA) in six Malian villages highly endemic for Wuchereria bancrofti (overall prevalence rate of 42.7%), treatment was discontinued in 2008. Surveillance was performed over the ensuing 5 years to detect recrudescence. Methods Circulating filarial antigen (CFA) was measured using immunochromatographic card tests (ICT) and Og4C3 ELISA in 6–7 year-olds. Antibody to the W. bancrofti infective larval stage (L3) antigen, Wb123, was tested in the same population in 2012. Microfilaraemia was assessed in ICT-positive subjects. Anopheles gambiae complex specimens were collected monthly using human landing catch (HLC) and pyrethrum spray catch (PSC). Anopheles gambiae complex infection with W. bancrofti was determined by dissection and reverse transcriptase polymerase chain reaction (RT-PCR) of mosquito pools. Results Annual CFA prevalence rates using ICT in children increased over time from 0% (0/289) in 2009 to 2.7% (8/301) in 2011, 3.9% (11/285) in 2012 and 4.5% (14/309) in 2013 (trend χ 2  = 11.85, df =3, P = 0.0006). Wb123 antibody positivity rates in 2013 were similar to the CFA prevalence by ELISA (5/285). Although two W. bancrofti-infected Anopheles were observed by dissection among 12,951 mosquitoes collected by HLC, none had L3 larvae when tested by L3-specific RT-PCR. No positive pools were detected among the mosquitoes collected by pyrethrum spray catch. Whereas ICT in 6–7 year-olds was the major surveillance tool, ICT positivity was also assessed in older children and adults (8–65 years old). CFA prevalence decreased in this group from 4.9% (39/800) to 3.5% (28/795) and 2.8% (50/1,812) in 2009, 2011 and 2012, respectively (trend χ 2  = 7.361, df =2, P = 0.0067). Some ICT-positive individuals were microfilaraemic in 2009 [2.6% (1/39)] and 2011 [8.3% (3/36)], but none were positive in 2012 or 2013. Conclusion Although ICT rates in children increased over the 5-year surveillance period, the decrease in ICT prevalence in the older group suggests a reduction in transmission intensity. This was consistent with the failure to detect infective mosquitoes or microfilaraemia. The threshold of ICT positivity in children may need to be re-assessed and other adjunct surveillance tools considered

    Utilisation des facteurs climatiques pour la surveillance de la fréquence des occurrences de méningite/ paludisme à Bamako

    Objectif : Notre travail consiste à : Déterminer les périodes de risque pour la méningite et le paludisme à Bamako; Etablir une corrélation entre : - d’une part les paramètres météorologiques (température de l’air, humidité relative de l’air, pluviométrie, insolation et vitesse du vent) et la fréquence des occurrences de paludisme ; - d’autre part les mêmes paramètres pour la méningite. Méthode : Nous avons fait une étude rétrospective de l’évolution du paludisme et de la méningite en fonction de la variation des paramètres météorologiques (température de l’air, humidité relative de l’air, pluviométrie, insolation et vitesse du vent) à Bamako. Résultats : Le paludisme, rare au 1er et 2èm trimestres, est très fréquent au 3è trimestre et au 4è trimestre. Quant à la méningite elle est fréquente au premier semestre et rare pendant le reste de l’année. La fréquence du paludisme est liée à la température moyenne de l’air, à la grande humidité de l’air, à la pluviométrie abondante, à la faible insolation et à la faible vitesse du vent. La fréquente de la méningite est liée à la haute température de l’air, à la faible humidité de l’air, à la faible pluviométrie, à la forte insolation et à la grande vitesse du vent. Conclusion : La fréquence du paludisme est liée à la grande humidité, et à la faible insolation. Quant à la méningite, sa fréquence est liée à la haute température, à la grande vitesse du vent et à la forte insolation

    Variety development for sorghum requires multi-location testing for yielding ability. In West-Africa, where adaptation to specific zones of rainfall distribution is crucial for the success of a variety, these multi-location trials need to be conducted within any specific zone of adaptation. As most West-African countries lack sufficient research station capacity for this type of yield testing, we have developed a trial design and experiments with a sharing of roles and responsibilities between farmers, NGO's and researchers that allow for effective varietal differentiation in the target environment across a wide range of production conditions within a zone. The added advantage of farmer participation in this early stage of variety evaluation is that farmers also evaluate a wide range of other traits that are essential for making a variety a success

    Polymerase Chain Reaction (PCR) provides a superior tool for the diagnosis of Pneumococcal Infection in Burkina Faso

    Purpose of study: The aim of this study was to determine the value of real-time Polymerase Chain Reaction (rt-PCR) in the routine surveillance of pneumococcal meningitis in Burkina Faso, compared to standard methods of culture, Gram stain and latex agglutination assay.Materiel and methods: A total of 385 specimens of cerebrospinal fluid were analyzed by the three standard bacteriological methods (Gram stain, latex agglutination assay, and culture) and real-time Polymerase Chain Reaction.Results: Of 385 specimens analyzed by these methods, 204 S. pneumoniae were detected by one or more  methods. Gram stain detected 36.4% (140/385) Gram positive encapsulated diplococci; 37.7% (145/385) and 20.8% (80/385) of the specimens were positive for pneumococci by latex agglutination assay and culture. These specimens were tested with rt-PCR, which confirmed 51.2% (197/385) S. pneumoniae positive. The sensitivity and specificity of culture were 54.4% and 31.5%, respectively, and the sensitivity and specificity of rt-PCR were 96.6% and 100%, respectively. These results showed that rt-PCR was more sensitive than Gram stain (p=0.0235), latex agglutination assay (p=0.0442)and culture (p=0.0006).The culture is the gold standard method; however, the result showed that rt-PCR had specificity and was as specific as Gram stain (p=0.3405) and latex agglutination assay (p=0.7745).Conclusion: rt-PCR was highly sensitive and specific. It could be used as a complementary diagnostic tool to  mprove case confirmation of bacterial meningitis. However,its high cost, the qualification of the technical staff and infrastructures required for its implementation, constitute obstacles to its widened use in countries with limited resources.Keywords: Streptococcus pneumoniae, meningitis, rt-PCR, standard bacteriological methods Objectif: Le but de cette étude était de déterminer la place de la rt-PCR dans la surveillance de routine deméningitespneumococciquesau Burkina Faso et la comparée avec les méthodes de la bactériologie classique: Culture, coloration de Gram et l’agglutination au latex.Matériel etméthodes: Au total, 385 échantillons deliquides céphalorachidiens (LCR)étaient analysés par les trois méthodes de la bactériologie classique (coloration de Gram, agglutination au latex, culture) et la PCR en temps réel.RESULTATS: Parmi 385 échantillons analysés, 204 cas de Streptococcuspneumoniae étaient détectés par une ou plusieurs méthodes. La coloration de Gram adétecté 36,4% (140/385) diplocoques encapsulés à Gram positif (DGP); 37,7% (145/385) et 20,8% (80/385) d’échantillons étaient positifs aux pneumocoques par l’agglutination au latex et la culture. Ces échantillons étaient aussi testés par rt-PCR qui a confirmé 51,2% (197/385) cas positifs de S. pneumoniae. La sensibilité et la spécificité de la culture étaient respectivement de 54,4% et 31,5%, et la sensibilité et la spécificité de rt-PCR étaient respectivement de 96,6% et 100%. Ces résultats ont montré que la rt-PCR était plus sensible que la coloration de Gram (p=0,0235), l’agglutination au latex (p=0,0442) et la  culture(p=0,0006).La culture est une méthode de référence; cependant, le résultat a montré que rt-PCR était plus sensibleet aussi spécifique que la coloration de Gram ((p=0,3405)et l’agglutination au latex (p=0,7745).Conclusion : :rt-PCR était plus sensible et plus spécifique. Elle pourrait être utilisée comme un outil de diagnosticcomplémentaire pour améliorer les cas de confirmation de méningites bactériennes.
