    Building a Movement: Community Development and Community Resilience in Response to Extreme Events

    There are gaps in understanding what community resilience is and means to those working in communities, academia, practice and policy and how it is currently enacted in Scotland. Greater understanding of existing experiences can be used to encourage development of community resilience.     A paradigm-shift in water treatment: in-reservoir UV-LED-driven TiO2 photocatalysis for the removal of cyanobacteria: a mesocosm study.

    Potentially harmful cyanobacteria challenge potable water treatment globally, with high biomass events, and dissolved toxic and nuisance metabolites. Retrofitting existing water treatment infrastructure is often impractical (if not impossible) and often prohibitively expensive. In a paradigm-shifting move, we propose in-reservoir pre-treatment of cyanobacteria-contaminated raw waters to ease the burden on existing water treatment infrastructure. In an iterative design approach over three years, treatment modules have been designed, refined and optimised, in bench and pilot-scale studies for in-reservoir deployment. TiO2-coated beads made from recycled glass are employed in conjunction with UV-light emitting diodes (LEDs), to create highly reactive hydroxyl radicals that preferably remove cyanobacteria and subsequently released cyanotoxins from raw water. In a mesocosm study using a drinking water reservoir in Brazil, water quality parameters were markedly improved within 72h of deployment and cyanobacterial presence was decreased by over 90% without affecting other phytoplankton communities. The treatment system is virtually plastic-free, low cost, utilises recycled materials and could ultimately be powered by renewable energies, thus providing a true green treatment option. We have conclusively demonstrated that a paradigm-shift towards in-reservoir treatment is not only possible but feasible and can provide a valuable addition to conventional water treatment methods

    The Role of CD 133+ Cells in a Recurrent Embryonal Tumor with Abundant Neuropil and True Rosettes ( ETANTR )

    Embryonal tumor with abundant neuropil and true rosettes ( ETANTR ) is a recently described embryonal neoplasm of the central nervous system, consisting of a well‐circumscribed embryonal tumor of infancy with mixed features of ependymoblastoma (multilayer ependymoblastic rosettes and pseudorosettes) and neuroblastoma (neuroblastic rosettes) in the presence of neuropil‐like islands. We present the case of a young child with a very aggressive tumor that rapidly recurred after gross total resection, chemotherapy and radiation. Prominent vascular sclerosis and circumscribed tumor led to the diagnosis of malignant astroblastoma; however, rapid recurrence and progression of this large tumor after gross total resection prompted review of the original pathology. ETANTR is histologically distinct with focal glial fibrillary acid protein ( GFAP ) and synaptophysin expression in the presence of neuronal and ependymoblastic rosettes with focal neuropil islands. These architectural features, combined with unique chromosome 19q13.42 amplification, confirmed the diagnosis. In this report, we describe tumor stem cell ( TSC ) marker CD 133, CD 15 and nestin alterations in ETANTR before and after chemotherapy. We found that TSC marker CD 133 was richly expressed after chemotherapy in recurrent ETANTR , while CD 15 is depleted compared with that expressed in the original tumor, suggesting that CD 133+ cells likely survived initial treatment, further contributing to formation of the recurrent tumor.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/102077/1/bpa12079.pd

    Case report: Adult-onset limb girdle muscular dystrophy in sibling pair due to novel homozygous LAMA2 missense variant

    Recessive pathogenic variants in the laminin subunit alpha 2 (LAMA2) gene cause a spectrum of disease ranging from severe congenital muscular dystrophy to later-onset limb girdle muscular dystrophy (LGMDR23). The phenotype of LGMDR23 is characterized by slowly progressive proximal limb weakness, contractures, raised creatine kinase, and sometimes distinctive cerebral white matter changes and/or epilepsy. We present two siblings, born to consanguineous parents, who developed adult-onset LGMDR23 associated with typical cerebral white matter changes and who both later developed dementia. The male proband also had epilepsy and upper motor neuron signs when he presented at age 72. Merosin immunohistochemistry and Western blot on muscle biopsies taken from both subjects was normal. Whole exome sequencing revealed a previously unreported homozygous missense variant in LAMA2 [Chr6(GRCh38):g.129297734G>A; NM_000426.3:c.2906G>A; p.(Cys969Tyr)] in the proband. The same homozygous LAMA2 variant was confirmed by Sanger sequencing in the proband's affected sister. These findings expand the genotypic and phenotypic spectrum of LGMDR23

    Cell arrest and cell death in mammalian preimplantation development

    The causes, modes, biological role and prospective significance of cell death in preimplantation development in humans and other mammals are still poorly understood. Early bovine embryos represent a very attractive experimental model for the investigation of this fundamental and important issue. To obtain reference data on the temporal and spatial occurrence of cell death in early bovine embryogenesis, three-dimensionally preserved embryos of different ages and stages of development up to hatched blastocysts were examined in toto by confocal laser scanning microscopy. In parallel, transcript abundance profiles for selected apoptosis-related genes were analyzed by real-time reverse transcriptase-polymerase chain reaction. Our study documents that in vitro as well as in vivo, the first four cleavage cycles are prone to a high failure rate including different types of permanent cell cycle arrest and subsequent non-apoptotic blastomere death. In vitro produced and in vivo derived blastocysts showed a significant incidence of cell death in the inner cell mass (ICM), but only in part with morphological features of apoptosis. Importantly, transcripts for CASP3, CASP9, CASP8 and FAS/FASLG were not detectable or found at very low abundances. In vitro and in vivo, errors and failures of the first and the next three cleavage divisions frequently cause immediate embryo death or lead to aberrant subsequent development, and are the main source of developmental heterogeneity. A substantial occurrence of cell death in the ICM even in fast developing blastocysts strongly suggests a regular developmentally controlled elimination of cells, while the nature and mechanisms of ICM cell death are unclear. Morphological findings as well as transcript levels measured for important apoptosis-related genes are in conflict with the view that classical caspase-mediated apoptosis is the major cause of cell death in early bovine development

    Neuroprotective therapies in the NICU in preterm infants:present and future (Neonatal Neurocritical Care Series)

    The survival of preterm infants has steadily improved thanks to advances in perinatal and neonatal intensive clinical care. The focus is now on finding ways to improve morbidities, especially neurological outcomes. Although antenatal steroids and magnesium for preterm infants have become routine therapies, studies have mainly demonstrated short-term benefits for antenatal steroid therapy but limited evidence for impact on long-term neurodevelopmental outcomes. Further advances in neuroprotective and neurorestorative therapies, improved neuromonitoring modalities to optimize recruitment in trials, and improved biomarkers to assess the response to treatment are essential. Among the most promising agents, multipotential stem cells, immunomodulation, and anti-inflammatory therapies can improve neural outcomes in preclinical studies and are the subject of considerable ongoing research. In the meantime, bundles of care protecting and nurturing the brain in the neonatal intensive care unit and beyond should be widely implemented in an effort to limit injury and promote neuroplasticity. IMPACT: With improved survival of preterm infants due to improved antenatal and neonatal care, our focus must now be to improve long-term neurological and neurodevelopmental outcomes. This review details the multifactorial pathogenesis of preterm brain injury and neuroprotective strategies in use at present, including antenatal care, seizure management and non-pharmacological NICU care. We discuss treatment strategies that are being evaluated as potential interventions to improve the neurodevelopmental outcomes of infants born prematurely.</p

    Evolution of the Stellar Mass Tully-Fisher Relation in Disk Galaxy Merger Simulations

    There is a large observational scatter toward low velocities in the stellar mass Tully-Fisher relation if disturbed and compact objects are included. However, this scatter can be eliminated if one replaces rotation velocity with S0.5\rm S_{\rm 0.5}, a quantity that includes a velocity dispersion term added in quadrature with the rotation velocity. In this work we use a large suite of hydrodynamic N-body galaxy merger simulations to explore a possible mechanism for creating the observed relations. Using mock observations of the simulations, we test for the presence of observational effects and explore the relationship between S0.5\rm S_{\rm 0.5} and intrinsic properties of the galaxies. We find that galaxy mergers can explain the scatter in the TF as well as the tight S0.5\rm S_{\rm 0.5}-stellar mass relation. Furthermore, S0.5\rm S_{\rm 0.5} is correlated with the total central mass of a galaxy, including contributions due to dark matter.Comment: ApJ accepte

    Academic success in schools in the Algarve: when leadership is part of the solution and not of the problem

    This paper presents the results of a case study conducted among school principals in the southern region of Portugal, located in urban, suburban and rural settings. The five cases that comprise this study were subjected to a final rating of “Very Good” in the external evaluation process of schools, whose data were collected through semi-structured interviews and documentary analysis (regulations and educational project, reports of the external evaluation of responsibility of Inspeção Geral de Educação e CiĂȘncia de Portugal). The data collected in this exploratory study were systematized and analyzed according to the following indicators: the students’ academic success, the teachers’ professional development, the organizational development and the identifying features of schools principals. Overall, we cannot affirm the existence of differences according to geographical area, although some particularities which proved to be interesting and indicative of good leadership practices could be highlighted.info:eu-repo/semantics/publishedVersio
