    Deployment and testing of a second prototype expandable surgical chamber in microgravity

    During microgravity exposure, two separate expandable surgical chambers were tested. Both chambers had been modified to fit the microgravity work station without extending over the sides of the table. Both chambers were attached to a portable laminar flow generator which served two purposes: to keep the chambers expanded during use; and to provide an operative area environment free of contamination. During the tests, the chambers were placed on various parts of a total body moulage to simulate management of several types of trauma. The tests consisted of cleansing contusions, debridement of burns, and suturing of lacerations. Also, indigo carmine dye was deliberately injected into the chamber during the tests to determine the ease of cleansing the chamber walls after contamination by escaping fluids. Upon completion of the tests, the expandable surgical chambers were deflated, folded, and placed in a flattened state back into their original containers for storage and later disposal.     Carbon-Rich Mira Variables: Radial Velocities and Distances

    Optical radial velocities have been measured for 38 C-type Mira variables. These data together with others in the literature are used to study the differences between optical and CO mm observations for C-Miras and the necessary corrections to the optical velocities are derived in order to obtain the true radial velocities of the variables. The difference between absorption and emission line velocities is also examined. A particularly large difference (+30 km\s) is found in the case of the H-alpha emission line. A catalogue is given of 177 C-Miras with estimated distances and radial velocities. The distances are based on bolometric magnitudes derived in Paper I using SAAO observations or (for 60 of the stars) using non-SAAO photometry. In the latter case the necessary transformations to the SAAO system are derived.     Solution pans and linear sand bedforms on the bare-rock limestone shelf of the Campeche Bank, Yucatán Peninsula, Mexico

    A high-resolution, near-surface geophysical survey was conducted in 2013 on the Campeche Bank, a carbonate platform offshore of Yucatán, Mexico, to provide a hazard assessment for future scientific drilling into the Chicxulub impact crater. It also provided an opportunity to obtain detailed information on the seafloor morphology and shallow stratigraphy of this understudied region. The seafloor exhibited two morphologies: (1) small-scale (<2 m) bare-rock karstic features, and (2) thin (<1 m) linear sand accumulations overlying the bedrock. Solution pans, circular to oblong depressions featured flat bottoms and steep sides, were the dominant karstic features; they are known to form subaerially by the pooling of rainwater and dissolution of carbonate. Observed pans were 10–50 cm deep and generally 1–8 m wide, but occasionally reach 15 m, significantly larger than any solution pan observed on land (maximum 6 m). These features likely grew over the course of many 10's of thousands of years in an arid environment while subaerially exposed during lowered sea levels. Surface sands are organized into linear bedforms oriented NE-SW, 10's to 100's meters wide, and kilometers long. These features are identified as sand ribbons (longitudinal bedforms), and contained asymmetric secondary transverse bedforms that indicate NE-directed flow. This orientation is incompatible with the prevalent westward current direction; we hypothesize that these features are storm-generated

    Deweyan tools for inquiry and the epistemological context of critical pedagogy

    This article develops the notion of resistance as articulated in the literature of critical pedagogy as being both culturally sponsored and cognitively manifested. To do so, the authors draw upon John Dewey\u27s conception of tools for inquiry. Dewey provides a way to conceptualize student resistance not as a form of willful disputation, but instead as a function of socialization into cultural models of thought that actively truncate inquiry. In other words, resistance can be construed as the cognitive and emotive dimensions of the ongoing failure of institutions to provide ideas that help individuals both recognize social problems and imagine possible solutions. Focusing on Dewey\u27s epistemological framework, specifically tools for inquiry, provides a way to grasp this problem. It also affords some innovative solutions; for instance, it helps conceive of possible links between the regular curriculum and the study of specific social justice issues, a relationship that is often under-examined. The aims of critical pedagogy depend upon students developing dexterity with the conceptual tools they use to make meaning of the evidence they confront; these are background skills that the regular curriculum can be made to serve even outside social justice-focused curricula. Furthermore, the article concludes that because such inquiry involves the exploration and potential revision of students\u27 world-ordering beliefs, developing flexibility in how one thinks may be better achieved within academic subjects and topics that are not so intimately connected to students\u27 current social lives, especially where students may be directly implicated

    Gramicidin Increases Lipid Flip-Flop in Symmetric and Asymmetric Lipid Vesicles

    © 2019 Biophysical Society Unlike most transmembrane proteins, phospholipids can migrate from one leaflet of the membrane to the other. Because this spontaneous lipid translocation (flip-flop) tends to be very slow, cells facilitate the process with enzymes that catalyze the transmembrane movement and thereby regulate the transbilayer lipid distribution. Nonenzymatic membrane-spanning proteins with unrelated primary functions have also been found to accelerate lipid flip-flop in a nonspecific manner and by various hypothesized mechanisms. Using deuterated phospholipids, we examined the acceleration of flip-flop by gramicidin channels, which have well-defined structures and known functions, features that make them ideal candidates for probing the protein-membrane interactions underlying lipid flip-flop. To study compositionally and isotopically asymmetric proteoliposomes containing gramicidin, we expanded a recently developed protocol for the preparation and characterization of lipid-only asymmetric vesicles. Channel incorporation, conformation, and function were examined with small angle x-ray scattering, circular dichroism, and a stopped-flow spectrofluorometric assay, respectively. As a measure of lipid scrambling, we used differential scanning calorimetry to monitor the effect of gramicidin on the melting transition temperatures of the two bilayer leaflets. The two calorimetric peaks of the individual leaflets merged into a single peak over time, suggestive of scrambling, and the effect of the channel on the transbilayer lipid distribution in both symmetric 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine and asymmetric 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine/1,2-dimyristoyl-sn-glycero-3-phosphocholine vesicles was quantified from proton NMR measurements. Our results show that gramicidin increases lipid flip-flop in a complex, concentration-dependent manner. To determine the molecular mechanism of the process, we used molecular dynamics simulations and further computational analysis of the trajectories to estimate the extent of membrane deformation. Together, the experimental and computational approaches were found to constitute an effective means for studying the effects of transmembrane proteins on lipid distribution in both symmetric and asymmetric model membranes
