    Review on Tomato (Solanum lycopersicum, L.) Improvement Programmes in Ghana

    Tomato is an important component of every Ghanaian meal, and its cultivation contributes significantly to livelihood improvement. The demand for tomato in Ghana outstrips supply, and therefore local production is augmented by imports from neighbouring countries. Despite the importance of tomato in Ghana, past tomato-breeding programmes have been unsystematic and had not led to the development of new varieties that meet the needs of consumers as well as environmental stresses. This review outlined tomato production trends, constraints and past tomato improvement programmes in Ghana, which mainly focused on germplasm collection, morphological and agronomic characterization, molecular evaluation, diversity study, as well as screening germplasm against biotic and abiotic stresses. The established variability and the outcomes of the evaluations against the various biotic and abiotic stresses have not been utilized in the development of new varieties. This work will serve as a reference for developing future tomato-breeding programmes

    Anti-proliferative effect of Ficus pumila Linn. on human leukemic cell lines

    Background: Cancer is one of the many diseases of global concern due to its high mortality rate with drug resistance becoming a major challenge to chemotherapy and this have propelled many cancer patients to seek alternative and complementary methods of treatment. The objective for this study was, therefore, to determine the antiproliferative activity as well as phytochemical, total phenolic content (TPC), and antioxidant activity of the stem and leaf extracts (FPS and FPL) of Ficus pumila (L.) using standard methods.Methods: The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay was used to evaluate anti-proliferative effect and spectrophotometric-based assays for antioxidant and TPC. Phytochemical constituents were accessed by standard methods.Results: The hydroethanolic extracts of the leaves and stems were rich in tannins, general glycosides, saponins, terpenoids, alkaloids, flavonoids (leaves only), and sterols (stem only). Strong total antioxidant activities were observed with FPL and FPS with EC50 values of 0.07 mg/ml and 0.089 mg/ml, respectively. All the crude extracts showed anti-proliferative effect towards the three human leukemic cell lines used (Jurkat, CEM, and HL-60). However, FPL gave the strongest inhibition concentration at 50% values of 130.97 µg/ml (Jurkat) and 56.31 µg/ml (HL-60).Conclusion: These findings suggest that crude extracts of FPS and FPL have anti-proliferative effect on the leukemia cells. The antioxidant properties of the plant including phenolics may be partly responsible for the anti-proliferative activity. Further studies are required to isolate chemical components of the plant and establish their anti-proliferative activities and mechanism of action

    Bulk genetic characterization of Ghanaian maize landraces using microsatellite markers

    Maize (Zea mays L) was first introduced into Ghana over five centuries ago and remains the most important cereal staple, grown in all agro-ecologies across the country. Yield from farmers’ fields are low, which is attributed in part to farmer’s preferences and/or reliance on local landraces for cultivation. Efforts are underway to improve some of these landraces for improved productivity. Seeds of maize landraces cultivated in all agro-ecologies were col¬lected for genetic characterization using a bulked fingerprinting technique and 20 SSR markers. In all, 20 popula¬tions of 15 plants each from Ghana and 4 control populations from Latin America were characterized. The cluster analysis grouped the 20 landraces into two major groups corresponding to the vegetation/climatic conditions of the north and south of the country. Genotypes from Ashanti, which is centrally located, fell into both major clus¬ters, which suggest its importance in maize seed distribution in Ghana and also the diverse climate/vegetation. A Structure analyses grouped the genotypes into two major clusters similar to the UPGMA cluster, and populations were not fully distinct according to F statistics. The results suggest that breeders should make performance data available to seed dealers for better productivity

    Combatting Cocoa Swollen Shoot Virus Disease: What do we know?

    Cocoa Swollen Shoot Virus Disease (CSSVD) is one of the major factors limiting cocoa (Theobroma cacao L.) productivity in West Africa. The only cure for CSSVD is to cut out visibly infected trees and the official eradication campaign in Ghana has cut out more than 200 million trees since 1946. 80 years of research on preventive control measures have mainly focused on resistance breeding, mild strain cross-protection (inoculation of cocoa seedlings with a mild strain of the virus to protect against the severe strain) and control of mealybug vectors. Meanwhile, diversification measures such as agroforestry (for shading) or barrier (strip) cropping have received less attention. Despite promising results, CSSVD is more prevalent in the field than ever before. The large body of knowledge on preventive control measures for CSSVD is fragmented and many publications are not easily accessible. Furthermore, the literature has never been systematically evaluated and quantitatively assessed. Hence, we consolidated this knowledge with an extensive literature review followed by meta-analysis to identify the pertinent research gaps. Out of 423 publications on CSSVD-related issues, we selected 34 studies, which contained 52 datasets on seven different preventive control measures. Results showed that resistance breeding and mild strain crossprotection may reduce CSSVD infection by 30 percent, while the potential of diversification measures (shading/agroforestry and barrier (strip) cropping) seems to be considerably higher (40 and 85 percent, respectively). However, there is a lack of evidence because of a low number of studies about diversification measures, indicating that our results have to be interpreted with care and calling for more research in this area. Future testing is needed to evaluate the efficacy of barrier (strip) cropping to reduce CSSVD, and address the effect of shade on CSSVD symptom severity. Furthermore, the practical relevance of different preventive control measures for farmers needs to be assessed, and shade should be considered in current breeding programs for CSSVD resistance

    Identification and molecular characterisation of Colletotrichum species from avocado, citrus and pawpaw in Ghana

    Owing to previous identifications based solely on morphological characteristics, the identity of the causal agents of anthracnose disease of pawpaw and avocado in Ghana is in doubt. In addition, the pathogen has not been identified previously on citrus. In this study, isolates of the pathogen were obtained from anthracnose lesions on avocado and pawpaw, and atypical anthracnose lesions on citrus. The isolates were identified using PCR with speciesspecific primers, complemented by phylogenetic analysis of nucleotide sequences of the internal transcribed spacer region and partial glyceraldehyde-3-phosphate dehydrogenase gene. The pathogenicity of the isolated fungi was determined on detached matured fruits. All isolates, including those isolated from citrus, were identified as Colletotrichum gloeosporioides from the expected 480 bp PCR products amplified by PCR. The phylogenetic analysis showed that isolates from avocado and pawpaw were C. siamense, rather than C. gloeosporioides, whereas isolates from citrus were C. gloeosporioides sensu stricto.     Genetic diversity and inter-trait relationship of tropical extra-early maturing quality protein maize inbred lines under low soil nitrogen stress.

    Information on the genetic diversity, population structure, and trait associations of germplasm resources is crucial for predicting hybrid performance. The objective of this study was to dissect the genetic diversity and population structure of extra-early yellow and orange quality protein maize (QPM) inbred lines and identify secondary traits for indirect selection for enhanced grain yield under low-soil nitrogen (LN). One hundred and ten inbred lines were assessed under LN (30 kg ha -1) and assayed for tryptophan content. The lines were genotyped using 2500 single nucleotide polymorphism (SNP) markers. Majority (85.4%) of the inbred lines exhibited wide pairwise genetic distances between 0.4801 and 0.600. Genetic distances were wider between yellow and orange endosperm lines and predicted high heterosis in crosses between parents of different endosperm colors. The unweighted pair group method with arithmetic mean (UPGMA) and the admixture model-based population structure method both grouped the lines into five clusters. The clustering was based on endosperm color, pedigree, and selection history but not on LN tolerance or tryptophan content. Genotype by trait biplot analysis revealed association of grain yield with plant height and ear height. TZEEQI 394 and TZEEIORQ 73A had high expressivity for these traits. Indirect selection for high grain yield among the inbred lines could be achieved using plant and ear heights as selection criteria. The wide genetic variability observed in this study suggested that the inbred lines could be important sources of beneficial alleles for LN breeding programs in SSA

    Crop loss, aetiology, and epidemiology of citrus black spot in Ghana

    Citrus Black Spot (CBS), caused by Guignardia citricarpa, was detected for the first time in Ghana and in West Africa. The disease was first observed in the Eastern Region in 1999 with typical disease symptoms including hard spot, virulent spot and false melanose were observed on several citrus species. A survey revealed that the disease has reached epidemic levels in the citrus-producing areas of the Eastern and Ashanti regions and is spreading rapidly within these areas and to other regions of the country. Currently, CBS is the most important fruit disease of citrus in Ghana, causing about 22% crop loss. Although the disease does not cause postharvest decay and the internal quality of the fruit is not affected, significant amounts of blemished fruit are discarded at the markets. Disease incidence and severity was found to be higher on mature than on young citrus trees. Pycnidia were found on fruit with hard spot symptoms, and pycnidia and pseudothecia typical of Guignardia spp. were found on decomposing leaves. Two species, G. citricarpa and G. mangiferae, were isolated from 15% of the samples collected and identified using the Oatmeal Agar test and by PCR with species-specific DNA primers. Isolates of G. citricarpa produced CBS symptoms after 80 to 233 days on 75% of the artificially inoculated young fruit of Valencia Late sweet orange. The fungus was re-isolated from symptomatic, inoculated fruit completing Koch's postulates. Isolates of the endophyte G. mangiferae did not induce symptoms in the pathogenicity tests. In epidemiological studies, infections were detected from November to February for the minor cropping season and from May to November for the major season. Fruit of Valencia Late sweet orange were susceptible to G. citricarpa infection for up to 7 months after petal fall. Knowledge of the disease cycle in Ghana will improve methods for disease control

    Currently, the only effective treatment for cocoa (Theobroma cacao L.) infected with the cocoa swollen shoot virus disease (CSSVD) is to cut and replant infected trees. Hence, the development of preventive control measures and strategies to mitigate the severity of the disease are of utmost importance. While past research has mainly focused on resistance breeding, mild strain cross protection and vector control, diversification measures such as agroforestry have received relatively less attention, despite their potential to mitigate CSSVD severity. Therefore, we studied the effects of shade on CSSVD symptom severity, capsid damage and cocoa yield along a gradient of increasing shade tree abundance in smallholder cocoa farms in Ghana. Furthermore, we measured photosynthetic active radiation and assessed soil fertility in order to elaborate on potential causal factors for possible shade effects on CSSVD symptom severity. Both CSSVD symptom severity and cocoa yields followed quadratic curves, and were found to be lowest and highest in plots with 54% and 39% shade, respectively. The simulated optimal shade levels for CSSVD symptom severity and cocoa yield overlapped between 45%-53%, indicating that agroforestry systems with around 50% shade cover may be an optimal coping strategy to balance CSSVD symptom severity versus reduced cocoa yield until diseased cocoa is replaced with more resistant varieties. Furthermore, our results suggest that rather than soil fertility, high-light and possibly also soil moisture stress may have been responsible for the shade effects on CSSVD symptom severity

    Social network to inform and prevent the spread of cocoa swollen shoot virus disease in Ghana

    The cocoa swollen shoot virus disease is a major factor limiting cocoa (Theobroma cacao L.) productivity for West African farmers. The only treatment against this disease is to cut infected trees and replant with disease-free planting material. Research has recommended the prevention measures: (i) cordon sanitaire (leaving 10-m-wide cocoa-free zone around cocoa), (ii) barrier cropping, (iii) using partly tolerant hybrids, and (iv) removing specific alternative host tree species. Here, we evaluate the current adoption of these measures and identify their adoption constraints. We conducted a quantitative survey with 396 farmers in the Eastern and Western Regions of Ghana, held six focus group discussions and hosted a multi-stakeholder validation workshop with 31 key actors in the cocoa value chain. Our results indicate that the adoption of prevention measure against the disease remains limited. Farmers with a more extensive social network (number of family members/close friends who already adopted a particular measure), a larger farm size, more secure land tenure rights, and more knowledge about the measures were more likely to adopt them, especially barrier cropping, hybrid seedlings, and removing alternative host trees. Lack of knowledge about the measures was the single biggest barrier for their adoption, with 51% of the participating farmers not even being aware of any prevention measures. Here, we show for the first time that the social network is the main information source for farmers, which agrees with the finding that the flow of information between farmers and other stakeholders is a critical factor affecting knowledge spread and consequently adoption. Our results provide crucial insights for the elaboration of an implementation action plan to boost the dissemination of feasible prevention measures against the cocoa swollen shoot virus disease in Ghana in order to efficiently cover farmers’ needs for information (technical advice) and inputs (access to hybrid seedlings).ISSN:1774-0746ISSN:1773-015
